ABSTRACT: The asymmetric unit of the anhydrous title compound, 2Na(+)·C(4)H(4)O(6) (2-), contains two sodium cations and one tartrate anion. Each sodium ion is six coordinate, with bonding to six O atoms from both the carboxyl-ate and hydroxyl groups of the anion. A three-dimensional coordination network is formed with sodium ions stacking in layers along the c-axis direction. This network is supported by additional O-H?O hydrogen bonds.

In the crystal, the hy-droxy H atom is equally disordered over two orientations, and links with an adjacent mol-ecule via an O-H?O hydrogen bond in both cases. Weak inter-molecular C-H?O hydrogen bonding is also a feature of the crystal structure.

The dihedral angle between the two benzene rings is 77.24?(6)°.

The two nonsolvent mol-ecules of the asymmetric unit are linked together by a weak C-H?O hydrogen bond. The ethyl acetate mol-ecule is present as a space filler and does not participate in the hydrogen-bonding network.

Fostamatinib Disodium.

Project description:The non-receptor tyrosine kinase Syk has a diverse range of biological functions, including a critical role in the intracellular signalling cascade for the surface immunoglobulin receptor on B lymphocytes, and the Fc receptor expressed on numerous immune effector cells. It is therefore seen as a potential therapeutic target in a variety of conditions, including autoimmune, allergic and malignant diseases. Fostamatinib disodium is the orally bioavailable prodrug of R406, a relatively selective small molecule inhibitor of Syk, that has accordingly shown activity in numerous cell types in vitro, and efficacy in a remarkable range of animal models in vivo, including rodent models of asthma, inflammatory arthritis, lupus, glomerulonephritis, diabetes and lymphoma. Success in these models has translated to phase II clinical trials in autoimmune thrombocytopenia, lymphoma and, most notably, rheumatoid arthritis, in which larger phase III trials are currently in progress. Whilst the diverse biological functions of Syk, coupled to the potential off-target effects of this kinase inhibitor are a source of possible toxicity, the available data thus far augurs well for future clinical use of Fostamatinib in a wide range of human diseases.

Project description:In the title salt, C23H26F3N2OS(+)·C4H5O6 (-) [systematic name: 1-(2-hy-droxy-eth-yl)-4-[3-(2-(tri-fluoro-meth-yl)thioxanthen-9-yl-idene)prop-yl]piperazin-1-ium 3-carb-oxy-2,3-di-hydroxy-pro-pion-ate], the monoprotonated piperazine ring in the cation adopts a chair conformation, while the thio-pyran ring of the thioxanthene group has a boat conformation. The dihedral angle between the mean planes of the two outer aromatic rings of the thioxanthene groups is 31.6?(2)°. In the crystal, the cations and anions are linked via O-H?O, N-H?O, O-H?N and C-H?O hydrogen bonds, forming chains propagating along [100]. In addition, R (2) 2(7), R (2) 2(11), R (2) 2(10) and R (2) 2(12) graph-set ring motifs involving the anions, and R (2) 2(9) graph-set ring motifs involving both the cations and anions are observed. The three F atoms of the tri-fluoro-methyl group are disordered over two sets of sites and the individual atoms were refined with occupancy ratios of 0.54?(6):0.46?(6), 0.72?(2):0.28?(2) and 0.67?(3):0.33?(3).

Project description:In the title compound (systematic name(R)-dimeth-yl{2-[1-phenyl-1-(pyridin-2-yl)eth-oxy]eth-yl}aza-nium (R,R)-3-carb-oxy-2,3-dihy-droxy-propano-ate), C(17)H(23)N(2)O(+)·C(4)H(5)O(6) (-), the doxylaminium cation is protonated at the N atom. The tartrate monoanions are linked by short, almost linear O-H?O hydrogen bonds into chains extended along [100]. These chains are inter-linked by anion-pyridine O-H?N hydrogen bonds into a two-dimensional grid structure. WeakC-H?O inter-actions also play a role in the crystal packing. An intra-molecular hy-droxy-carboxyl-ate O-H?O hydrogen bond influences the conformation of the anion: the hydrogen-bonded fragment is almost planar, the maximum deviation from the mean plane being 0.059?(14)?Å. In the cation, the aromatic rings are almost perpendicular [dihedral angle = 84.94?(8)°] and the conformation of the O-C-C-N chain is gauche(-), the dihedral angle is -76.6?(2)°. The absolute configuration was assigned on the basis of known chirality of the parent compound.

The conformation of the C-C-C-C backbone of the monotartrate anion is close to anti [torsion angle = 173.18 (17)°], which is supported by two intra-molecular O-H⋯O hydrogen bonds. In the crystal, the components are linked by N-H-O and O-H-O hydrogen bonds, generating (001) sheets.

Poly[[?(7)-l-cysteato(2-)]disodium].

Project description:The title compound {systematic name: poly[[?(7)-(2R)-2-amino-3-sulfonato-propano-ato]disodium]}, [Na(2)(C(3)H(5)NO(5)S)](n), was obtained through solvent-thermal reaction of l-cysteic acid and aqueous sodium hydroxide. The monomer consists of two Na(+) cations that are coordinated to the deprotonated amino acid. The latter acts as donor utilizing all available coordination sites, viz. the amino, the carboxyl-ate and the sulfonate residues, so producing a monomeric framework in which the two coordinated Na(+) ions have different coordination spheres and geometries. One of the Na(+) ions has an O(5) coordination sphere with a typical geometric arrangement, inter-mediate between trigonal-bipyramidal and square-pyramidal; all the O atoms from the amino acid (three from the sulfonate and two from the caboxylate residues) act as donors. The second Na(+) ion is tetracoordinated within an NO(3) coordination sphere. The Na(+) ion binds to the amino N atom, to one of the O atom of the carb-oxy-lic residue and to two O atoms of the sulfonate group in a distorted tetra-hedral arrangement. As the sulfonate O atoms bind to both Na(+) ions, a three-dimensional polymeric framework is obtained.
