Project description:Multicomponent hydrogels offer a tremendous opportunity for preparing useful and exciting materials that cannot be accessed using a single component. Here, we describe an unusual multi-component low-molecular weight gelling system that exhibits pH-responsive behavior involving cooperative hydrogen bonding between the components, allowing it to maintain a gel phase across a wide pH range. Unlike traditional acid-triggered gels, our system undergoes a change in the underlying molecular packing and maintains the β-sheet structure both at acidic and basic pH. We further establish that autonomous programming between these two gel states is possible by an enzymatic reaction which allows us to prepare gels with improved mechanical properties.

Project description:Many efforts have been made to develop bifunctional electrocatalysts to facilitate overall water splitting. Here, a fibrous bifunctional 3D electrocatalyst is reported for both the hydrogen evolution reaction (HER) and the oxygen evolution reaction (OER) with high performance. The remarkable electrochemical performance is attributed of the catalysts to a number of factors: the metallic character of the three components (i.e., Ni3N, CoN, and NiCo2O4); the electronic structure, nanoflake-nanosphere network with abundant electroactive sites, and the electric field effect at the interfaces between different components. The oxide-nitride/graphite fibers have the lowest overpotential requirements of 71 and 183 mV at 10 mA cm-2 for HER and OER in alkaline medium, respectively. These values are comparable to those of commercial Pt/C (20 wt%) and RuO2. The electrodes also show a response to HER and OER in both neutral and acid media. Furthermore, the 3D structure can be highlighted by all-round electrodes for overall water splitting. The calculations on the changes in electrons transfer and the Femi level from oxides to oxides/nitrides reveal that the observed superb electrocatalytic performance can be attributed to the presence of Ni3N and CoN derived from the in situ nitridation of NiCo2O4.

Project description:BACKGROUND:Glycerol is currently an over-produced chemical that can be used as substrate for the production of high value products such as 1,3-propanediol (1,3-PDO) in fermentation processes. The aim of this study was to investigate the effect of initial pH on a batch mixed culture fermentation of glycerol, considering both the bacterial community composition and the fermentation patterns. RESULTS:For pH values between 5 and 9, 1,3-PDO production yields ranged from 0.52 ± 0.01 to 0.64 ± 0.00 [Formula: see text], with the highest values obtained at pH 7 and 8. An Enterobacteriaceae member closely related to Citrobacter freundii was strongly enriched at all pH values. Within the less dominant bacterial species, two different microbial community structures were found, one at acid pH values and another at neutral to basic pH values. CONCLUSIONS:1,3-PDO production was improved at pH values over 7. It was anti-correlated with lactate and ethanol production but positively correlated with acetate production. No direct correlation between 1,3-PDO production and a specific family of bacteria was found, suggesting functional redundancies in the microbial community. However, 1,3-PDO production yield remained high over the range of pH studied and was comparable to the best obtained in the same conditions in the literature.

Project description:The water-soluble quencher hydrodabcyl can be activated as an N-succinimidyl ester that is readily accessible from crude hydrodabcyl and storable for a long time. With primary and secondary amines, it reacts swiftly and chemoselectively, even in the presence of other competing nucleophiles such as those typically present in natural peptides. One of the three phenolic OH groups of hydrodabcyl is amenable to selective mono-Boc protection resulting in reduced polarity, advantageous to its further use in organic synthesis. The advantages of hydrodabcyl over dabcyl in spectrometric applications are exemplified by the pH dependence of its absorbance spectra.

Project description:We have developed a convenient and selective method for the detection of Gram-positive bacteria using a ditopic poly(amidoamine) (PAMAM) dendrimer probe. The dendrimer that was modified with dipicolylamine (dpa) and phenylboronic acid groups showed selectivity toward Staphylococcus aureus. The ditopic dendrimer system had higher sensitivity and better pH tolerance than the monotopic PAMAM dendrimer probe. We also investigated the mechanisms of various ditopic PAMAM dendrimer probes and found that the selectivity toward Gram-positive bacteria was dependent on a variety of interactions. Supramolecular interactions, such as electrostatic interaction and hydrophobic interaction, per se, did not contribute to the bacterial recognition ability, nor did they improve the selectivity of the ditopic dendrimer system. In contrast, the ditopic PAMAM dendrimer probe that had a phosphate-sensing dpa group and formed a chelate with metal ions showed improved selectivity toward S. aureus. The results suggested that the targeted ditopic PAMAM dendrimer probe showed selectivity toward Gram-positive bacteria. This study is expected to contribute to the elucidation of the interaction between synthetic molecules and bacterial surface. Moreover, our novel method showed potential for the rapid and species-specific recognition of various bacteria.

Project description:Nucleation and growth of aerosol particles from atmospheric vapors constitutes a major source of global cloud condensation nuclei (CCN). The fraction of newly formed particles that reaches CCN sizes is highly sensitive to particle growth rates, especially for particle sizes <10 nm, where coagulation losses to larger aerosol particles are greatest. Recent results show that some oxidation products from biogenic volatile organic compounds are major contributors to particle formation and initial growth. However, whether oxidized organics contribute to particle growth over the broad span of tropospheric temperatures remains an open question, and quantitative mass balance for organic growth has yet to be demonstrated at any temperature. Here, in experiments performed under atmospheric conditions in the Cosmics Leaving Outdoor Droplets (CLOUD) chamber at the European Organization for Nuclear Research (CERN), we show that rapid growth of organic particles occurs over the range from [Formula: see text]C to [Formula: see text]C. The lower extent of autoxidation at reduced temperatures is compensated by the decreased volatility of all oxidized molecules. This is confirmed by particle-phase composition measurements, showing enhanced uptake of relatively less oxygenated products at cold temperatures. We can reproduce the measured growth rates using an aerosol growth model based entirely on the experimentally measured gas-phase spectra of oxidized organic molecules obtained from two complementary mass spectrometers. We show that the growth rates are sensitive to particle curvature, explaining widespread atmospheric observations that particle growth rates increase in the single-digit-nanometer size range. Our results demonstrate that organic vapors can contribute to particle growth over a wide range of tropospheric temperatures from molecular cluster sizes onward.

Project description:Bio-mediated ground improvement is an attractive alternative to traditional admixtures for strength improvement of shallow surfaces because it is environmentally friendly. Since biofilms contain extracellular polymeric substances (EPS), they can be considered as an alternative to current technologies to improve soil strength. EPS containing biofilms are porous materials with charged surfaces, and therefore adsorption and capillary condensation can result in water retention. Currently, most of the literature work only tested soil improvement under the dry condition. Therefore, the influence of water retention by biofilms on the strength of improved soil remains unclear. Our goal is to evaluate the strength of a biofilm-enhanced sand over a wide range of saturations and explain the trends through the suction stress characteristic curve (SSCC), which quantifies interparticle adsorptive, capillary, and cementation forces as a function of saturation. We used homogenized anaerobic granule biofilms from an existing upflow anaerobic sludge blanket reactor and mixed it with sand to test the strength of a poorly-graded sand in a wide range of saturations, S (between 0.02 S and 0.74 S). We found that biofilm treatment of sand increases soil strength through cementation over a wide range of saturations and through adsorptive forces among sand, biofilm surfaces, and water molecules at low saturations (S < ~0.3). Our results suggested that homogenized biofilms mixed with sand can be used to improve the strength of sand over a wide range of saturations.

Project description:The biochemical activation of amino acids by adenosine triphosphate (ATP) drives the synthesis of proteins that are essential for all life. On the early Earth, before the emergence of cellular life, the chemical condensation of amino acids to form prebiotic peptides or proteins may have been activated by inorganic polyphosphates, such as tri metaphosphate (TP). Plausible volcanic and other potential sources of TP are known, and TP readily activates amino acids for peptide synthesis. But de novo peptide synthesis also depends on pH, temperature, and processes of solvent drying, which together define a varied range of potential activating conditions. Although we cannot replay the tape of life on Earth, we can examine how activator, temperature, acidity and other conditions may have collectively shaped its prebiotic evolution. Here, reactions of two simple amino acids, glycine and alanine, were tested, with or without TP, over a wide range of temperature (0-100 °C) and acidity (pH 1-12), while open to the atmosphere. After 24 h, products were analyzed by HPLC and mass spectrometry. In the absence of TP, glycine and alanine readily formed peptides under harsh near-boiling temperatures, extremes of pH, and within dry solid residues. In the presence of TP, however, peptides arose over a much wider range of conditions, including ambient temperature, neutral pH, and in water. These results show how polyphosphates such as TP may have enabled the transition of peptide synthesis from harsh to mild early Earth environments, setting the stage for the emergence of more complex prebiotic chemistries.

Project description:Phosphorylation is one of the main regulators of cellular signaling typically occurring in flexible parts of folded proteins and in intrinsically disordered regions. It can have distinct effects on the chemical environment as well as on the structural properties near the modification site. Secondary chemical shift analysis is the main NMR method for detection of transiently formed secondary structure in intrinsically disordered proteins (IDPs) and the reliability of the analysis depends on an appropriate choice of random coil model. Random coil chemical shifts and sequence correction factors were previously determined for an Ac-QQXQQ-NH2-peptide series with X being any of the 20 common amino acids. However, a matching dataset on the phosphorylated states has so far only been incompletely determined or determined only at a single pH value. Here we extend the database by the addition of the random coil chemical shifts of the phosphorylated states of serine, threonine and tyrosine measured over a range of pH values covering the pKas of the phosphates and at several temperatures (www.bio.ku.dk/sbinlab/randomcoil). The combined results allow for accurate random coil chemical shift determination of phosphorylated regions at any pH and temperature, minimizing systematic biases of the secondary chemical shifts. Comparison of chemical shifts using random coil sets with and without inclusion of the phosphoryl group, revealed under/over estimations of helicity of up to 33%. The expanded set of random coil values will improve the reliability in detection and quantification of transient secondary structure in phosphorylation-modified IDPs.
