ABSTRACT: A reliable, high yielding cyclization protocol for the macrocycle of tamandarin B is presented. This strategy will facilitate the synthesis of side chain analogues.

Macrocycle-assisted synthesis of non-stoichiometric silver(i) halide electrocatalysts for efficient chlorine evolution reaction.

Project description:The electrocatalytic oxidation of chloride to chlorine is a fundamental and important electrochemical reaction in industry. Herein we report the synthesis of non-stoichiometric silver halide nanoparticles through a novel macrocycle-assisted bulk-to-cluster-to-nano transformation. The acquired positively charged nanoparticles expedite chloride transportation by electrostatic attraction and facilitate the formation of silver polychloride catalytic species on the surface, thus functioning as efficient and selective electrocatalysts for the chlorine evolution reaction (CER) at a very low overpotential and within a wide concentration range of chloride. The formation of uncommon non-stoichiometric nanoparticles prevents the formation of a AgCl precipitate and exposes more coordination unsaturated silver atoms to catalyze CER, finally causing a large enhancement of the atomic catalytic efficiency of silver. This study showcases a promising approach to achieve efficient catalysts from a bottom-up design.

Concise Synthesis of Tetrazole Macrocycle.

Project description:A concise two step synthesis of tetrazole containing macrocycles from readily accessible starting materials is presented. The first step comprises a chemoselective amidation of amino acid derived isocyanocarboxylicacid esters with unprotected symmetrical diamines to afford diverse ?-isocyano-?-amines. In the second step, the ?-isocyano-?-amines undergo an Ugi tetrazole reaction to close the macrocycle. Advantageously, this strategy allows short access to 11-19-membered macrocycles in which substituents can be independently varied at three different positions.

Two-Step Macrocycle Synthesis by Classical Ugi Reaction.

Project description:The direct nonpeptidic macrocycle synthesis of α-isocyano-ω-amines via the classical Ugi four-component reaction (U-4CR) is introduced. Herein an efficient and flexible two-step procedure to complex macrocycles is reported. In the first step, the reaction between unprotected diamines and isocyanocarboxylic acids gives high diversity of unprecedented building blocks in high yield. In the next step, the α-isocyano-ω-amines undergo a U-4CR with high diversity of aldehydes and carboxylic acids in a one-pot procedure. This synthetic approach is short and efficient and leads to a wide range of macrocycles with different ring sizes.

Total Synthesis of Njaoamine C by Concurrent Macrocycle Formation.

Project description:In conceptual terms, the first total synthesis of the cytotoxic marine natural product njaoamine C differs from all known approaches toward related alkaloids of the manzamine superfamily in that both macrocyclic rings enveloping the diazatricyclic core are concomitantly formed; this goal was reached by double ring closing alkyne metathesis (dRCAM). The success of this maneuver does not merely reflect a favorable preorientation of the four alkyne chains that need to be concatenated in the proper pairwise manner but is also the outcome of dynamic covalent chemistry involving error correction by the chosen "canopy" molybdenum alkylidyne catalyst. The end game downstream of dRCAM capitalizes on the striking chemoselectivity of palladium-catalyzed hydrostannation, which selects for (hetero)arylalkynes even in the presence of sterically much more accessible dialkylalkynes or alkenes; for this preference, the method complements the classical repertoire of hydrometalation and semireduction reactions.

Oxidative carbocation formation in macrocycles: synthesis of the neopeltolide macrocycle.

Project description:Macrocyclic oxocarbenium ions can be formed from macrolactones that contain benzylic or allylic ether groups through oxidative carbon-hydrogen-bond activation mediated by 2,3-dichloro-4,5-dicyanoquinone (DDQ). The applicability of this efficient reaction to complex-molecule synthesis was demonstrated by its use in a brief formal synthesis of neopeltolide (see retrosynthetic scheme) to form the tetrahydropyrone ring.

Synthesis of a sucrose-based macrocycle with unsymmetrical monosaccharides "arms".

Project description:An efficient methodology for the selective substitution of both terminal positions (C6 and C6') in 1',2,3,3',4,4'-hexa-O-benzylsucrose with different unsaturated monosaccharide units is presented. Such a highly functionalized intermediate was cyclized under RCM conditions to afford a macrocyclic derivative containing a 31-membered ring in 26% yield.

Synthesis of a Macrocycle Based on Linked Amino Acid Mimetics (LAAM).

Project description:We report the synthesis of a macrocycle utilizing a novel framework of standard amino acids in combination with subunits that we have named as Linked Amino Acid Mimetics (LAAM's). Macrocycles based on the LAAM concept provide both a peptide targeting region and two independently variable functional regions. In the prototype structure, the commonly known Arg-Gly-Asp (RGD) sequence was used for the targeting region. The functional regions contain a phenyl group, and the linkage was formed via a Ring-Closing Metathesis (RCM) reaction.

Recognition Site Modifiable Macrocycle: Synthesis, Functional Group Variation and Structural Inspection.

Project description:Traditional macrocyclic molecules encode recognition sites in their structural backbones, which limits the variation of the recognition sites and thus, would restrict the adjustment of recognition properties. Here, we report a new oligoamide-based macrocycle capable of varying the recognition functional groups by post-synthesis modification on its structural backbone. Through six steps of common reactions, the parent macrocycle (9) can be produced in gram scale with an overall yield of 31%. The post-synthesis modification of 9 to vary the recognition sites are demonstrated by producing four different macrocycles (10-13) with distinct functional groups, 2-methoxyethoxyl (10), hydroxyl (11), carboxyl (12) and amide (13), respectively. The 1H NMR study suggests that the structure of these macrocycles is consistent with our design, i.e., forming hydrogen bonding network at both rims of the macrocyclic backbone. The 1H-1H NOESY NMR study indicates the recognition functional groups are located inside the cavity of macrocycles. At last, a preliminary molecular recognition study shows 10 can recognize n-octyl-β-D-glucopyranoside (14) in chloroform.

Formal synthesis of (-)-kendomycin featuring a Prins-cyclization to construct the macrocycle.

Project description:The kendomycin skeleton was prepared by a highly convergent strategy in which the benzofuran fragment and the acyclic iodide fragment were prepared by standard methods and joined using a Suzuki coupling. The distinctive reaction in our approach was an intramolecular Prins cyclization that assembles the macrocyclic ring in good yield. Modeling studies demonstrate that the acyclic chain is predisposed for macrocycle formation. Ultimately, the product was correlated with one of Lee's advanced intermediates for a formal total synthesis of kendomycin.
