PU.1 (<i>SPI1</i>) is pivotal in hematopoiesis, yet its role in human endothelial-to-hematopoietic transition (EHT) remains unclear. Comparing human <i>in vivo</i> and <i>in vitro</i> EHT transcriptomes revealed <i>SPI1</i>'s regulatory role.
