Proteomics analysis of Lactobacillus casei Zhang, a new probiotic bacterium isolated from traditional home-made koumiss in Inner Mongolia of China.

Project description:Lactobacillus casei Zhang, isolated from traditional home-made koumiss in Inner Mongolia of China, was considered as a new probiotic bacterium by probiotic selection tests. We carried out a proteomics study to identify and characterize proteins expressed by L. casei Zhang in the exponential phase and stationary phase. Cytosolic proteins of the strain cultivated in de Man, Rogosa, and Sharpe broth were resolved by two-dimensional gel electrophoresis using pH 4-7 linear gradients. The number of protein spots quantified from the gels was 487 +/- 21 (exponential phase) and 494 +/- 13 (stationary phase) among which a total of 131 spots were identified by MALDI-TOF/MS and/or MALDI-TOF/TOF according to significant growth phase-related differences or high expression intensity proteins. Accompanied by the cluster of orthologous groups (COG), codon adaptation index (CAI), and GRAVY value analysis, the study provided a very first insight into the profile of protein expression as a reference map of L. casei. Forty-seven spots were also found in the study that showed statistically significant differences between exponential phase and stationary phase. Thirty-three of the spots increased at least 2.5-fold in the stationary phase in comparison with the exponential phase, including 19 protein spots (e.g. Hsp20, DnaK, GroEL, LuxS, pyruvate kinase, and GalU) whose intensity up-shifted above 3.0-fold. Transcriptional profiles were conducted to confirm several important differentially expressed proteins by using real time quantitative PCR. The analysis suggests that the differentially expressed proteins were mainly categorized as stress response proteins and key components of central and intermediary metabolism, indicating that these proteins might play a potential important role for the adaptation to the surroundings, especially the accumulation of lactic acid in the course of growth, and the physiological processes in bacteria cell.

<i>Lactobacillus casei</i> Zhang exerts probiotic effects to antibiotic-treated rats.

Project description:Probiotics administration can facilitate the restoration of host gut microbiota/metabolome after antibiotic treatment. Yet, the mechanism behind such beneficial effects remains unclear. This study constructed a rat model of antibiotic-induced gut dysbiosis to monitor the effects and mechanism of probiotic (Lactobacillus casei Zhang) treatment in maintaining gut homeostasis and restoring the gut microbiota/metabolome. Forty rats were randomly divided into four groups (n = 10 per group): control receiving only saline (Ctrl), antibiotic (AB-Ctrl), antibiotic followed by probiotic (AB-Prob), and antibiotic plus probiotic followed by probiotic (AB + Prob). Rat fecal microbiota and sera were collected at four time points from pre-treatment to post-treatment. The probiotic-treated group (AB + Prob) had significantly more Parabacteroides (P.) goldsteinii after one week of antibiotic and probiotic intervention but fewer antibiotic resistance genes (ARGs)-possessing bacteria (Clostridioides difficile and Burkholderiales bacterium). Consistently, metabolomics data revealed that both probiotic groups had more acetic acid, propionic acid, butyric acid, and valeric acid post treatment. Moreover, a potential probiotic species, P. goldsteinii, strongly correlated with L. casei, as well as propionic acid, butyric acid, and valeric acid. Furthermore, administering probiotic lowered the serum IL-1α level. In contrast, the antibiotic-recipients had a higher irreversible level of IL-1α, suggesting inflammation of the rats. Thus, antibiotic treatment not only led to host gut dysbiosis, but inflammatory responses and an increase in gut ARGs. Daily L. casei Zhang supplementation could alleviate the side effect of cefdinir intervention and facilitate the restoration of gut microbial homeostasis, and these probiotic effects might involve P. goldsteinii-mediated beneficial activities.

Transcriptome analysis of probiotic Lactobacillus casei Zhang during fermentation in soymilk

Project description:Lactobacillus casei Zhang is a probiotic bacterium isolated from koumiss in Inner Mongolia of China. Gene expression dynamics of L. casei Zhang during growth in soymilk was investigated in attempt to reveal the mechanisms involved in growth stimulation for growing probiotics in. Comparison of different transcripts next to each other revealed 162 and 63 significantly induced genes in late logarithmic phase and stationary phase, of which the expression was at least 3 fold up-regulated and down-regulated, respectively. Approximately, 38.4% of the up-regulated genes were associated with amino acid transport and metabolism notably for histidine and lysine biosynthesis, followed by genes/gene clusters involved in carbohydrate transport and metabolism, lipid transport and metabolism, and inorganic ion transport and metabolism. The analysis results suggest that the stimulatory effect of soymilk-based ecosystem on the L. casei Zhang growth is more complex than amino acids or oligopeptides supply.

Gene expression profile of probiotic Lactobacillus casei Zhang during the late stage of milk fermentation

Project description:Lactobacillus casei Zhang is a probiotic bacterium isolated from koumiss in Inner Mongolia of China that has been commercially used as a starter in the manufacture of dairy products. To study the gene expression profiles of L. casei Zhang during growth in milk, a whole-genome microarray was used. Compared to L. casei Zhang grown to late logarithmic phase in milk, 61 genes were significantly up-regulated (>5 fold) in stationary phase, whereas 26 genes were down-regulated. Collectively, these data showed that the majority of the identified genes was involved in carbohydrate metabolism and energy production, followed by genes involved in nucleotide metabolism, inorganic ion transport, amino acid transport and metabolism, chaperone, etc. This study demonstrates the fundamental effects of cultural conditions on the transcriptome of L. casei Zhang. Moreover, it improves the understanding of the growth and survival mechanism of the bacterium during the late stage of milk fermentation.

Metagenomic and metatranscriptomic profiling of Lactobacillus casei Zhang in the human gut.

Project description:Little is known about the replication and dynamic transcription of probiotics during their "passenger" journey in the human GI tract, which has therefore limited the understanding of their probiotic mechanisms. Here, metagenomic and metatranscriptomic sequencing was used to expose the in vivo expression patterns of the probiotic Lactobacillus casei Zhang (LcZ), which was compared with its in vitro growth transcriptomes, as well as the dynamics of the indigenous microbiome response to probiotic consumption. Extraction of the strain-specific reads revealed that replication and transcripts from the ingested LcZ were increased, while those from the resident L. casei strains remained unchanged. Mapping of all sequencing reads to LcZ genome showed that gene expression in vitro and in vivo differed dramatically. Approximately 39% of mRNAs and 45% of sRNAs of LcZ well-expressed were repressed after ingestion into human gut. The expression of ABC transporter genes and amino acid metabolism genes was induced at day 14 of ingestion, and genes for sugar and SCFA metabolism were activated at day 28 of ingestion. Expression of rli28c sRNA with peaked expression during the in vitro stationary phase was also activated in the human gut; this sRNA repressed LcZ growth and lactic acid production in vitro. However, the response of the human gut microbiome to LcZ was limited and heterogeneous. These findings implicate the ingested probiotic has to change its transcription patterns to survive and adapt in the human gut, and the time-dependent activation patterns indicate highly dynamic cross-talk between the probiotic and human gut microbes.
