Sea surface temperature and current-related parameters affecting local adaptation of scalloped spiny lobster population in Indonesia's archipelagic system.

ABSTRACT: The Scalloped Spiny lobster (Panulirus homarus, Linnaeus 1758) known as one of the commercially harvested Panilurid lobster. This species was distributed widely across continents. Indonesia, as one of the largest archipelagic systems in the world, was also distribution area of the Scalloped Spiny lobster. These facts have led to questions regarding spiny lobster harvest and culture management by considering population differentiation and habitat fragmentation on complex and distinct archipelagic islands. Our investigation was conducted using high-density SNPs datasets from several spiny lobsters harvested from five locations in Indonesia. We found strong differentiation among spiny lobster populations clustered into 3 sub-populations. Environment association analysis and Fst analysis revealed outlier loci significantly associated with Sea Surface Temperature variation and potentially correlated with Sea Current-related parameters. The evidence of a structured population of scalloped spiny lobsters in Indonesia can serve as a consideration in the management of spiny lobsters.

Project description:BackgroundThe scalloped spiny lobster (Panulirus homarus) is a popular seafood commodity worldwide and an important export item from Oman. Annual catches in commercial fisheries are in serious decline, which has resulted in calls for the development of an integrated stock management approach. In Oman, the scalloped spiny lobster is currently treated as a single management unit (MU) or stock and there is an absence of information on the genetic population structure of the species that can inform management decisions, particularly at a fine-scale level. This work is the first to identify genome-wide single nucleotide polymorphisms (SNPs) for P. homarus using Diversity Arrays Technology sequencing (DArT-seq) and to elucidate any stock structure in the species.ResultsAfter stringent filtering, 7988 high utility SNPs were discovered and used to assess the genetic diversity, connectivity and structure of P. homarus populations from Al Ashkharah, Masirah Island, Duqm, Ras Madrakah, Haitam, Ashuwaymiyah, Mirbat and Dhalkut landing sites. Pairwise FST estimates revealed low differentiation among populations (pairwise FST range = - 0.0008 - 0.0021). Analysis of genetic variation using putatively directional FST outliers (504 SNPs) revealed higher and significant pairwise differentiation (p < 0.01) for all locations, with Ashuwaymiyah being the most diverged population (Ashuwaymiyah pairwise FST range = 0.0288-0.0736). Analysis of population structure using Discriminant Analysis of Principal Components (DAPC) revealed a broad admixture among P. homarus, however, Ashuwaymiyah stock appeared to be potentially under local adaptive pressures. Fine scale analysis using Netview R provided further support for the general admixture of P. homarus.ConclusionsFindings here suggested that stocks of P. homarus along the Omani coastline are admixed. Yet, fishery managers need to treat the lobster stock from Ashuwaymiyah with caution as it might be subject to local adaptive pressures. We emphasize further study with larger number of samples to confirm the genetic status of the Ashuwaymiyah stock. The approach utilised in this study has high transferability in conservation and management of other marine stocks with similar biological and ecological attributes.

Project description:The non-feeding postlarva (puerulus) of spiny lobsters actively swims from the open ocean to the coastal habitats where it settles and molts to the first-stage juvenile (JI). Because pueruli use much of their energy reserves swimming and preparing for the post-settlement molt, the survival of JIs presumably depends on resuming feeding as soon as possible. To test this hypothesis, the resistance to starvation of JIs of the Caribbean spiny lobster, Panulirus argus, was evaluated by measuring their point-of-no-return (PNR, minimum time of initial starvation preventing recovery after later feeding) and point-of-reserve-saturation (PRS, minimum time of initial feeding allowing for food-independent development through the rest of the molting cycle) in a warm and a cold season. Each experiment consisted of eight groups: a continuously fed control (FC) group, a continuously starved control (SC) group, and six groups subjected to differential periods of either initial starvation and subsequent feeding (PNR experiments) or initial feeding and subsequent starvation (PSR experiments). No JIs molted under continuous absence of food (SC). In both PNR experiments (temperature in warm season: 29.79 ± 0.07°C, mean ± 95% CI; in cold season: 25.63 ± 0.12°C) mortality increased sharply after 9 d of initial starvation and intermolt periods increased with period of initial starvation, but were longer in the cold season. The PNR50 was longer in the warm season (12.1 ± 1.2 d, mean ± 95% CI) than in the cold season (9.5 ± 2.1 d). In PRS experiments (temperature in warm season: 29.54 ± 0.07 °C; in cold season: 26.20 ± 0.12 °C), JIs that molted did so near the end of the feeding period; all JIs initially fed for up to 6 d succumbed, and no JIs molted after 13 d of starvation despite having fed previously. The PRS50 did not differ between the cold (13.1 ± 0.7 d) and warm seasons (12.1 ± 1.1 d). JIs of P. argus exhibit a remarkable resistance to starvation considering that the previous non-feeding, energy-demanding puerulus phase lasts for ∼3 weeks. However, JIs appear to have a relatively higher degree of dependence on food to complete development to JII during the cold season than during the warm season. Therefore, JIs of P. argus would appear to be more resistant to starvation during the warm season.
