Pupillometric and behavioral markers of a developmental shift in the temporal dynamics of cognitive control.

ABSTRACT: The capacity to anticipate and prepare for future events is thought to be critical for cognitive control. Dominant accounts of cognitive control treat the developing system as merely a weaker version of the adult system, progressively strengthening over time. Using the AX Continuous Performance Task (AX-CPT) in combination with high-resolution pupillometry, we find that whereas 8-year-old children resemble adults in their proactive use of cognitive control, 3.5-year-old children exhibit a qualitatively different, reactive form of cognitive control, responding to events only as they unfold and retrieving information from memory as needed in the moment. These results demonstrate the need to reconsider the origins of cognitive control and the basis for children's behaviors across domains.

A Pupillometric Examination of Cognitive Control in Taxonomic and Thematic Semantic Memory.

Project description:Semantic cognition includes taxonomic and thematic relationships, as well as control systems to retrieve and manipulate semantic knowledge to suit specific tasks or contexts. A recent report (Thompson et al., 2017) suggested that retrieving thematic relationships (i.e., relations based on participation in the same event or scenarios) requires more effort or cognitive control, especially when the relevant relations are weak, than retrieving identity relations that are based on sensory-motor features. It is not clear whether the same contrast applies to the broader set of taxonomic relations, which are also based on shared sensory-motor features. In this study we tested cognitive control requirements of retrieving taxonomic and thematic knowledge using a physiological measure of cognitive effort: pupil dilation. Participants completed a semantic relatedness judgement task that manipulated semantic type (thematic vs. taxonomic) and relatedness strength (high vs. low) of word pairs. Cognitive control in the similarity task was examined using task-evoked pupillary responses (TEPRs), as well as standard behavioral measures (reaction times and accuracy). Compared with high-strength relations, low-strength semantic relations elicited larger TERPs, slower reaction times, and lower accuracy, consistent with higher control demands. Compared to thematic relations, taxonomic relations also elicited larger TERPs and slower reaction times, suggesting that retrieving taxonomic relations required more cognitive effort. Critically, our pupillometric data indicated that controlled processing was particularly important for low-strength taxonomic pairs rather than low-strength thematic pairs. These findings indicate that semantic control demands are primarily determined by relatedness strength, not whether the relationship is taxonomic or thematic.

A pupillometric study of developmental and individual differences in cognitive effort in visual word recognition.

Project description:Throughout the history of modern psychology, the neural basis of cognitive performance, and particularly its efficiency, has been assumed to be an essential determinant of developmental and individual differences in a wide range of human behaviors. Here, we examine one aspect of cognitive efficiency-cognitive effort, using pupillometry to examine differences in word reading among adults (N = 34) and children (N = 34). The developmental analyses confirmed that children invested more effort in reading than adults, as indicated by larger and sustained pupillary responses. The within-age (individual difference) analyses comparing faster (N = 10) and slower (N = 10) performers revealed that in both age groups, the faster readers demonstrated accelerated pupillary responses compared to slower readers, although both groups invested a similar overall degree of cognitive effort. These findings have the potential to open up new avenues of research in the study of skill growth in word recognition and many other domains of skill learning.

Neural and behavioral markers of inhibitory control predict symptom improvement during internet-delivered cognitive behavioral therapy for depression.

Project description:Poor inhibitory control contributes to deficits in emotion regulation, which are often targeted by treatments for major depressive disorder (MDD), including cognitive behavioral therapy (CBT). Brain regions that contribute to inhibitory control and emotion regulation overlap; thus, inhibitory control might relate to response to CBT. In this study, we examined whether baseline inhibitory control and resting state functional connectivity (rsFC) within overlapping emotion regulation-inhibitory control regions predicted treatment response to internet-based CBT (iCBT). Participants with MDD were randomly assigned to iCBT (N = 30) or a monitored attention control (MAC) condition (N = 30). Elastic net regression was used to predict post-treatment Patient Health Questionnaire-9 (PHQ-9) scores from baseline variables, including demographic variables, PHQ-9 scores, Flanker effects (interference, sequential dependency, post-error slowing), and rsFC between the dorsal anterior cingulate cortex, bilateral anterior insula (AI), and right temporoparietal junction (TPJ). Essential prognostic predictor variables retained in the elastic net regression included treatment group, gender, Flanker interference response time (RT), right AI-TPJ rsFC, and left AI-right AI rsFC. Prescriptive predictor variables retained included interactions between treatment group and baseline PHQ-9 scores, age, gender, Flanker RT, sequential dependency effects on accuracy, post-error accuracy, right AI-TPJ rsFC, and left AI-right AI rsFC. Inhibitory control and rsFC within inhibitory control-emotion regulation regions predicted reduced symptom severity following iCBT, and these effects were stronger in the iCBT group than in the MAC group. These findings contribute to a growing literature indicating that stronger inhibitory control at baseline predicts better outcomes to psychotherapy, including iCBT.

Temporal dynamics of reactive cognitive control as revealed by event-related brain potentials.

Project description:Reactive cognitive control refers to a complementary set of cognitive operations by which individuals monitor for and detect the presence of goal-interfering conflict (i.e., conflict monitoring/evaluation) and, subsequently, initiate attention-focusing and response selection processes to bolster goal-directed action in the face of such conflict (regulative control). The purpose of the current study was to characterize the nature of conflict adaptation in both components of this dynamic process across sequences of trials and, more broadly, across time as participants complete a cognitive control task. Fifty-two young adults completed a standard arrow flanker task while behavioral and ERP data were recorded. Multilevel modeling of sequences of compatible and incompatible trials over time showed that, whereas response time data demonstrated a typical conflict adaptation effect throughout the task, N2 and frontal slow wave (FSW) indices of conflict monitoring and regulative control, respectively, demonstrated significant conflict adaptation only during the early part of the task. Moreover, although differential change in N2 and FSW over time suggested that conflict monitoring and regulative control were dissociable, a reciprocal relation between them was maintained throughout the task and was not present in a component theoretically unrelated to conflict adaptation (visual attention-related N1). Findings are discussed in terms of compensatory processes that help to maintain goal-directed performance even as control-related neural responses become fatigued.

Temporal Dynamics of Human Frontal and Cingulate Neural Activity During Conflict and Cognitive Control.

Project description:Cognitive control refers to the ability to produce flexible, goal-oriented behavior in the face of changing task demands and conflicting response tendencies. A classic cognitive control experiment is the Stroop-color naming task, which requires participants to name the color in which a word is written while inhibiting the tendency to read the word. By comparing stimuli with conflicting word-color associations to congruent ones, control processes over response tendencies can be isolated. We assessed the spatial specificity and temporal dynamics in the theta and gamma bands for regions engaged in detecting and resolving conflict in a cohort of 13 patients using a combination of high-resolution surface and depth recordings. We show that cognitive control manifests as a sustained increase in gamma band power, which correlates with response time. Conflict elicits a sustained gamma power increase but a transient theta power increase, specifically localized to the left cingulate sulcus and bilateral dorsolateral prefrontal cortex (DLPFC). Additionally, activity in DLPFC is affected by trial-by-trial modulation of cognitive control (the Gratton effect). Altogether, the sustained local neural activity in dorsolateral and medial regions is what determines the timing of the correct response.

Cognitive behavioral markers of neurodevelopmental trajectories in rodents.

Project description:Between adolescence and adulthood, the brain critically undergoes maturation and refinement of synaptic and neural circuits that shape cognitive processing. Adolescence also represents a vulnerable period for the onset of symptoms in neurodevelopmental psychiatric disorders. Despite the wide use of rodent models to unravel neurobiological mechanisms underlying neurodevelopmental disorders, there is a surprising paucity of rigorous studies focusing on normal cognitive-developmental trajectories in such models. Here, we sought to behaviorally capture maturational changes in cognitive trajectories during adolescence and into adulthood in male and female mice using distinct behavioral paradigms. C57 BL/6J mice (4.5, 6, and 12 weeks of age) were assessed on three behavioral paradigms: drug-induced locomotor hyperactivity, prepulse inhibition, and a novel validated version of a visuospatial paired-associate learning touchscreen task. We show that the normal maturational trajectories of behavioral performance on these paradigms are dissociable. Responses in drug-induced locomotor hyperactivity and prepulse inhibition both displayed a 'U-shaped' developmental trajectory; lower during mid-adolescence relative to early adolescence and adulthood. In contrast, visuospatial learning and memory, memory retention, and response times indicative of motivational processing progressively improved with age. Our study offers a framework to investigate how insults at different developmental stages might perturb normal trajectories in cognitive development. We provide a brain maturational approach to understand resilience factors of brain plasticity in the face of adversity and to examine pharmacological and non-pharmacological interventions directed at ameliorating or rescuing perturbed trajectories in neurodevelopmental and neuropsychiatric disorders.

Comprehensive Temporal Protein Dynamics during the Diauxic Shift in Saccharomyces cerevisiae.

Project description:Yeast (Saccharomyces cerevisiae) has served as a key model system in biology and as a benchmark for "omics" technology. Although near-complete proteomes of log phase yeast have been measured, protein abundance in yeast is dynamic, particularly during the transition from log to stationary phase. Defining the dynamics of proteomic changes during this transition, termed the diauxic shift, is important to understand the basic biology of proliferative versus quiescent cells. Here, we perform temporal quantitative proteomics to fully capture protein induction and repression during the diauxic shift. Accurate and sensitive quantitation at a high temporal resolution and depth of proteome coverage was achieved using TMT10 reagents and LC-MS3 analysis on an Orbitrap Fusion tribrid mass spectrometer deploying synchronous precursor selection. Triplicate experiments were analyzed using the time-course R package and a simple template matching strategy was used to reveal groups of proteins with similar temporal patterns of protein induction and repression. Within these groups are functionally distinct types of proteins such as those of glyoxylate metabolism and many proteins of unknown function not previously associated with the diauxic shift (e.g. YNR034W-A and FMP16). We also perform a dual time-course experiment to determine Hap2-dependent proteins during the diauxic shift. These data serve as an important basic model for fermentative versus respiratory growth of yeast and other eukaryotes and are a benchmark for temporal quantitative proteomics.

Cognitive-behavioral correlates of pupil control ideology.

Project description:Teacher's pupil control ideology is a central feature for the quality of the teacher-student relationship, which, in turn, impacts the teacher's level of well-being. The pupil control ideology refers to a teacher's belief system along a continuum from humanistic to custodial views. Teachers with humanistic orientation view students as responsible and, therefore, they exert a lower degree of control to manage students' classroom behaviors. Teachers with a custodial orientation view students as untrustworthy and, therefore, they exert a higher degree of control to manage students' classroom behaviors. The relationship between pupil control ideology and dysfunctional beliefs originated from the cognitive-behavioral therapy framework has not been investigated, despite existing evidence suggesting that the pupil control ideology is linked to stress and burnout. One hundred fifty-five teachers completed a set of self-report questionnaires measuring: (i) teacher's pupil-control ideology; (ii) perfectionistic and hostile automatic thoughts; (iii) irrational beliefs; (iv) unconditional self-acceptance; (v) early maladaptive schemas; and (vi) dimensions of perfectionism. The result suggests that teachers who adopt a custodial view on pupil control ideology endorse more dysfunctional beliefs than teachers who adopt a humanistic view. They tend to present a higher level of perfectionism, unrelenting standards, and problematic relational beliefs, including schemas of mistrust and entitlement. They also present more often other-directed demands and derogation of other thoughts. Such results picture a dysfunctional view on pupils who misbehave, as adversaries who threaten their rigid and/or perfectionistic expectations.

Temporal Dynamics of Motor Functioning and Cognitive Aging.

Project description:BackgroundBecause of the possible implications for intervention and thus successful aging, researchers have striven to determine whether the age changes in physical and cognitive functioning are coincident or does functioning in one domain change before, and possibly contribute to, functioning in the other.MethodsBivariate dual change score models were applied to four cognitive factors and three motor functioning factors available from 813 adults who participated in the Swedish Adoption/Twin Study of Aging. Participants were aged 50-88 at the first of six waves of testing covering a 19-year follow-up period; 68% participated in at least three waves.ResultsModel comparisons indicated dynamic coupling relationships between Balance and Fine Motor factors and the Speed cognitive factor. Decline in motor function precedes decline in performance on processing speed tasks, even though the motor function tasks were not timed. Results indicated possible bidirectional coupling between Fine Motor and Speed.ConclusionsCombined with other dual change score model analyses of cognition and physical function, a picture is beginning to emerge of the cascade of events that may lead to cognitive aging.
