Project description:Uncontrolled bleeding is the main cause of mortality from trauma. Collagen has been developed as an important hemostatic material due to its platelet affinity function. A bath sponge skeleton is rich in collagen, also known as spongin. To understand the hemostatic effect of spongin, spongin materials, SX, SFM and SR were prepared from the bath sponge Spongia officinalis, and hemostatic experiments were performed. The SX, SFM and SR were significantly better than the positive control, type I collagen, in shortening the whole blood clotting time in vitro and hemostasis upon rat tail amputation. In a hemostatic experiment of rabbit common carotid artery injury, the hemostatic time and 3 h survival rate of the SFM group were 3.00 ± 1.53 min and 100%, respectively, which are significantly better than those of the commercial hemostat CELOX-A (10.33 ± 1.37 min and 67%, respectively). Additionally, the SFM showed good coagulation effects in platelet-deficient blood and defibrinated blood, while also showing good biocompatibility. Through a variety of tests, we speculated that the hemostatic activity of the SFM is mainly caused by its hyperabsorbency, high affinity to platelets and high effective concentration. Overall, the SFM and spongin derivates could be potential hemostatic agents for uncontrolled bleeding and hemorrhagic diseases caused by deficiency or dysfunction of coagulation factors.

Project description:Secondary marine adaptation is a major pattern in amniote evolution, accompanied by specific bone histological adaptations. In the aftermath of the end-Permian extinction, diverse marine reptiles evolved early in the Triassic. Plesiosauria is the most diverse and one of the longest-lived clades of marine reptiles, but its bone histology is least known among the major marine amniote clades. Plesiosaurians had a unique and puzzling body plan, sporting four evenly shaped pointed flippers and (in most clades) a small head on a long, stiffened neck. The flippers were used as hydrofoils in underwater flight. A wide temporal, morphological, and morphometric gap separates plesiosaurians from their closest relatives (basal pistosaurs, Bobosaurus). For nearly two centuries, plesiosaurians were thought to appear suddenly in the earliest Jurassic after the end-Triassic extinctions. We describe the first Triassic plesiosaurian, from the Rhaetian of Germany, and compare its long bone histology to that of later plesiosaurians sampled for this study. The new taxon is recovered as a basal member of the Pliosauridae, revealing that diversification of plesiosaurians was a Triassic event and that several lineages must have crossed into the Jurassic. Plesiosaurian histology is strikingly uniform and different from stem sauropterygians. Histology suggests the concurrent evolution of fast growth and an elevated metabolic rate as an adaptation to cruising and efficient foraging in the open sea. The new specimen corroborates the hypothesis that open ocean life of plesiosaurians facilitated their survival of the end-Triassic extinctions.

Project description:Microtubule assembly is vital for many fundamental cellular processes. Current models for microtubule assembly kinetics assume that the subunit dissociation rate from a microtubule tip is independent of free subunit concentration. Total-Internal-Reflection-Fluorescence (TIRF) microscopy experiments and data from a laser tweezers assay that measures in vitro microtubule assembly with nanometer resolution, provides evidence that the subunit dissociation rate from a microtubule tip increases as the free subunit concentration increases. These data are consistent with a two-dimensional model for microtubule assembly, and are explained by a shift in microtubule tip structure from a relatively blunt shape at low free concentrations to relatively tapered at high free concentrations. We find that because both the association and the dissociation rates increase at higher free subunit concentrations, the kinetics of microtubule assembly are an order-of-magnitude higher than currently estimated in the literature.

Project description:Access to carbocyclic C-nucleosides (CC-Ns) is currently restricted. The few methods available to make CC-Ns suffer from long syntheses and poor modularity, hindering the examination of potentially important chemical space. Here we report an approach to CC-Ns which uses an asymmetric Suzuki-Miyaura type reaction as the key C-C bond forming step. After coupling the densely functionalized racemic bicyclic allyl chloride and heterocyclic boronic acids, the trisubstituted cyclopentenyl core is elaborated to RNA analogues via a hydroborylation-homologation-oxidation sequence. We demonstrate that the approach can be used to produce a variety of enantiomerically enriched CC-Ns, including a carbocyclic derivative of Showdomycin.

Project description:Allylic cyclitols were investigated as covalent inhibitors of glycoside hydrolases by chemical, enzymatic, proteomic, and computational methods. This approach was inspired by the C7 cyclitol natural product streptol glucoside, which features a potential carbohydrate leaving group in the 4-position (carbohydrate numbering). To test this hypothesis, carbocyclic inhibitors with leaving groups in the 4- and 6- positions were prepared. The results of enzyme kinetics analyses demonstrated that dinitrophenyl ethers covalently inhibit α-glucosidases of the GH13 family without reactivation. The labeled enzyme was studied by proteomics, and the active site residue Asp214 was identified as modified. Additionally, computational studies, including enzyme homology modeling and density functional theory (DFT) calculations, further delineate the electronic and structural requirements for activity. This study demonstrates that previously unexplored 4- and 6-positions can be exploited for successful inhibitor design.

Project description:Antimicrobial nanobunches with different amounts of chitosan-capped Ag were prepared by continuous gas-liquid green route under ultrasound irradiation. Spark-produced aerosol Cu nanoparticles were directly injected into an ultrasound Ag(I)-chitosan reaction cell for efficient hydrosolization of the Cu particles and the subsequent incorporation of Ag and chitosan on Cu. Subsequently, electrospraying was used to form of chitosan-capped Cu-Ag nanobunch coatings. The time required for reducing the bacterial proliferation to 50% dropped to ~1?h at a nanobunch concentration of 10??g mL(-1) from the 2.0?min Ag(I) reaction time, and was further decreased to ~0.5?h by increasing the concentration of the nanobunches to 90??g mL(-1). The nanobunches were directly coated onto the substrate using an electrospray device to fabricate transparent films and composite fibers. The antimicrobial activity of the composite carbon fibers was then evaluated via the disc diffusion method.

Project description:The mineralogy and geochemistry associated with Rodinian assembly (~1.3-0.9?Ga) are significantly different from those of other supercontinents. Compared to other supercontinents, relatively more Nb-bearing minerals, Y-bearing minerals, and zircons formed during Rodinian assembly, with corresponding enrichments of Nb, Y, and Zr concentrations in igneous rocks. By contrast, minerals bearing many other elements (e.g., Ni, Co, Au, Se, and platinum group elements) are significantly less abundant, without corresponding depletion of Ni and Co concentrations in igneous rocks. Here we suggest that the Nb, Y, and Zr enrichments in igneous rocks and relatively more occurrences of corresponding Nb-bearing minerals, Y-bearing minerals, and zircons result from significant non-arc magmatism during the mid-Proterozoic, while fewer occurrences of many other minerals suggest enhanced erosion of Rodinian volcanic arcs and orogens. The prolonged, extrovert assembly of Rodinia from thickened mid-Proterozoic continental crust via two-sided subduction can account for both the prevalence of non-arc magmatism and the enhanced erosion.

Project description:The natural nucleoside antibiotic, bredinin, exhibits antiviral and other biological activities. While various nucleosides related to bredinin have been synthesized, its carbocyclic analog has remained unknown. Synthesis of this heretofore unknown analog of bredinin is described. The key precursor, (3aS,4R,6R,6aR)-6-((methoxy-methoxy)methyl)-2,2-dimethyltetrahydro-3aH-cyclopenta[d][1,3]dioxol-4-amine (5), was prepared from the commercially available compound, (1R,4S)-2-azabicyclo[2.2.1] hept-5-en-3-one (4). Our initial approach used intermediate 6, derived in three transformations from 5, for the key photolytic step to produce the desired ring-opened precursor to the target compound. This photochemical transformation was unsuccessful. However, an appropriately protected and related precursor was synthesized from 5 through the following side-chain functional group transformations: elaboration of the amino group through malonyl ester formation, oximation at the central carbon, conversion of ester to amide and catalytic reduction of the oxime group. This precursor, on treatment with triethylorthoformate and catalytic acetic acid in ethanol, underwent cyclization to produce the desired 4-carbamoyl-imidazolium-5-olate ring. Deprotection of the latter product proceeded smoothly to give the carbocyclic analog of bredinin. This target molecule exhibits antiviral activity, albeit low, against a number of RNA viruses. Further biological evaluations are in progress.

Project description:It is well-recognized that block copolymer self-assembly in solution typically produces spheres, worms or vesicles, with the relative volume fraction of each block dictating the copolymer morphology. Stimulus-responsive diblock copolymers that can undergo either sphere/worm or vesicle/worm transitions are also well-documented. Herein we report a new amphiphilic diblock copolymer that can form spheres, worms, vesicles or lamellae in aqueous solution. Such self-assembly behavior is unprecedented for a single diblock copolymer of fixed composition yet is achieved simply by raising the solution temperature from 1 °C (spheres) to 25 °C (worms) to 50 °C (vesicles) to 70 °C (lamellae). Heating increases the degree of hydration (and hence the effective volume fraction) of the core-forming block, with this parameter being solely responsible for driving the sphere-to-worm, worm-to-vesicle and vesicle-to-lamellae transitions. The first two transitions exhibit excellent reversibility but the vesicle-to-lamellae transition exhibits hysteresis on cooling. This new thermoresponsive diblock copolymer provides a useful model for studying such morphological transitions and is likely to be of significant interest for theoretical studies.
