ABSTRACT: A protocol for forming a highly active Pd(0) catalyst from Pd(OAc) 2, water, and biaryldialkylphosphine ligands has been developed. This protocol generates a catalyst system, which exhibits excellent reactivity and efficiency in the coupling of a variety of amides and anilines with aryl chlorides.

Palladium-bearing intermetallic electride as an efficient and stable catalyst for Suzuki cross-coupling reactions.

Project description:Suzuki cross-coupling reactions catalyzed by palladium are powerful tools for the synthesis of functional organic compounds. Excellent catalytic activity and stability require negatively charged Pd species and the avoidance of metal leaching or clustering in a heterogeneous system. Here we report a Pd-based electride material, Y3Pd2, in which active Pd atoms are incorporated in a lattice together with Y. As evidenced from detailed characterization and density functional theory (DFT) calculations, Y3Pd2 realizes negatively charged Pd species, a low work function and a high carrier density, which are expected to be beneficial for the efficient Suzuki coupling reaction of activated aryl halides with various coupling partners under mild conditions. The catalytic activity of Y3Pd2 is ten times higher than that of pure Pd and the activation energy is lower by nearly 35%. The Y3Pd2 intermetallic electride catalyst also exhibited extremely good catalytic stability during long-term coupling reactions.

Synergistic Catalyst-Mediator Pairings for Electroreductive Cross-Electrophile Coupling Reactions.

Project description:Electroreductive cross-electrophile coupling (eXEC) represents an attractive approach for the direct C-C coupling of two electrophiles but generally suffers from limited scope compared to reactions with chemical reductants. This work demonstrates that mediator-assisted electrocatalysis is a general strategy for the enhancement of eXEC reactions. While eXEC reactions catalyzed by a variety of widely available ligand-nickel complexes are low yielding when applied to reductive couplings of challenging substrates, reactions with the same complexes generate products in near-quantitative yield when a redox-matched mediator is included. We identify a library of catalyst-mediator systems that provide complementary reactivity and enable coupling of a range of substrate classes in high yields. These catalyst systems are applicable to both chemical and electrochemical reduction, but some require electroreduction due to the low potentials required for activation. Finally, mechanistic studies offer insights that facilitate catalyst-mediator pairing.

A multiligand based Pd catalyst for C-N cross-coupling reactions.

Project description:An alternative approach to catalyst development, which led to a Pd catalyst based on two biarylphosphine ligands for C-N cross-coupling reactions, is reported. By effectively being able to take the form of multiple catalysts this system manifests the best properties that catalysts based on either of the two ligands exhibit separately and displays the highest reactivity and substrate scope of any system that has been reported to date for these reactions.

Palladium Catalyst Recycling for Heck-Cassar-Sonogashira Cross-Coupling Reactions in Green Solvent/Base Blend.

Project description:The identification of a green, versatile, user-friendly, and efficient methodology is necessary to facilitate the use of Heck-Cassar-Sonogashira (HCS) cross-coupling reaction in drug discovery and industrial production in the pharmaceutical segment. The Heck-Cassar and Sonogashira protocols, using N-hydroxyethylpyrrolidone (HEP)/water/N,N,N',N'-tetramethyl guanidine (TMG) as green solvent/base mixture and sulfonated phosphine ligands, allowed to recycle the catalyst, always guaranteeing high yields and fast conversion under mild conditions, with aryl iodides, bromides, and triflates. No catalyst leakage or metal contamination of the final product were observed during the HCS recycling. To our knowledge, a turnover number (TON) up to 2375, a turnover frequency (TOF) of 158 h-1 , and a process mass intensity (PMI) around 7 that decreased around 3 after solvent, base, and palladium recovery, represent one of the best results to date using a sustainable protocol. The Heck-Cassar protocol using sSPhos was successfully applied to the telescoped synthesis of Erlotinib (TON: 1380; TOF: 46 h-1 ).

Heterometallic palladium-iron metal-organic framework as a highly active catalyst for cross-coupling reactions.

Project description:Palladium-based metal-organic frameworks (Pd-MOFs) are an emerging class of heterogeneous catalysts extremely challenging to achieve due to the facile leaching of palladium and its tendency to be reduced. Herein, Pd(ii) was successfully incorporated in the framework of a MOF denoted as MUV-22 using a solvent assisted reaction. This stable MOF, with square-octahedron (soc) topology as MIL-127, and a porosity of 710 m2 g-1, is highly active, selective, and recyclable for the Suzuki-Miyaura allylation of aryl and alkyl boronates as exemplified with the coupling between cinnamyl bromide and Me-Bpin, a typically reluctant reagent in cross-coupling reactions.

Dramatic improvement in catalyst loadings and molar ratios of coupling partners for Ni/Cr-mediated coupling reactions: heterobimetallic catalysts.

Project description:Two new ligands 1a,b are reported. Upon treatment with 1 equiv of NiCl(2) x (MeOCH(2))(2), 1a,b give the corresponding Ni complexes. X-ray analysis of 1a x NiCl(2) established that the NiCl(2) is selectively coordinated to the phenanthroline nitrogens. Ni/Cr heterobimetallic catalysts 1a,b x CrCl(2)/NiCl(2), prepared from 1a,b x NiCl(2), have been shown to behave exceptionally well in catalytic asymmetric Ni/Cr-mediated couplings, with highlights including the following: (1) 1-2 mol % catalyst is sufficient to complete the coupling; (2) only negligible amounts of the dimers, byproducts formed through the alkenyl Ni species, are observed; (3) the coupling goes to completion even with a 1:1 molar ratio of the coupling partners; and (4) the asymmetric induction is practically identical with that obtained from the coupling with the Cr catalysts prepared from (S)-sulfonamides 2a,b. The scope of the new Ni/Cr heterobimetallic catalysts was briefly studied using four additional aldehydes. The applicability of the new catalysts to polyfunctional substrates was demonstrated by two C-C bond formations chosen from the halichondrin/E7389 synthesis as examples.

This magnetic mesoporous compound is characterized by various techniques including FT-IR, BET, VSM, SEM, XRD, and TGA.

Titanium-mediated reductive cross-coupling reactions of imines with terminal alkynes: An efficient route for the synthesis of stereodefined allylic amines.

Project description:Low-valency titanium species, generated in situ by using Ti(OiPr)4/2 c-C5H9MgCl reagent, react with imines to give a titanium-imine complex that can couple with terminal alkynes to provide azatitanacyclopentenes with excellent regioselectivity. Stereodefined allylic amines are obtained in good yields after hydrolysis or iodonolysis of the corresponding azatitanacyclopentenes. When ethynylcyclopropane is used as the coupling partner to react with imines in this reaction, the initially generated allylic amine undergoes an unexpected 1,3-amino migration on silica gel during the column chromatography.

Chemical and Photochemical Water Oxidation Mediated by an Efficient Single-Site Ruthenium Catalyst.

Project description:Water oxidation is a fundamental step in artificial photosynthesis for solar fuels production. In this study, we report a single-site Ru-based water oxidation catalyst, housing a dicarboxylate-benzimidazole ligand, that mediates both chemical and light-driven oxidation of water efficiently under neutral conditions. The importance of the incorporation of the negatively charged ligand framework is manifested in the low redox potentials of the developed complex, which allows water oxidation to be driven by the mild one-electron oxidant [Ru(bpy)3 ]3+ (bpy=2,2'-bipyridine). Furthermore, combined experimental and DFT studies provide insight into the mechanistic details of the catalytic cycle.
