Microfluidic devices integrating microcavity surface-plasmon-resonance sensors: glucose oxidase binding-activity detection.

ABSTRACT: We have developed miniature (approximately 1 microm diameter) microcavity surface-plasmon-resonance sensors (MSPRS), integrated them with microfluidics, and tested their sensitivity to refractive-index changes. We tested their biosensing capability by distinguishing the interaction of glucose oxidase (M(r) 160 kDa) with its natural substrate (beta-D-glucose, M(r) 180 Da) from its interactions with nonspecific substrates (L-glucose, D-mannose, and 2-deoxy-D-glucose). We ran the identical protocol we had used with the MSPRS on a Biacore 3000 instrument using their bare gold chip. Only the MSPRS was able to detect beta-D-glucose binding to glucose oxidase. Each MSPRS can detect the binding to its surface of fewer than 35,000 glucose oxidase molecules (representing 9.6 fg or 60 zmol of protein), about 10(6) times fewer than classical surface-plasmon-resonance biosensors.

Electrochemical surface plasmon resonance and waveguide-enhanced glucose biosensing with N-alkylaminated polypyrrole/glucose oxidase multilayers.

Project description:In this work, we report an electrochemical surface plasmon resonance/waveguide (EC-SPR/waveguide) glucose biosensor that could detect enzymatic reactions in a conducting polymer/glucose oxidase (GO(x)) multilayer thin film. In order to achieve a controlled enzyme electrode and waveguide mode, GO(x) (negatively charged) was immobilized with a water-soluble, conducting N-alkylaminated polypyrrole (positively charged) using the layer-by-layer (LbL) electrostatic self-assembly technique. The electrochemical and optical signals were simultaneously obtained from the composite LbL enzyme electrode upon the addition of glucose as mediated by the electroactivity and electrochromic property of the polypyrrole layers. Signal enhancement in EC-SPR detection is obtained by monitoring the doping-dedoping events on the polypyrrole. The real-time optical signal could be distinguished between the change in the dielectric constant of the enzyme layer and other nonenzymatic reaction events such as adsorption of glucose and the change of the refractive index of the solution. This was possible by correlation of both the SPR mode and the m = 0 and 1 modes of the waveguide in an SPR/waveguide spectroscopy experiment.

Nanostructured surface plasmon resonance sensors: Toward narrow linewidths.

Project description:Surface plasmon resonance sensors have found wide applications in optical sensing field due to their excellent sensitivity to the slight refractive index change of surrounding medium. However, the intrinsically high optical losses in metals make it nontrivial to obtain narrow resonance spectra, which greatly limits the performance of surface plasmon resonance sensors. This review first introduces the influence factors of plasmon linewidths of metallic nanostructures. Then, various approaches to achieve narrow resonance linewidths are summarized, including the fabrication of nanostructured surface plasmon resonance sensors supporting surface lattice resonance/plasmonic Fano resonance or coupling with a photonic cavity, the preparation of surface plasmon resonance sensors with ultra-narrow resonators, as well as strategies such as platform-induced modification, alternating different dielectric layers, and the coupling with whispering-gallery-modes. Lastly, the applications and some existing challenges of surface plasmon resonance sensors are discussed. This review aims to provide guidance for the further development of nanostructured surface plasmon resonance sensors.

Metal Nanoparticles/Porous Silicon Microcavity Enhanced Surface Plasmon Resonance Fluorescence for the Detection of DNA.

Project description:A porous silicon microcavity (PSiMC) with resonant peak wavelength of 635 nm was fabricated by electrochemical etching. Metal nanoparticles (NPs)/PSiMC enhanced fluorescence substrates were prepared by the electrostatic adherence of Au NPs that were distributed in PSiMC. The Au NPs/PSiMC device was used to characterize the target DNA immobilization and hybridization with its complementary DNA sequences marked with Rhodamine red (RRA). Fluorescence enhancement was observed on the Au NPs/PSiMC device substrate; and the minimum detection concentration of DNA ran up to 10 pM. The surface plasmon resonance (SPR) of the MC substrate; which is so well-positioned to improve fluorescence enhancement rather the fluorescence enhancement of the high reflection band of the Bragg reflector; would welcome such a highly sensitive in biosensor.

Bovine serum albumin detection by using molecularly imprinted surface plasmon resonance sensors.

Project description:Molecular imprinted polymers (MIP) have key-lock pattern binding properties specific to the size and shape of target molecules. In this study, we have prepared detection platforms based on a molecularly imprinted surface plasmon resonance (SPR) sensor that can detect bovine serum albumin (BSA) sensitively, selectively, quickly, and in real time. The polymeric film prepared on the SPR sensor surface by molecular imprinting method was obtained by selecting the N-methacryloyl-(L)-glutamic acid molecule as a suitable functional monomer using ultraviolet polymerization. Three different imprinting methods, such as epitope, bulk, and surface imprinting methods, were used to examine the imprinting efficiency. Real-time measurements were performed with BSA imprinted SPR sensor provide linearity in the concentration range from 0.10 to 7.50 nM and indicate a detection limit value of 0.015 nM. Furthermore, we performed the selectivity experiments, where transferrin and hemoglobin were chosen as competitor agents. Overall, the SPR sensor prepared by the epitope imprinting approach has been found to be highly selective and sensitive for bovine serum albumin. To statistically assess the reusability of the sensor, intraday experiments were tested three times with five replicates. The RSD% value less than <1.3 indicates high reproducibility for both sensor production and reproducibility of the method. Validation studies were carried out via enzyme-linked immunosorbent analysis technique (ELISA) in order to demonstrate the applicability of the BSA imprinted SPR sensor. Due to their features such as reusability, fast response time, and ease of use, these SPR sensors, which could be used as an alternative to albumin monitoring approaches, can also be adapted to detect and monitor other proteins in real time.

Sensitivity Comparison of Surface Plasmon Resonance and Plasmon-Waveguide Resonance Biosensors.

Project description:Plasmon-waveguide resonance (PWR) sensors are particularly useful for investigation of biomolecular interactions with or within lipid bilayer membranes. Many studies demonstrated their ability to provide unique qualitative information, but the evaluation of their sensitivity as compared to other surface plasmon resonance (SPR) sensors has not been broadly investigated. We report here a comprehensive sensitivity comparison of SPR and PWR biosensors for the p-polarized light component. The sensitivity of five different biosensor designs to changes in refractive index, thickness and mass are determined and discussed. Although numerical simulations show an increase of the electric field intensity by 30-35 % and the penetration depth by four times in PWR, the waveguide-based method is 0.5 to 8 fold less sensitive than conventional SPR in all considered analytical parameters. The experimental results also suggest that the increase in the penetration depth in PWR is made at the expense of the surface sensitivity. The physical and structural reasons for PWR sensor limitations are discussed and a general viewpoint for designing more efficient SPR sensors based on dielectric slab waveguides is provided.

Applying Machine Learning with Localized Surface Plasmon Resonance Sensors to Detect SARS-CoV-2 Particles.

Project description:The sudden outbreak of COVID-19 rapidly developed into a global pandemic, which caused tens of millions of infections and millions of deaths. Although SARS-CoV-2 is known to cause COVID-19, effective approaches to detect SARS-CoV-2 using a convenient, rapid, accurate, and low-cost method are lacking. To date, most of the diagnostic methods for patients with early infections are limited to the detection of viral nucleic acids via polymerase chain reaction (PCR), or antigens, using an enzyme-linked immunosorbent assay or a chemiluminescence immunoassay. This study developed a novel method that uses localized surface plasmon resonance (LSPR) sensors, optical imaging, and artificial intelligence methods to directly detect the SARS-CoV-2 virus particles without any sample preparation. The virus concentration can be qualitatively and quantitatively detected in the range of 125.28 to 106 vp/mL through a few steps within 12 min with a limit of detection (LOD) of 100 vp/mL. The accuracy of the SARS-CoV-2 positive or negative assessment was found to be greater than 97%, and this was demonstrated by establishing a regression machine learning model for the virus concentration prediction (R2 > 0.95).

A method for the controllable fabrication of optical fiber-based localized surface plasmon resonance sensors.

Project description:Optical fiber-based Localized Surface Plasmon Resonance (OF-LSPR) biosensors have emerged as an ultra-sensitive miniaturized tool for a great variety of applications. Their fabrication by the chemical immobilization of gold nanoparticles (AuNPs) on the optic fiber end face is a simple and versatile method. However, it can render poor reproducibility given the number of parameters that influence the binding of the AuNPs. In order to develop a method to obtain OF-LSPR sensors with high reproducibility, we studied the effect that factors such as temperature, AuNPs concentration, fiber core size and time of immersion had on the number and aggregation of AuNPs on the surface of the fibers and their resonance signal. Our method consisted in controlling the deposition of a determined AuNPs density on the tip of the fiber by measuring its LSPR signal (or plasmonic signal, Sp) in real-time. Sensors created thus were used to measure changes in the refractive index of their surroundings and the results showed that, as the number of AuNPs on the probes increased, the changes in the Sp maximum values were ever lower but the wavelength shifts were higher. These results highlighted the relevance of controlling the relationship between the sensor composition and its performance.

A Genetic Algorithm for Universal Optimization of Ultrasensitive Surface Plasmon Resonance Sensors with 2D Materials.

Project description:We present a general optimization technique for surface plasmon resonance, (SPR) yielding a range of ultrasensitive SPR sensors from a materials database with an enhancement of ∼100%. Applying the algorithm, we propose and demonstrate a novel dual-mode SPR structure coupling SPP and a waveguide mode within GeO2 featuring an anticrossing behavior and an unprecedented sensitivity of 1364 deg/RIU. An SPR sensor operating at wavelengths of 633 nm having a bimetal Al/Ag structure sandwiched between hBN can achieve a sensitivity of 578 deg/RIU. For a wavelength of 785 nm, we optimized a sensor as a Ag layer sandwiched between hBN/MoS2/hBN heterostructures achieving a sensitivity of 676 deg/RIU. Our work provides a guideline and general technique for the design and optimization of high sensitivity SPR sensors for various sensing applications in the future.

Metal-coated microsphere monolayers as surface plasmon resonance sensors operating in both transmission and reflection modes.

Project description:Metal-coated microsphere monolayers (MCM) are a class of plasmonic crystals consisting of noble metal films over arrays of self-assembled colloidal microspheres. Despite their ease of fabrication and tunable plasmonic response, their optical sensing potential has been scarcely explored. Here, silver coated polystyrene sphere monolayers are proposed as surface plasmon resonance sensors capable of functioning in both transmission (T) and reflection (R) readout modes. An original and key point is the use of ~200?nm colloids, smaller than in MCM studied before. It allowed us to reveal a previously unobserved, additional/secondary Enhanced Optical Transmission band, which can be exploited in sensing, with higher sensitivity than the better-known main transmission band. The reflection configuration however, is almost an order of magnitude more efficient for sensing than the transmission one. We also evidenced a strong impact of the adsorbate location on the metal surface on the sensing efficiency. Electric field distribution analysis is performed to explain these results. Proof-of-concept experiments on the detection of 11-MUA molecular monolayers, performed in both readout modes, confirm the behaviors observed through FDTD simulations. Results in this paper can serve as guidelines for designing optimized sensors based on metal-coated colloidal monolayers, and more generally for plasmonic sensors based on metal nanostructured films.

Flexible hyperspectral surface plasmon resonance microscopy.

Project description:Optical techniques for visualization and quantification of chemical and biological analytes are always highly desirable. Here we show a hyperspectral surface plasmon resonance microscopy (HSPRM) system that uses a hyperspectral microscope to analyze the selected area of SPR image produced by a prism-based spectral SPR sensor. The HSPRM system enables monochromatic and polychromatic SPR imaging and single-pixel spectral SPR sensing, as well as two-dimensional quantification of thin films with the measured resonance-wavelength images. We performed pixel-by-pixel calibration of the incident angle to remove pixel-to-pixel differences in SPR sensitivity, and demonstrated the HSPRM's capabilities by using it to quantify monolayer graphene thickness distribution, inhomogeneous protein adsorption and single-cell adhesion. The HSPRM system has a wide spectral range from 400 nm to 1000 nm, an optional field of view from 0.884 mm2 to 0.003 mm2 and a high lateral resolution of 1.2 μm, demonstrating an innovative breakthrough in SPR sensor technology.
