Project description:Common bottlenose dolphins serve as sentinels for the health of their coastal environments as they are susceptible to health impacts from anthropogenic inputs through both direct exposure and food web magnification. Remote biopsy samples have been widely used to reveal contaminant burdens in free-ranging bottlenose dolphins, but do not address the health consequences of this exposure. To gain insight into whether remote biopsies can also identify health impacts associated with contaminant burdens, we employed RNA sequencing (RNA-seq) to interrogate the transcriptomes of remote skin biopsies from 116 bottlenose dolphins from the northern Gulf of Mexico and southeastern U.S. Atlantic coasts. Gene expression was analyzed using principal component analysis, differential expression testing, and gene co-expression networks, and the results correlated to season, location, and contaminant burden. Season had a significant impact, with over 30% of genes differentially expressed between spring/summer and winter months. Geographic location exhibited lesser effects on the transcriptome, with 15% of genes differentially expressed between the northern Gulf of Mexico and the southeastern U.S. Atlantic locations. Despite a large overlap between the seasonal and geographical gene sets, the pathways altered in the observed gene expression profiles were somewhat distinct. Co-regulated gene modules and differential expression analysis both identified epidermal development and cellular architecture pathways to be expressed at lower levels in animals from the northern Gulf of Mexico. Although contaminant burdens measured were not significantly different between regions, some correlation with contaminant loads in individuals was observed among co-expressed gene modules, but these did not include classical detoxification pathways. Instead, this study identified other, possibly downstream pathways, including those involved in cellular architecture, immune response, and oxidative stress, that may prove to be contaminant responsive markers in bottlenose dolphin skin.

Project description:The aim of this study was to generate an easy to use index to evaluate the ecological state of agricultural land from a sustainability perspective. We selected environmental indicators, such as the use of organic soil amendments (green manure) versus chemical fertilizers, plant biodiversity (including crop associations), variables which characterize soil conservation of conventional agricultural systems, pesticide use, method and frequency of tillage. We monitored the ecological state of 52 agricultural plots to test the performance of the index. The variables were hierarchically aggregated with simple mathematical algorithms, if-then rules, and rule-based fuzzy models, yielding the final multi-criteria index with values from 0 (worst) to 1 (best conditions). We validated the model through independent evaluation by experts, and we obtained a linear regression with an r2 = 0.61 (p = 2.4e-06, d.f. = 49) between index output and the experts' evaluation.

Project description:A new tiger beetle species, Cicindelidiamelissa Duran & Roman, sp. n., of the tribe Cicindelini, is described from high elevation montane forests of southeastern Arizona and Mexico. It appears to be most closely related to Cicindelidianebuligera (Bates) but is distinguished on the basis of multiple morphological characters and geographic range. The new species is also superficially similar to the widespread Cicindelidiasedecimpunctata (Klug), but distinguished on the basis of multiple morphological characters and habitat. Habitus, male and female reproductive structures, and known distribution map are presented.

Project description:From January to May 2015, a sample of 50 individuals of the Pacific sierra Scomberomorus sierra Jordan and Starks, 1895 captured off Mazatlán (southeastern Gulf of California, Mexico) were reviewed for helminths. A total of 6, 255 parasitic worms belonging to 11 species (9 in adult stage and 2 larvae) were obtained. Trematoda was the best represented group with 6 species of Didymozoidae Monticelli, 1888 and 1 of Bucephalidae Poche, 1907. In addition, 2 monogenean species and 2 of Nematoda were collected. In this study, the first molecular sequences for didymozoid species in Mexico were generated, and for Glomeritrema sp. at worldwide level. The most prevalent species were Didymocylindrus sp. (92 %) and Didymocystis scomberomori (MacCallum & MacCallum, 1916) (88 %), whereas the monogenean Thoracocotyle crocea MacCallum, 1913 reached the highest value of mean intensity (75.2). The coincidence between the helminthological composition established in our study and that reported for the same scombrid in 4 localities from the Mexican South Pacific (sharing 10 species) suggests that this group of species persistently parasitize S. sierra throughout its distribution along the Mexican Pacific coast; furthermore, due to the richness of didymozoids and the affinity of Thoracocotylidae species for S. sierra, both groups can be considered typical parasites of this fi sh.

Project description:Leishmaniasis, a vector-borne disease transmitted to humans through the bite of phlebotomine sand flies, is of public health significance in southeastern Mexico. Active and continuous monitoring of vectors is an important aspect of disease control for the prediction of potential outbreaks. Thus, the correct identification of vectors is paramount in this regard. In this study, we employed DNA barcoding as a tool for identifying phlebotomine sand flies collected in localized cutaneous leishmaniasis endemic areas of Quintana Roo, Mexico. Specimens were collected using CDC light and Shannon traps as part of the Mexican Ministry of Health surveillance program. DNA extraction was carried out using a nondestructive protocol, and morphological identification based on taxonomic keys was conducted on slide-mounted specimens. Molecular taxonomic resolution using the 658-bp fragment of the mitochondrial cytochrome c oxidase subunit 1 (cox1) gene was 100% congruent with the morphological identification. Seven species were identified: Lutzomyia cruciata (Coquillett 1907), Lutzomyia longipalpis (Lutz & Neiva 1912), Psathyromyia shannoni (Dyar 1929), Dampfomyia deleoni (Fairchild & Hertig 1947), Dampfomyia beltrani/steatopyga (Vargas & Díaz-Nájera 1951), Bichromomyia olmeca olmeca (Vargas & Díaz-Nájera, 1959), and Brumptomyia mesai (Sherlock 1962). Mean intraspecific divergence ranged from 0.12% to 1.22%, while interspecific distances ranged from 11.59% to 19.29%. Neighbor-joining (NJ) analysis using the Kimura 2-parameter model also showed specimens of the same species to be clustered together. The study provides the first cox1 sequences for three species of sand flies and indicates the utility of DNA barcoding for phlebotomine sand flies species identification in southeastern Mexico.

Project description:The role of invasive species in ecosystem functioning represents one of the main challenges in ecology. Pteridium aquilinum is a successful cosmopolitan invasive species with negative effects on the ecological mechanisms that allow secondary succession. In this study, we evaluated the influence of P. aquilinumon secondary succession under different disturbances in a seasonal dry forest of the Yucatán Peninsula. We determined species richness, composition and the relative importance value in four sampling units. Fabaceae followed by Asteraceae, Meliaceae, Rubiaceae, Sapindaceae and Verbenaceae were the most species rich families. A dissimilarity analysis determined significant differences in beta diversity between sampling units. With a generalized linear model we found that species richness was best explained by site conditions, followed by calcium and soil organic matter. Also, the generalized linear model showed that abundance resulted in a strong correlation with site conditions and soil characteristics. Specific soil conditions related to phosphoro and calcium were also detected as beneficiary to the successional processes. Our results suggest that applying fire restriction and periodic cutting of the bracken fern, this can increase a higher diversity of species.

Project description:AimsIn this research we assessed the prevalence of prediabetes and type 2 diabetes and its association with social determinants such as indigenous origin and residence area in population from Comitan, Chiapas, Mexico.MethodsThe Comitan Study is a population-based study carried out from 2010 to 2012 that included 1844 participants aged ≥ 20 years, 880 indigenous and 964 nonindigenous participants. Ethnicity was ascertained by self-report and speaking an indigenous language was also recorded. Prediabetes was defined as fasting serum glucose 5.6-6.9 mmol/l or 2-hour post load serum glucose 7.8-11.0 mmol/l. Type 2 diabetes was defined as fasting serum glucose ≥ 7.0 mmol/l or 2-h post load serum glucose ≥ 11.1 mmol/l or previous clinical diagnosis.ResultsAge-sex-adjusted prevalence of prediabetes and type 2 diabetes was 18.0% (95%CI 15.3-20.6) and 11.0% (95%CI 8.9-131.1) in nonindigenous and 10.6% (95%CI 8.4-12.7) and 4.7% (95%CI 3.3-6.1) in indigenous individuals, respectively. After stratifying by ethnicity, in both indigenous and nonindigenous participants the probability of prediabetes and type 2 diabetes increased with age and BMI. In both indigenous and nonindigenous participants the probability of type 2 diabetes was lower in those living in rural compared with urban areas.ConclusionsThe prevalence of prediabetes and type 2 diabetes was significantly lower in indigenous than in nonindigenous participants. Also, the prevalence of type 2 diabetes was lower in those living in rural areas. Health benefits of a traditional lifestyle may partially account for these differences.

Project description:In 2015, we detected clinical cases of babesiosis caused by Babesia microti in Yucatán State, Mexico. Cases occurred in 4 children from a small town who became ill during the same month. Diagnosis was confirmed using conventional PCR followed by sequencing of the DNA fragment obtained.
