Low genetic diversity in Polish populations of sibling ant species: Lasius niger (L.) and Lasius platythorax Seifert (Hymenoptera, Formicidae).

ABSTRACT: We present preliminary data on mitochondrial DNA diversity within and among populations of the ants Lasius niger and Lasius platythorax in Poland. Phylogenetic analysis based on the mitochondrial DNA markers: cytochrome c oxidase subunit I (cox1) and 16S ribosomal RNA (16S rRNA) confirms the species status of L. niger and L. platythorax. Intraspecific variability is low in both species, which might be a result of severe bottlenecks and rapid postglacial expansion into Central Europe.

Project description:Lasiustapinomoides sp. n. from Crete, Greece, is described and illustrated. It belongs to L.turcicus complex and is well characterized by very small body, extremely shallow metanotal groove and presence of suberect to erect setae on the apical part of scape. New records of Cretan members of the genus Lasius Fabricius, 1804 are provided, their checklist is updated, and the key to their determination is presented.

Morphology of the tentorium in the ant genus Lasius Fabricius (Hymenoptera: Formicidae).

Project description:The tentorium is the internal skeleton of the head capsule of insects. Several studies have shown that the structure of the tentorium is an important factor not only for the morphology and systematics but also for the phylogeny and evolution. In ants, however, only three studies have reported tentorial morphology so far. We reveal the fundamental structure of the tentorium of the genus Lasius (Hymenoptera, Formicidae) and its minor variation among six species of the genus. Based on the results, we give new terminologies of the organ, presenting a schematic diagram of the tentorium. We clarify muscle attachment to the tentorium by constructing a three-dimension image of the tentorium and muscles. We then verify the attachment areas of the antennal muscles and maxillary adductor muscles on the tentorium. The results show that the muscular attachment areas are broader than previously thought. Our study indicates that the key to understanding the evolution of the tentorium is its functional morphology, in relation to the attachments of the muscles originating from the tentorium within the head capsule. This is the first report of the three-dimensional images of the ant tentorium and its attached muscles. The intra- and inter-specific variations of the tentorium is also reported for the first time.

Proteinaceous Venom Expression of the Yellow Meadow Ant, Lasius flavus (Hymenoptera: Formicidae).

Project description:Ants are one of the important groups of venomous animals with about 14,000 described species. Studies so far focused on the discovery of venom proteins are only available for limited stinging ants, and the proteinaceous compositions of the stingless ants are completely unknown. Here, we used the transcriptomic approach to identify venom components from the yellow meadow ant, Lasius flavus, a stingless ant. The transcriptomic analysis yielded an extraordinary simplicity of the venom expression profile, with 17 venom proteins, such as phospholipase B, odorant binding protein, and apolipoprotein D. Ten of them were discovered as novel toxins for future functional investigations. Quantitative real time PCR analysis revealed that genes encoding the identified venom proteins display exclusively or highly expression profiles in venom glands, validating them as venom compositions. Our findings contribute to the understanding of the evolutional diversity of toxins between stinging and stingless ants.

Complete mitochondrial genome of the jet ant Lasius spathepus Wheeler, W.M., 1910 (Formicidae; Hymenoptera).

Project description:The genus Lasius is a conspicuous and popular genus of ants found in the Holarctic regions. We have completed the mitochondrial genome of Lasius spathepus as the first mitochondrial genome of Lasius. The mitochondrial genome is 18,951 bp long, which is the sixth longest ant mitochondrial genome known to science. It contains 13 protein-coding genes, 2 ribosomal RNAs, 22 transfer RNAs, and a control region in a gene order shared with other species of subfamily Formicinae. The control region is 2,147 bp long, longest of all ants. Phylogenetic analysis shows L. spathepus groups with Nylanderia flavipes of the same tribe Lasiini.

New and little-known ant species (Hymenoptera, Formicidae) from Bulgaria.

Project description:BackgroundMany faunistic studies on the myrmecofauna of Bulgaria have been carried out and about 180 ant species (Hymenoptera, Formicidae) from 43 genera and six subfamilies have been discovered as a result. Although the Bulgarian ant fauna is considered to be relatively well studied, the finding of unrecorded species continues, especially amongst the rare social parasites and the species with a more southern distribution in the Balkans.New informationThe current study presents data on 11 ant species recorded for the first time in Bulgaria (Messorhellenius Agosti & Collingwood, 1987, M.mcarthuri Steiner et al., 2018, Crematogasterionia Forel, 1911, Monomoriummonomorium Bolton, 1987, Temnothoraxaeolius (Forel, 1911), T.cf.exilis(formdarii Forel, 1911), T.finzii (Menozzi, 1925), T.rogeri Emery, 1869, Tetramoriumcf.punicum, Plagiolepisxene Stärcke, 1936 and Lasiusreginae Faber, 1967), as well as new locality data on 15 rarely found species. Some of the species, such as Hypoponeraeduardi (Forel, 1894), Strumigenysargiola (Emery, 1869), Temnothoraxgraecus (Forel, 1911), Tetramoriumdiomedeum Emery, 1908, Camponotusionius Emery, 1920 and C.tergestinus Müller, 1921, have been known so far only from a single locality in Bulgaria.The dataset of all records presented in this work was published separately through Global Biodiversity Information Facility (GBIF, https://doi.org/10.15468/mngbzp).

Varying impact of neonicotinoid insecticide and acute bee paralysis virus across castes and colonies of black garden ants, Lasius niger (Hymenoptera: Formicidae).

Project description:Pesticides and pathogens are known drivers of declines in global entomofauna. However, interactions between pesticides and viruses, which could range from antagonistic, over additive to synergistic, are poorly understood in ants. Here, we show that in ants the impact of single and combined pesticide and virus stressors can vary across castes and at the colony level. A fully-crossed laboratory assay was used to evaluate interactions between a sublethal dose of the neonicotinoid thiamethoxam and Acute bee paralysis virus (ABPV) in black garden ants, Lasius niger. After monitoring colonies over 64 weeks, body mass, neonicotinoid residues and virus titres of workers and queens, as well as worker behavioural activity were measured. ABPV, but not thiamethoxam, reduced activity of workers. Neonicotinoid exposure resulted in reduced body mass of workers, but not of queens. Further, thiamethoxam facilitated ABPV infections in queens, but not in workers. Overall, virus exposure did not compromise detoxification and body mass, but one colony showed high virus titres and worker mortality. Although the data suggest additive effects at the level of individuals and castes, co-exposure with both stressors elicited antagonistic effects on colony size. Our results create demand for long-term holistic risk assessment of individual stressors and their interactions to protect biodiversity.

Genetic modification of the ant Lasius niger using CRISPR-Cas9 technology.

Project description:CRISPR-Cas9 has become one of the most prominent gene editing tools available and it has been utilized in various organisms from bacteria to fungi, plants, and animals. In this study, we developed a CRISPR-Cas9 protocol for the black garden ant Lasius niger, a common and easily available study species for lab and field experiments. To create indel mutations using CRISPR-Cas9 in L. niger, we targeted three different locations in a well-studied eye pigmentation gene cinnabar, generating several mutations that disrupt the ommochrome biosynthesis pathway and result in the lack of the pigment and therefore, abnormal eye coloration in adult workers. We also developed a protocol to collect L. niger eggs, inject them with CRISPR-Cas9 construct, and rear the eggs into mature adult workers with the assistance of nursing workers. We demonstrated for the first time in L. niger that CRISPR-Cas9 is an excellent tool to create targeted mutations for this species. Our protocol can be referred to when developing similar studies for other species of ants and eusocial insects.

Ant Lasius niger joining one-way trails go against the flow.

Project description:Social insects, such as ants, use various pheromones as their social signal. In addition, they use the presence of other ants for decision-making. In this study, we attempted to evaluate if individual decision-making is influenced by the complementary use of pheromones and presence of other ants. Ants were induced to form a one-way flow system. We found that when ants entered such a system at a right angle, they tended to move in the opposite direction of the one-way flow system. Interestingly, the target ants moved randomly in the experiments in which no ant and/or no pheromone trails were present. We also developed simulation algorithms and found that artificial ant foragers could reach a certain goal more often if they adopted the reverse run (similar mechanism found in ant experiments) over the forward run (moving in the same direction as their nestmates).

Antimicrobial Activity of Microorganisms Isolated from Ant Nests of Lasius niger.

Project description:In this study, the microbial communities of two nests of black garden ants (Lasius niger) in the hollows of stem branches of old apple trees were found to have similar species compositions: each community contained representatives of three species from the Bacillaceae family and one species of actinomycetes from the genus Streptomyces. In total, four types of bacilli and two actinomycetes were isolated. Actinomycetes were identified as Streptomyces antibioticus-like and Streptomyces sp. None of the bacilli had antibiotic activity, whereas both streptomycetes produced antibiotics that inhibited the growth of Gram-positive bacteria in vitro, including isolates from their community. Antibiotic compounds of S. antibioticus-like strain INA 01148 (Institute of New Antibiotics) were identified as actinomycin D and its closest homologue, actinomycin A. Actinomycins presumably change the microbial community of the ant nest substrate as they act against Gram-positive bacteria and against fungi and Gram-negative bacteria. The antibiotic activity of the isolated Streptomyces sp. INA 01156 is of interest, since the substances produced by this strain inhibit the growth of drug-resistant bacteria, including methicillin-resistant Staphylococcus aureus INA 00761 (MRSA) and vancomycin-resistant strain Leuconostoc mesenteroides VKPM B-4177 (VR) (VKPM-National Collection of Industrial Microorganisms (Russian acronym)).

Evolution of morphological crypsis in the Tetramorium caespitum ant species complex (Hymenoptera: Formicidae).

Project description:Cryptic species are morphologically very similar to each other. To what extent stasis or convergence causes crypsis and whether ecology influences the evolution of crypsis has remained unclear. The Tetramorium caespitum complex is one of the most intricate examples of cryptic species in ants. Here, we test three hypotheses concerning the evolution of its crypsis: H1: The complex is monophyletic. H2: Morphology resulted from evolutionary stasis. H3: Ecology and morphology evolved concertedly. We confirmed (H1) monophyly of the complex; (H2) a positive relation between morphological and phylogenetic distances, which indicates a very slow loss of similarity over time and thus stasis; and (H3) a positive relation between only one morphological character and a proxy of the ecological niche, which indicates concerted evolution of these two characters, as well as a negative relation between p-values of correct species identification and altitude, which suggests that species occurring in higher altitudes are more cryptic. Our data suggest that species-specific morphological adaptations to the ecological niche are exceptions in the complex, and we consider the worker morphology in this complex as an adaptive solution for various environments.
