ABSTRACT: The title compound, C(16)H(14)O(3), adopts a conformation in which each functional group is almost coplanar with its adjacent ring, while the two aromatic rings are twisted with respect to one another with a dihedral angle of 78.51?(3)°. The compound dimerizes by standard centrosymmetric hydrogen-bonded carboxyl pairing [O?O = 2.6218?(11)?Å and O-H?O = 176?(2)°]. The packing includes two inter-molecular C-H?O close contacts with the ketone group.

Project description:In the title compound, C(9)H(10)N(2)O(4), an intra-molecular N-H⋯O hydrogen-bond inter-action generates an S(6) ring motif. The nitro group is slightly twisted away from its attached benzene ring [dihedral angle = 15.29 (15)°]. In the crystal structure, mol-ecules are stacked down the a axis caused by short O⋯O(-1-x, -y, 2-z) contacts of 2.6481 (16) Å involving the O atoms of the nitro groups. The crystal packing is consolidated by inter-molecular O-H⋯O hydrogen bonds, linking the mol-ecules into centrosymmetric dimers.

Benzyl and benzoyl benzoic acid inhibitors of bacterial RNA polymerase-sigma factor interaction.

Project description:Transcription is an essential biological process in bacteria requiring a core enzyme, RNA polymerase (RNAP). Bacterial RNAP is catalytically active but requires sigma (σ) factors for transcription of natural DNA templates. σ factor binds to RNAP to form a holoenzyme which specifically recognizes a promoter, melts the DNA duplex, and commences RNA synthesis. Inhibiting the binding of σ to RNAP is expected to inhibit bacterial transcription and growth. We previously identified a triaryl hit compound that mimics σ at its major binding site of RNAP, thereby inhibiting the RNAP holoenzyme formation. In this study, we modified this scaffold to provide a series of benzyl and benzoyl benzoic acid derivatives possessing improved antimicrobial activity. A representative compound demonstrated excellent activity against Staphylococcus epidermidis with minimum inhibitory concentrations reduced to 0.5 μg/mL, matching that of vancomycin. The molecular mechanism of inhibition was confirmed using biochemical and cellular assays. Low cytotoxicity and metabolic stability of compounds demonstrated the potential for further studies.

Project description:The reaction of lanthanum nitrate and 4-ethyl-benzoic acid (EBAH) in aqueous solution yielded the title polymer, [La(C(9)H(9)O(2))(3)(C(9)H(10)O(2))(H(2)O)](n). The asymmetric unit contains one La(III) atom, three 4-ethyl-benzoate (EBA) ligands, one neutral EBAH ligand and one coordinated water mol-ecule. Each La(III) ion is eight-coordinated by six O atoms from six bridging-bidentate EBA ligands, one O atom from a monodentate EBAH ligand and one water O atom in a distorted bicapped trigonal-prismatic geometry. The adjacent La(III) ions are linked by the carboxyl-ate groups of EBA ligands in a bridging-bidetate coordination mode, resulting in an infinite chain structure along the c axis. O-H⋯O hydrogen-bonding inter-actions involving the water mol-ecules, carboxyl-ate groups and carboxyl H atoms are formed within the one-dimensional polymer. One of the ethyl groups is disordered over two positions with occupancies of 0.717 (7) and 0.283 (7).
