Fabrication of silica-coated gold nanorods functionalized with DNA for enhanced surface plasmon resonance imaging biosensing applications.

ABSTRACT: A novel method for preparing gold nanorods that are first coated with a thin silica film and then functionalized with single-stranded DNA (ssDNA) is presented. Coating the nanorods with 3-5 nm of silica improves their solubility and stability. Amine-modified ssDNA is attached to the silica-coated gold nanorods via a reductive amination reaction with an aldehyde trimethoxysilane monolayer. The nanorods exhibit an intense absorption band at 780 nm, and are used to enhance the sensitivity of surface plasmon resonance imaging (SPRI) measurements on DNA microarrays.

Induced pH-dependent shift by local surface plasmon resonance in functionalized gold nanorods.

Project description:Localized surface plasmon resonance (LSPR) spectroscopy of metallic nanoparticles is a powerful tool for chemical and biological sensing experiments. In this study, we observed LSPR shifts of 11-mercaptoundecanoic acid modified gold nanorods (GNR-MUA) for the pH range of 6.41 to 8.88. We proposed a mechanism involving changes of the dipole moment after protonation/deprotonation carboxylic groups of 11-mercaptoundecanoic acid (MUA) which plays an important role by modulating LSPR around the functionalized GNR. Such a stable and easily prepared GNR-MUA has potential to become one of the most efficient and promising pH nanosensors to study intra- or extra-cellular pH in a wide range of chemical or biological systems.

Silica-coated gold nanorods as photoacoustic signal nanoamplifiers.

Project description:Photoacoustic signal generation by metal nanoparticles relies on the efficient conversion of light to heat, its transfer to the environment, and the production of pressure transients. In this study we demonstrate that a dielectric shell has a strong influence on the amplitude of the generated photoacoustic signal and that silica-coated gold nanorods of the same optical density are capable of producing about 3-fold higher photoacoustic signals than nanorods without silica coating. Spectrophotometry measurements and finite difference time domain (FDTD) analysis of gold nanorods before and after silica coating showed only an insignificant change of the extinction and absorption cross sections, hence indicating that the enhancement is not attributable to changes in absorption cross section resulting from the silica coating. Several factors including the silica thickness, the gold/silica interface, and the surrounding solvent were varied to investigate their effect on the photoacoustic signal produced from silica-coated gold nanorods. The results suggest that the enhancement is caused by the reduction of the gold interfacial thermal resistance with the solvent due to the silica coating. The strong contrast enhancement in photoacoustic imaging, demonstrated using phantoms with silica-coated nanorods, shows that these hybrid particles acting as "photoacoustic nanoamplifiers" are high efficiency contrast agents for photoacoustic imaging or photoacoustic image-guided therapy.

Template-stripped smooth Ag nanohole arrays with silica shells for surface plasmon resonance biosensing.

Project description:Inexpensive, reproducible, and high-throughput fabrication of nanometric apertures in metallic films can benefit many applications in plasmonics, sensing, spectroscopy, lithography, and imaging. Here we use template-stripping to pattern periodic nanohole arrays in optically thick, smooth Ag films with a silicon template made via nanoimprint lithography. Ag is a low-cost material with good optical properties, but it suffers from poor chemical stability and biocompatibility. However, a thin silica shell encapsulating our template-stripped Ag nanoholes facilitates biosensing applications by protecting the Ag from oxidation as well as providing a robust surface that can be readily modified with a variety of biomolecules using well-established silane chemistry. The thickness of the conformal silica shell can be precisely tuned by atomic layer deposition, and a 15 nm thick silica shell can effectively prevent fluorophore quenching. The Ag nanohole arrays with silica shells can also be bonded to polydimethylsiloxane (PDMS) microfluidic channels for fluorescence imaging, formation of supported lipid bilayers, and real-time, label-free SPR sensing. Additionally, the smooth surfaces of the template-stripped Ag films enhance refractive index sensitivity compared with as-deposited, rough Ag films. Because nearly centimeter-sized nanohole arrays can be produced inexpensively without using any additional lithography, etching, or lift-off, this method can facilitate widespread applications of metallic nanohole arrays for plasmonics and biosensing.

Multipole plasmon resonances in gold nanorods.

Project description:The optical properties of gold rods electrochemically deposited in anodic aluminum oxide templates have been investigated. Homogeneous suspensions of rods with an average diameter of 85 nm and varying lengths of 96, 186, 321, 465, 495, 578, 641, 735, and 1175 nm were fabricated. The purity and dimensions of these rod nanostructures allowed us to observe higher-order multipole resonances for the first time in a colloidal suspension. The experimental optical spectra agree with discrete dipole approximation calculations that have been modeled from the dimensions of the gold nanorods.

Investigation of Performance Parameters of Spherical Gold Nanoparticles in Localized Surface Plasmon Resonance Biosensing.

Project description:In this paper, we present numerical and experimental results on Localized Surface Plasmon Resonance (LSPR) refractive index (RI) sensitivity, Figure of Merit (FoM), and penetration depth (dp) dependence on spherical gold nanoparticles (AuNPs) size, and the effects of AuNP dimer interparticle distance (ds) studied numerically. These parameters were calculated and observed for d = 20, 40, 60, 80, and 100 nm diameter spherical AuNPs. Our investigation shows d = 60 nm AuNPs give the best FoM. The AuNP dimer interparticle distance can significantly influence the RI sensitivity. Therefore, the effect of distances between pairs of d = 20 nm and 60 nm AuNPs is shown. We discuss the importance of penetration depth information for AuNPs functionalized with aptamers for biosensing in the context of aptamer size.

Photoacoustic imaging of mesenchymal stem cells in living mice via silica-coated gold nanorods.

Project description:Improved imaging modalities are critically needed for optimizing stem cell therapy. Techniques with real-time content to guide and quantitate cell implantation are especially important in applications such as musculoskeletal regenerative medicine. Here, we report the use of silica-coated gold nanorods as a contrast agent for photoacoustic imaging and quantitation of mesenchymal stem cells in rodent muscle tissue. The silica coating increased the uptake of gold into the cell more than 5-fold, yet no toxicity or proliferation changes were observed in cells loaded with this contrast agent. Pluripotency of the cells was retained, and secretome analysis indicated that only IL-6 was disregulated more than 2-fold from a pool of 26 cytokines. The low background of the technique allowed imaging of down to 100,000 cells in vivo. The spatial resolution is 340 ?m, and the temporal resolution is 0.2 s, which is at least an order of magnitude below existing cell imaging approaches. This approach has significant advantages over traditional cell imaging techniques like positron emission tomography and magnetic resonance imaging including real time monitoring of stem cell therapy.

Applications of Mesoporous Silica-Encapsulated Gold Nanorods Loaded Doxorubicin in Chemo-photothermal Therapy.

Project description:We investigate the chemo-photothermal effects of gold nanorods (GNRs) coated using mesoporous silica (mSiO2) loading doxorubicin (DOX). When the mesoporous silica layer is embedded by doxorubicin drugs, a significant change in absorption spectra enables to quantify the drug loading. We carried out photothermal experiments on saline and livers of mice having GNRs@mSiO2 and GNRs@mSiO2-DOX. We also injected the gold nanostructures into many tumor-implanted mice and used laser illumination on some of them. By measuring the weight and size of tumors, the distinct efficiency of photothermal therapy and chemotherapy on treatment is determined. We experimentally confirm the accumulation of gold nanostructures in the liver.

Nanocomposites Based on Spin-Crossover Nanoparticles and Silica-Coated Gold Nanorods: A Nonlinear Optical Study.

Project description:A nanocomposite based on silica-coated AuNRs with the aminated silica-covered spin-crossover nanoparticles (SCO NPs) of the 1D iron(II) coordination polymer with the formula [Fe(Htrz)2(trz)](BF4) is presented. For the synthesis of the SCO NPs, the reverse micelle method was used, while the gold nanorods (AuNRs) were prepared with the aspect ratio AR = 6.0 using the seeded-growth method and a binary surfactant mixture composed of cetyltrimethylammonium bromide (CTAB) and sodium oleate (NaOL). The final nanocomposite was prepared using the heteroaggregation method of combining different amounts of SCO NPs with the AuNRs. The nonlinear optical (NLO) properties of the hybrid AuNRs coated with different amounts of SCO NPs were studied in detail by means of the Z-scan technique, revealing that the third-order NLO properties of the AuNRs@SCO are dependent on the amount of SCO NPs grafted onto them. However, due to the resonant nature of the excitation, SCO-induced NLO switching was not observed.

Localized surface plasmon resonance-based abscisic acid biosensor using aptamer-functionalized gold nanoparticles.

Project description:Abscisic acid (ABA) plays an important role in abiotic stress response and physiological signal transduction resisting to the adverse environment. Therefore, it is very essential for the quantitative detection of abscisic acid (ABA) due to its indispensable role in plant physiological activities. Herein, a new detection method based on localized surface plasmon resonance (LSPR) using aptamer-functionalized gold nanoparticles (AuNPs) is developed without using expensive instrument and antibody. In the presence of ABA, ABA specifically bind with their aptamers to form the ABA-aptamer complexes with G-quadruplex-like structure and lose the ability to stabilize AuNPs against NaCl-induced aggregation. Meanwhile, the changes of the LSPR spectra of AuNP solution occur and therefore the detection of ABA achieved. Under optimized conditions, this method showed a good linear range covering from 5×10-7 M to 5×10-5 M with a detection limit of 0.33 μM. In practice, the usage of this novel method has been demonstrated by its application to detect ABA from fresh leaves of rice with the relative error of 6.59%-7.93% compared with ELISA bioassay. The experimental results confirmed that this LSPR-based biosensor is simple, selective and sensitive for the detection of ABA. The proposed LSPR method could offer a new analytical platform for the detection of other plant hormones by changing the corresponding aptamer.
