Project description:Acetate is a major source of energy and substrate for milk fat synthesis in the dairy cow. We recently reported a linear increase in milk fat yield and greater than a 30% net apparent transfer of acetate to milk fat with ruminal infusion of neutralized acetate. Additionally, ruminal acetate infusion linearly increases plasma β-hydroxybutyrate. The objective of the current study was to investigate the ability of acetate and butyrate fed in a diet to increase milk fat synthesis. Twelve multiparous lactating Holstein cows were randomly assigned to treatments in a 3 × 3 Latin square design with 14-d periods that included a 7-d washout followed by 7 d of treatment. Cows were fed ad libitum a basal diet with a low risk for biohydrogenation-induced milk fat depression, and treatments were mixed into the basal diet. Treatments were 3.2% NaHCO3 (control), 2.9% sodium acetate, and 2.5% calcium butyrate (carbon equivalent to acetate treatment) as a percent of diet dry matter. Feeding sodium acetate increased dry matter intake by 2.7 kg, had no effect on milk yield, and increased milk fat yield by 90 g/d and concentration by 0.2 percentage units, compared with control. Calcium butyrate decreased dry matter intake by 2.6 kg/d, milk yield by 1.65 kg/d, and milk fat yield by 60 g/d, compared with control. Sodium acetate increased concentration and yield of 16 carbon mixed source fatty acids (FA) and myristic acid, while decreasing the concentration of preformed FA, compared with control. Calcium butyrate had no effect on concentration of milk FA by source, but increased concentration of trans-10 C18:1 in milk by 18%, indicating a shift in rumen biohydrogenation pathways. Our data demonstrate that milk fat yield and concentration can be increased by feeding sodium acetate at 2.9% of diet dry matter, but not by feeding calcium butyrate at an equivalent carbon mass.

Project description:Heat stress (HS) damages the global dairy industry by reducing milk yields and quality, harming health, and damaging the reproduction of dairy cows, causing huge economic losses each year. However, an understanding of the physiological mechanism of HS lactating dairy cows remains elusive. Here, a metabolomics study using LC-MS and (1)H NMR spectroscopy was performed to analyze the metabolomic differences in the milk between HS-free and HS dairy cows, and discover diagnostic biomarkers and changes in the metabolic pathway. A total of 53 discriminating metabolites were significantly up- or down-regulated in the HS group compared with the HS-free group (P?<?0.05). These biomarkers were involved in pathways of carbohydrate, amino acid, lipid, and gut microbiome-derived metabolism. Comparing these potential biomarkers with previously identified HS candidate biomarkers in plasma, significant correlations between the levels of lactate, pyruvate, creatine, acetone, ?-hydroxybutyrate, trimethylamine, oleic acid, linoleic acid, lysophosphatidylcholine 16:0, and phosphatidylcholine 42:2 in milk and plasma were found, indicating that the blood-milk barrier became leaky and the levels of these 10 biomarkers in milk can reflect HS-induced metabolomic alterations in blood. These novel findings can support more in-depth research to elucidate the milk-based changes in metabolic pathways in HS lactating dairy cows.

Project description:Growth hormone (GH) has a well established galactopoietic effect on ruminant lactation, however the molecular mechanisms that mediate these effects are not fully understood. This study aimed to define the effects of GH on the production of milk protein by the bovine mammary gland and to establish the potential molecular mechanisms mediating these effects. Elevated levels of GH increased milk yield with corresponding increases in protein, fat and lactose yield, while milk composition remained unaffected by treatment. Increased yield (but not concentration) of all the individual milk proteins was not accompanied by changes in the levels of the milk protein transcripts, with the exception of S2-casein, suggesting post-transcriptional regulation of the milk protein genes is a main factor in the GH-mediated effects on bovine lactation. There were significant transcriptional changes in a wide range of pathways including cell signaling, cell death, cell growth and proliferation, cell cycle regulation and metabolism, consistent with the broad effects of GH. Differential expression of genes involved in the PI3K, ERK/MAPK, insulin receptor, JAK/STAT and IGF-1 signaling pathways were also evident in response to GH treatment supporting potential cross-talk between the GH and insulin signaling pathways in the bovine mammary gland. Transcript levels of the SHC1 gene were elevated in GH-treated cows. SHC1 activates many pathways including those associated with translational regulation and gene transcription and has been implicated in mediating cross-talk between the insulin and GH signaling pathways, providing some insights into the potential pathways mediating the effects of GH on milk protein synthesis in the bovine mammary gland.

Project description:Supplementation with selenium is common for dairy cows, but the importance of selenium source is not clear. This study aimed to compare nano-selenium (Nano-Se) and sodium selenite supplements for dairy cows on lactation performance, milk Se levels and selenoprotein (Sel) gene expression. Twelve multiparous Holstein cows were randomly divided into two groups: a control group fed a basal diet plus 0.30 mg Se/kg of DM as sodium selenite or Nano-Se for 30 days. Dry matter intake, milk yield and composition were not affected by dietary Se source (P?> 0.05); however, the milk total Se levels and milk glutathione peroxidase (GSH-Px) activities were higher with Nano-Se supplementation than sodium selenite (P?<?0.05). At the end of the experiment, Nano-Se supplementation significantly increased plasma Se levels and GSH-Px activity, compared with the sodium selenite supplement. The mRNA expression levels of glutathione peroxidase 1, 2 and 4; thioredoxin reductase 2 and 3; and selenoproteins W, T, K and F were markedly upregulated (P?<?0.05) in the mammary gland of the Nano-Se group. Thus, the source of selenium plays an important role in the antioxidant status and in particular the Sel gene expression in the mammary glands of dairy cows, both being stimulated by nano sources.

Project description:The goals of the current study were to investigate the effects of including chicory (Cichorium intybus L.) into the traditional feeding regime of ryegrass/white clover (Lolium perenne L./Trifolium repens L.), and time of its allocation on milk production, rumen fermentation, and FA composition of milk and rumen digesta of dairy cows. Nine groups of four cows were allocated one of three replicated feeding regimes: (1) ryegrass/white clover only (RGWC), (2) ryegrass/white clover + morning allocation of chicory (CHAM), and (3) ryegrass/white clover + afternoon allocation of chicory (CHPM). One cow per group had a rumen cannulae fitted. Treatment did not affect total grazing time or estimated dry matter intake, but cows ruminated more when fed RGWC than chicory. Allocating chicory in the afternoon elevated milk production compared with RGWC and CHAM. Milk from cows grazing chicory contained greater concentrations of polyunsaturated FA (PUFA) such as C18:3 c9, 12, 15 and C18:2 c9, 12 than those on RGWC. As with milk, rumen digesta concentration of PUFA increased when cows grazed on chicory rather than RGWC, which corresponded with lower concentrations of intermediate vaccenic and biohydrogenation end-product stearic acid for cows grazing on chicory. Mean ruminal pH was lower for cows offered chicory than those on RGWC, reflecting greater rumen concentrations of volatile fatty acids (VFA) for cows fed chicory. Allocating chicory during the afternoon is a useful strategy that can translate to improved milk production. The lower rumen pH, lower concentration of vaccenic and stearic acids, and elevated concentration of PUFA in the rumen of cows fed chicory suggest reduced biohydrogenation and may explain the elevated concentration of PUFA in the milk of cows fed chicory compared with those fed RGWC.

Project description:ObjectiveThe objective of the present study was to elucidate the mechanism underlying liver metabolic perturbations in dairy cows exposed to heat stress (HS).MethodsLiquid chromatography massabl spectrometry was used to analyze metabolic differences in livers of 20 dairy cows, with and without exposure to HS.ResultsThe results revealed 33 potential metabolite candidate biomarkers for the detection of HS in dairy cows. Fifteen of these metabolites (glucose, lactate, pyruvate, acetoacetate, ?-hydroxybutyrate, fumaric acid, citric acid, choline, glycine, proline, isoleucine, leucine, urea, creatinine, and orotic acid) were previously found to be potential biomarkers of HS in plasma or milk, discriminating dairy cows with and without HS.ConclusionAll the potential diagnostic biomarkers were involved in glycolysis, amino acid, ketone, tricarboxylic acid, or nucleotide metabolism, indicating that HS mainly affected energy and nucleotide metabolism in lactating dairy cows.

Project description:Grape pomace (GP) is the main solid by-product of winemaking and represents a rich source of potent bioactive compounds which could display a wide range of beneficial effects in human health for their association with reduced risk of several chronic diseases. Several studies have proposed the use of GP as a macro-ingredient to obtain economically worthwhile animal feedstuffs naturally enriched by polyphenols and dietary fibers. Moreover, the research carried out in this field in the last two decades evidences the ability of GP to induce beneficial effects in cow milk and its derived dairy products. First of all, a general increase in concentration of polyunsaturated fatty acids (PUFA) was observed, and this could be considered the reflection of the high content of these compounds in the by-product. Furthermore, an improvement in the oxidative stability of dairy products was observed, presumably as a direct consequence of the high content of bioactive compounds in GP that are credited with high and well-characterized antioxidant functions. Last but not least, particularly in ripened cheeses, volatile compounds (VOCs) were identified, arising both from lipolytic and proteolytic processes and commonly associated with pleasant aromatic notes. In conclusion, the GP introduction in the diet of lactating cows made it possible to obtain dairy products characterized by improved nutritional properties and high health functionality. Furthermore, the presumable improvement of organoleptic properties seems to be effective in contributing to an increase in the consumer acceptability of the novel products. This review aims to evaluate the effect of the dietary GP supplementation on the quality of milk and dairy products deriving from lactating dairy cows.

Project description:Enteric methane (CH4) is the main source of greenhouse gas emissions from ruminants. The red seaweeds Asparagopsis taxiformis (AT) and Asparagopsis armata contain halogenated compounds, including bromoform (CHBr3), which may strongly decrease enteric CH4 emissions. Bromoform is known to have several toxicological effects in rats and mice and is quickly excreted by the animals. This study investigated the transfer of CHBr3 present in AT to milk, urine, feces, and animal tissue when incorporated in the diet of dairy cows. Twelve lactating Holstein-Friesian dairy cows were randomly assigned to three treatment groups, representing the target dose (low), 2× target dose (medium), and 5× target dose (high). The adaptation period lasted seven days, and subsequently cows were fed AT for 22 days maximally. The transfer of CHBr3 to the urine at days 1 and 10 (10-148 µg/L) was found with all treatments. On day 1, CHBr3 was detected in the milk of most cows in the low and medium treatment groups (9.1 and 11 µg/L, respectively), and detected in the milk of one cow in the high treatment group on day 9 (35 µg/L). Bromoform was not detected in milk and urine at day 17, nor at concentrations above the detection limit in feces and collected animal tissues. Two animals (low) were sacrificed, and their rumen wall showed abnormalities. Upon histological examination, signs of inflammation became visible. Animals regularly refused the feed or distinctively selected against AT. In conclusion, within the confines of the present experiment, CHBr3 does not accumulate in animal tissue, but can be excreted in urine and milk.

Project description:A total of 31 differentially expressed genes in the mammary glands were identified in our previous study using RNA sequencing (RNA-Seq), for lactating cows with extremely high and low milk protein and fat percentages. To determine the regulation of milk composition traits, we herein investigated the expression profiles of microRNA (miRNA) using small RNA sequencing based on the same samples as in the previous RNA-Seq experiment. A total of 497 known miRNAs (miRBase, release 22.1) and 49 novel miRNAs among the reads were identified. Among these miRNAs, 71 were found differentially expressed between the high and low groups (p < 0.05, q < 0.05). Furthermore, 21 of the differentially expressed genes reported in our previous RNA-Seq study were predicted as target genes for some of the 71 miRNAs. Gene ontology and KEGG pathway analyses showed that these targets were enriched for functions such as metabolism of protein and fat, and development of mammary gland, which indicating the critical role of these miRNAs in regulating the formation of milk protein and fat. With dual luciferase report assay, we further validated the regulatory role of 7 differentially expressed miRNAs through interaction with the specific sequences in 3'UTR of the targets. In conclusion, the current study investigated the complexity of the mammary gland transcriptome in dairy cattle using small RNA-seq. Comprehensive analysis of differential miRNAs expression and the data from previous study RNA-seq provided the opportunity to identify the key candidate genes for milk composition traits.

EBseq analysis determined no differentially expressed genes due to postruminal lysine infusion, therefore, read counts were averaged across treatment and block. DAVID functional annotation analysis was utilized to determine specific gene ontologies and KEGG pathways to describe the data by magnitude of reads.
