Project description:Detection of the relevant brain regions for characterizing the distinction between cognitive conditions is one of the most sought after objectives in neuroimaging research. A popular approach for achieving this goal is the multivariate pattern analysis which is currently conducted through a number of approaches such as the popular searchlight procedure. This is due to several advantages such as being automatic and flexible with regards to size of the search region. However, these approaches suffer from a number of limitations which can lead to misidentification of truly informative regions which in turn results in imprecise information maps. These limitations mainly stem from several factors such as the fact that the information value of the search spheres are assigned to the voxel at the center of them (in case of searchlight), the requirement for manual tuning of parameters such as searchlight radius and shape, and high complexity and low interpretability in commonly used machine learning-based approaches. Other drawbacks include overlooking the structure and interactions within the regions, and the disadvantages of using certain regularization techniques in analysis of datasets with characteristics of common functional magnetic resonance imaging data. In this article, we propose a fully data-driven maximum relevance minimum redundancy search algorithm for detecting precise information value of the clusters within brain regions while alleviating the above-mentioned limitations. Moreover, in order to make the proposed method faster, we propose an efficient algorithmic implementation. We evaluate and compare the proposed algorithm with the searchlight procedure as well as least absolute shrinkage and selection operator regularization-based mapping approach using both real and synthetic datasets. The analysis results of the proposed approach demonstrate higher information detection precision and map specificity compared to the benchmark approaches.

Project description:Prevention of age-related cognitive decline is an increasingly important topic. Recently, increased attention is being directed at understanding biological models of successful cognitive aging. Here, we examined resting-state brain regional low-frequency oscillations using functional magnetic resonance imaging in 19 older adults with excellent cognitive abilities (Supernormals), 28 older adults with normative cognition, 57 older adults with amnestic mild cognitive impairment, and 26 with Alzheimer's disease. We identified a "Supernormal map", a set of regions whose oscillations were resistant to the aging-associated neurodegenerative process, including the right fusiform gyrus, right middle frontal gyrus, right anterior cingulate cortex, left middle temporal gyrus, left precentral gyrus, and left orbitofrontal cortex. The map was unique to the Supernormals, differentiated this group from cognitive average-ager comparisons, and predicted a 1-year change in global cognition (indexed by the Montreal Cognitive Assessment scores, adjusted R2 = 0.68). The map was also correlated to Alzheimer's pathophysiological features (beta-amyloid/pTau ratio, adjusted R2 = 0.66) in participants with and without cognitive impairment. These findings in phenotypically successful cognitive agers suggest a divergent pattern of brain regions that may either reflect inherent neural integrity that contributes to Supernormals' cognitive success, or alternatively indicate adaptive reorganization to the demands of aging.

Project description:Over the last decade, there has been growing interest in learning the mapping from structural connectivity (SC) to functional connectivity (FC) of the brain. The spontaneous fluctuations of the brain activity during the resting-state as captured by functional MRI (rsfMRI) contain rich non-stationary dynamics over a relatively fixed structural connectome. Among the modeling approaches, graph diffusion-based methods with single and multiple diffusion kernels approximating static or dynamic functional connectivity have shown promise in predicting the FC given the SC. However, these methods are computationally expensive, not scalable, and fail to capture the complex dynamics underlying the whole process. Recently, deep learning methods such as GraphHeat networks and graph diffusion have been shown to handle complex relational structures while preserving global information. In this paper, we propose a novel attention-based fusion of multiple GraphHeat networks (A-GHN) for mapping SC-FC. A-GHN enables us to model multiple heat kernel diffusion over the brain graph for approximating the complex Reaction Diffusion phenomenon. We argue that the proposed deep learning method overcomes the scalability and computational inefficiency issues but can still learn the SC-FC mapping successfully. Training and testing were done using the rsfMRI data of 1058 participants from the human connectome project (HCP), and the results establish the viability of the proposed model. On HCP data, we achieve a high Pearson correlation of 0.788 (Desikan-Killiany atlas with 87 regions) and 0.773 (AAL atlas with 86 regions). Furthermore, experiments demonstrate that A-GHN outperforms the existing methods in learning the complex nature of the structure-function relation of the human brain.

Project description:(1) Background: Directed functional connectivity (DFC) alterations within brain networks are described using fMRI. EEG has been scarcely used. We aimed to explore changes in DFC in the sensory-motor network (SMN), ventral-attention network (VAN), dorsal-attention network (DAN), and central-executive network (CEN) using an EEG-based mapping between PD patients and healthy controls (HCs). (2) Methods: Four-minutes resting EEG was recorded from 29 PD patients and 28 HCs. Network's hubs were defined using fMRI-based binary masks and their electrical activity was calculated using the LORETA. DFC between each network's hub-pairs was calculated for theta, alpha and beta bands using temporal partial directed coherence (tPDC). (3) Results: tPDCs percent was lower in the CEN and DAN in PD patients compared to HCs, while no differences were observed in the SMN and VAN (group*network: F = 5.943, p < 0.001) in all bands (group*band: F = 0.914, p = 0.401). However, in the VAN, PD patients showed greater tPDCs strength compared to HCs (p < 0.001). (4) Conclusions: Our results demonstrated reduced connectivity in the CEN and DAN, and increased connectivity in the VAN in PD patients. These results indicate a complex pattern of DFC alteration within major brain networks, reflecting the co-occurrence of impairment and compensatory mechanisms processes taking place in PD.

Project description:How is working memory for different visual categories supported in the brain? Do the same principles of cortical specialization that govern the initial processing and encoding of visual stimuli also apply to their short-term maintenance? We investigated these questions with a delayed discrimination paradigm for faces, bodies, flowers, and scenes and applied both univariate and multivariate analyses to functional magnetic resonance imaging (fMRI) data. Activity during encoding followed the well-known specialization in posterior areas. During the delay interval, activity shifted to frontal and parietal regions but was not specialized for category. Conversely, activity in visual areas returned to baseline during that interval but showed some evidence of category specialization on multivariate pattern analysis (MVPA). We conclude that principles of cortical activation differ between encoding and maintenance of visual material. Whereas perceptual processes rely on specialized regions in occipitotemporal cortex, maintenance involves the activation of a frontoparietal network that seems to require little specialization at the category level. We also confirm previous findings that MVPA can extract information from fMRI signals in the absence of suprathreshold activation and that such signals from visual areas can reflect the material stored in memory.

Project description:Typical brain networks consist of many peripheral regions and a few highly central ones, i.e., hubs, playing key functional roles in cerebral inter-regional interactions. Studies have shown that networks, obtained from the analysis of specific frequency components of brain activity, present peculiar architectures with unique profiles of region centrality. However, the identification of hubs in networks built from different frequency bands simultaneously is still a challenging problem, remaining largely unexplored. Here we identify each frequency component with one layer of a multiplex network and face this challenge by exploiting the recent advances in the analysis of multiplex topologies. First, we show that each frequency band carries unique topological information, fundamental to accurately model brain functional networks. We then demonstrate that hubs in the multiplex network, in general different from those ones obtained after discarding or aggregating the measured signals as usual, provide a more accurate map of brain's most important functional regions, allowing to distinguish between healthy and schizophrenic populations better than conventional network approaches.

Project description:The relationship between structural and functional brain networks is still highly debated. Most previous studies have used a single functional imaging modality to analyze this relationship. In this work, we use multimodal data, from functional MRI, magnetoencephalography, and diffusion tensor imaging, and assume that there exists a mapping between the connectivity matrices of the resting-state functional and structural networks. We investigate this mapping employing group averaged as well as individual data. We indeed find a significantly high goodness of fit level for this structure-function mapping. Our analysis suggests that a functional connection is shaped by all walks up to the diameter in the structural network in both modality cases. When analyzing the inverse mapping, from function to structure, longer walks in the functional network also seem to possess minor influence on the structural connection strengths. Even though similar overall properties for the structure-function mapping are found for different functional modalities, our results indicate that the structure-function relationship is modality dependent.

Project description:Theory and modelling suggest that detection of neuronal activity may be feasible using phase sensitive MRI methods. Successful detection of neuronal activity both in vitro and in vivo has been described while others have reported negative results. Magnetic resonance electrical properties tomography may be a route by which signal changes can be identified. Here, we report successful and repeatable detection at 3 Tesla of human brain activation in response to visual and somatosensory stimuli using a functional version of tissue conductivity imaging (funCI). This detects activation in both white and grey matter with apparent tissue conductivity changes of 0.1 S/m (17-20%, depending on the tissue baseline conductivity measure) allowing visualization of complete system circuitry. The degree of activation scales with the degree of the stimulus (duration or contrast). The conductivity response functions show a distinct timecourse from that of traditional fMRI haemodynamic (BOLD or Blood Oxygenation Level Dependent) response functions, peaking within milliseconds of stimulus cessation and returning to baseline within 3-4 s. We demonstrate the utility of the funCI approach by showing robust activation of the lateral somatosensory circuitry on stimulation of an index finger, on stimulation of a big toe or of noxious (heat) stimulation of the face as well as activation of visual circuitry on visual stimulation in up to five different individuals. The sensitivity and repeatability of this approach provides further evidence that magnetic resonance imaging approaches can detect brain activation beyond changes in blood supply.

Project description:Both the putative mirror neuron system (pMNS) and the ventral medial prefrontal cortex (vmPFC) are deemed important for social interaction: the pMNS because it supposedly "resonates" with the actions of others, the vmPFC because it is involved in mentalizing. Strictly speaking, the resonance property of the pMNS has never been investigated. Classical functional MRI experiments have only investigated whether pMNS regions augment their activity when an action is seen or executed. Resonance, however, entails more than only "going on and off together". Activity in the pMNS of an observer should continuously follow the more subtle changes over time in activity of the pMNS of the actor. Here we directly explore whether such resonance indeed occurs during continuous streams of actions. We let participants play the game of charades while we measured brain activity of both gesturer and guesser. We then applied a method to localize directed influences between the brains of the participants: between-brain Granger-causality mapping. Results show that a guesser's brain activity in regions involved in mentalizing and mirroring echoes the temporal structure of a gesturer's brain activity. This provides evidence for resonance theories and indicates a fine-grained temporal interplay between regions involved in motor planning and regions involved in thinking about the mental states of others. Furthermore, this method enables experiments to be more ecologically valid by providing the opportunity to leave social interaction unconstrained. This, in turn, would allow us to tap into the neural substrates of social deficits such as autism spectrum disorder.

Project description:Understanding the relationship between the dynamics of neural processes and the anatomical substrate of the brain is a central question in neuroscience. On the one hand, modern neuroimaging technologies, such as diffusion tensor imaging, can be used to construct structural graphs representing the architecture of white matter streamlines linking cortical and subcortical structures. On the other hand, temporal patterns of neural activity can be used to construct functional graphs representing temporal correlations between brain regions. Although some studies provide evidence that whole-brain functional connectivity is shaped by the underlying anatomy, the observed relationship between function and structure is weak, and the rules by which anatomy constrains brain dynamics remain elusive. In this article, we introduce a methodology to map the functional connectivity of a subject at rest from his or her structural graph. Using our methodology, we are able to systematically account for the role of structural walks in the formation of functional correlations. Furthermore, in our empirical evaluations, we observe that the eigenmodes of the mapped functional connectivity are associated with activity patterns associated with different cognitive systems.
