Recovery of Scots Pine Seedlings from Long-Term Zinc Toxicity.

ABSTRACT: We studied the recovery of the growth and physiological parameters of Scots pine seedlings after long-term zinc toxicity. The removal of excess zinc from the nutrient solution resulted in the rapid recovery of primary root growth but did not promote the initiation and growth of lateral roots. The recovery of root growth was accompanied by the rapid uptake of manganese, magnesium, and copper. Despite the maximum rate of manganese uptake by the roots, the manganese content in the needles of the recovering plants did not reach control values during the 28 days of the experiment, unlike magnesium, iron, and copper. In general, the recovery of ion homeostasis eliminated all of the negative effects on the photosynthetic pigment content in the needles. However, these changes, along with recovery of the water content in the needles, were not accompanied by an increase in the weight gain of the recovering seedlings compared with that of the Zn-stressed seedlings. The increased accumulation of phenolic compounds in the needles persisted for a long period after excess zinc was removed from the nutrient solution. The decreased lignin content in the roots and needles is a characteristic feature of Zn-stressed plants. Moreover, the removal of excess zinc from the nutrient solution did not lead to an increase in the lignin content in the organs.

Project description:Tree seedling resistance to and recovery from abiotic stressors such as drought and warming are crucial for forest regeneration and persistence. Selection of more resilient provenances and their use in forest management programs might alleviate pressures of climate change on forest ecosystems. Scots pine forests in particular have suffered frequent drought-induced mortality, suggesting high vulnerability to extreme events. Here, we conducted an experiment using potted Scots pine seedlings from ten provenances of its south-western distribution range to investigate provenance-specific impacts of multiple drought events. Seedlings were grown under ambient and elevated temperatures for 1.5 years and were subjected to consecutive droughts during spring and summer. Growth (height, diameter, and needle) and spring phenology were monitored during the whole study period and complemented by biomass assessments (bud, needle, wood, and needle/wood ratio) as well as measurements of chlorophyll fluorescence and of needle stable carbon isotope ratio. Phenology, growth and biomass parameters as well as carbon isotope ratio and their (direct) responses to reoccurring droughts differed between provenances, indicating genotypic adaptation. Seedling growth was plastic during drought with intra- and inter-annual compensatory growth after drought stress release (carryover effects), however, not fully compensating the initial impact. For (smaller) seedlings from southern/drier origins, sometimes greater drought resistance was observed which diminished under warmer conditions in the greenhouse. Warming increased diameter growth and advanced phenological development, which was (partly) delayed by drought in 2013, but advanced in 2014. Earlier phenology was linked to higher growth in 2013, but interestingly later phenology had positive effects on wood and needle biomass when subjected to drought. Lastly, stable carbon isotope ratios indicated a clear drought response of carbon assimilation. Drought-induced reduction of the photosystem II efficiency was only observed under warmer conditions but showed compensation under ambient temperatures. Besides these direct drought impacts, also interactive effects of previous drought events were shown, either reinforcing or sometimes attenuating the actual impact. Thus, depending on amount and timing of events, Scots pine seedlings, particularly from southern origins, might be well adapted and resilient to drought stress and should be considered when discussing assisted migration under changing climatic conditions.

Project description:Dendroclimatic research offers insight into tree growth-climate response as a solution to the forward problem and provides reconstructions of climatic variables as products of the reverse problem. Methodological developments in dendroclimatology have led to the inclusion of a variety of tree growth parameters in this field. Tree-ring traits developed during short time intervals of a growing season can potentially provide a finer temporal scale of both dendroclimatic applications and offer a better understanding of the mechanisms of tree growth reaction to climatic variations. Furthermore, the transition from classical dendroclimatic studies based on a single integral variable (tree-ring width) to the modern multitude of quantitative variables (e.g., wood anatomical structure) adds a lot of complexity, which mainly arises from intrinsic feedbacks between wood traits and muddles seasonality of registered climatic signal. This study utilized life-long wood anatomical measurements of 150- to 280-year-old trees of Pinus sylvestris L. growing in a moisture-sensitive habitat of the forest-steppe of Southern Siberia (Russia) to investigate and eliminate legacy effect from cell production in tracheid traits. Anatomical parameters were calculated to describe the results of the three main subsequent stages of conifer xylem tracheid development, namely, cell number per radial file in the ring, mean and maximum cell radial diameter, and mean and maximum cell-wall thickness. Although tree-ring width was almost directly proportional to cell number, non-linear relationships with cell number were revealed in tracheid measurements. They exhibited a stronger relationship in the areas of narrow rings and stable anatomical structure in wider rings. The exponential models proposed in this study demonstrated these relationships in numerical terms with morphometric meaning. The ratio of anatomical measurements to their modeled values was used to develop long-term anatomical chronologies, which proved to retain information about climatic fluctuations independent of tree-ring width (cell number), despite decreased common signal.
