ABSTRACT: In the crystal structure of the title compound, C(21)H(15)N(5)O(2), the planar indolone unit and the pyran ring are almost perpendicular to each other [dihedral angle = 89.41?(2)°], and the pyrazole and phenyl rings are oriented at an angle of 25.74?(1)°. The mol-ecular packing is stabilized by inter- and intra-molecular C-H?O, N-H?O and C-H?? hydrogen bonds.

Project description:The crystal structure of the title compound, C(25)H(18)ClN(5)O, was determined in the course of our studies on the synthesis of 1,4-dihydro-pyrano[2,3-c]pyrazole as an inhibitor of the p38 mitogen-activated protein kinase (MAPK). The compound was prepared via a base-catalysed synthesis from 1-benzyl-3-(4-pyrid-yl)-1H-pyrazol-5(4H)-one with p-chloro-aldehyde and malononitrile. The crystal data obtained were used to generate a three-dimensional pharmacophore model for in silico database screening. The phenyl ring is disordered over two positions, with site occupancy factors of 0.55 and 0.45. The dihedral angles between the 1,4-dihydropyrano[2,3-c]pyrazole unit and the chloro-phenyl and pyridine rings are 83.7 (1) and 16.0 (1)°, respectively. The chloro-phenyl and pyridine rings make a dihedral angle of 86.8 (2)°.

Project description:The title compound, C(20)H(15)N(5)O(3), was synthesized by the one-pot reaction of a four-component reaction protocol in aqueous medium. The pyrano[2,3-c]pyrazole system is essentially planar, with a maximum deviation of 0.026?(2)?Å. The 3-nitro-phenyl and phenyl rings make dihedral angles of 81.11?(5) and 13.36?(1)°, respectively, with the mean plane of the pyrano[2,3-c]pyrazole ring. The crystal structure is stabilized by N-H?N hydrogen bonds, which form infinite chain propagating along the c axis and by N-H?O hydrogen bonds, which form infinite chains propagating along the a axis. There are also N-O?N-C dipole-dipole inter-actions along the a axis with an O?N distance of 3.061?(3)?Å, which is shorter than that of the N-H?O hydrogen bond [3.196?(3)?Å].

Synthesis, Molecular Docking, and Antimalarial Activity of Hybrid 4-Aminoquinoline-pyrano[2,3-c]pyrazole Derivatives.

Project description:Widespread resistance of Plasmodium falciparum to current artemisinin-based combination therapies necessitate the discovery of new medicines. Pharmacophoric hybridization has become an alternative for drug resistance that lowers the risk of drug-drug adverse interactions. In this study, we synthesized a new series of hybrids by covalently linking the scaffolds of pyrano[2,3-c]pyrazole with 4-aminoquinoline via an ethyl linker. All synthesized hybrid molecules were evaluated through in vitro screenings against chloroquine-resistant (K1) and -sensitive (3D7) P. falciparum strains, respectively. Data from in vitro assessments showed that hybrid 4b displayed significant antiplasmodial activities against the 3D7 strain (EC50 = 0.0130 ± 0.0002 μM) and the K1 strain (EC50 = 0.02 ± 0.01 μM), with low cytotoxic effect against Vero mammalian cells. The high selectivity index value on the 3D7 strain (SI > 1000) and the K1 strain (SI > 800) and the low resistance index value from compound 4b suggested that the pharmacological effects of this compound were due to selective inhibition on the 3D7 and K1 strains. Molecular docking analysis also showed that 4b recorded the highest binding energy on P. falciparum lactate dehydrogenase. Thus, P. falciparum lactate dehydrogenase is considered a potential molecular target for the synthesized compound.

Project description:In the title compound, C(9)H(8)N(2)OS, the benzothio-phene ring is substituted with amino, oxo and carbonitrile groups. The thio-phene ring is essentially planar (r.m.s. deviation = 0.0003?Å), while the cyclo-hexene ring is in a half-chair conformation. In the crystal, N-H?O hydrogen bonds generate chains of mol-ecules in a zigzag pattern along the b axis. Pairs of N-H?N hydrogen bonds form centrosymmetric head-to-head dimers about inversion centres, corresponding to an R(2) (2)(12) graph-set motif. In addition, rather weak N-H?S inter-actions are also present in the structure and the supra-molecular assembly is further consolidated by ?-? stacking inter-actions between the benzothio-phene rings, disposed at a distance of 3.742?(3)?Å.
