Project description:Significant attention is paid to the design of magnetoplasmonic nanohybrids, which exploit synergistic properties for biomedical applications. Here, a facile method was employed to prepare plasmonic magnetic Au-MnO heterostructured hybrid nanoparticles for imaging-guided photothermal therapy of cancers in vitro, with the view to reducing the serious drawbacks of chemotherapy and gadolinium-based contrast agents. The biocompatibility of the prepared Au-MnO nanocomposites was further enhanced by Food and Drug Administration (FDA)-approved triblock copolymers Pluronic® F-127 and chitosan oligosaccharide (COS), with complementary support to enhance the absorption in the near-infrared (NIR) region. In addition, synthesized COS-PF127@Au-MnO nanocomposites exhibited promising contrast enhancement in T1 MR imaging with a good r1 relaxivity value (1.2 mM-1 s-1), demonstrating a capable substitute to Gd-based toxic contrast agents. In addition, prepared COS-PF127@Au-MnO hybrid nanoparticles (HNPs) produced sufficient heat (62 °C at 200 μg/mL) to ablate cancerous cells upon 808 nm laser irradiation, inducing cell toxicity, and apoptosis. The promising diagnostic and photothermal therapeutic performance demonstrated the appropriateness of the COS-PF127@Au-MnO HNPs as a potential theranostic agent.

Project description:Magnetically recoverable noble metal nanoparticles are promising catalysts for chemical reactions. However, the chemical synthesis of these nanocatalysts generally causes environmental concern due to usage of toxic chemicals under extreme conditions. Here, Pd/Fe3O4, Au/Fe3O4 and PdAu/Fe3O4 nanocomposites are biosynthesized under ambient and physiological conditions by Shewanella oneidensis MR-1. Microbial cells firstly transform akaganeite into magnetite, which then serves as support for the further synthesis of Pd, Au and PdAu nanoparticles from respective precursor salts. Surface-bound cellular components and exopolysaccharides not only function as shape-directing agent to convert some Fe3O4 nanoparticles to nanorods, but also participate in the formation of PdAu alloy nanoparticles on magnetite. All these three kinds of magnetic nanocomposites can catalyze the reduction of 4-nitrophenol and some other nitroaromatic compounds by NaBH4. PdAu/Fe3O4 demonstrates higher catalytic activity than Pd/Fe3O4 and Au/Fe3O4. Moreover, the magnetic nanocomposites can be easily recovered through magnetic decantation after catalysis reaction. PdAu/Fe3O4 can be reused in at least eight successive cycles of 4-nitrophenol reduction. The biosynthesis approach presented here does not require harmful agents or rigorous conditions and thus provides facile and environmentally benign choice for the preparation of magnetic noble metal nanocatalysts.

Project description:BACKGROUND:Identifying the precise location of cells and their migration dynamics is of utmost importance for achieving the therapeutic potential of cells after implantation into a host. Magnetic resonance imaging is a suitable, non-invasive technique for cell monitoring when used in combination with contrast agents. RESULTS:This work shows that nanowires with an iron core and an iron oxide shell are excellent materials for this application, due to their customizable magnetic properties and biocompatibility. The longitudinal and transverse magnetic relaxivities of the core-shell nanowires were evaluated at 1.5 T, revealing a high performance as T2 contrast agents. Different levels of oxidation and various surface coatings were tested at 7 T. Their effects on the T2 contrast were reflected in the tailored transverse relaxivities. Finally, the detection of nanowire-labeled breast cancer cells was demonstrated in T2-weighted images of cells implanted in both, in vitro in tissue-mimicking phantoms and in vivo in mouse brain. Labeling the cells with a nanowire concentration of 0.8 ?g of Fe/mL allowed the detection of 25 cells/µL in vitro, diminishing the possibility of side effects. This performance enabled an efficient labelling for high-resolution cell detection after in vivo implantation (~?10 nanowire-labeled cells) over a minimum of 40 days. CONCLUSIONS:Iron-iron oxide core-shell nanowires enabled the efficient and longitudinal cellular detection through magnetic resonance imaging acting as T2 contrast agents. Combined with the possibility of magnetic guidance as well as triggering of cellular responses, for instance by the recently discovered strong photothermal response, opens the door to new horizons in cell therapy and make iron-iron oxide core-shell nanowires a promising theranostic platform.

Project description:The ternary nanocomposites Fe3O4/Ag/polyoxometalates (Fe3O4/Ag/POMs) with core-shell-core nanostructure were synthesized by coating [Cu(C6H6N2O)2(H2O)]H2[Cu(C6H6N2O)2(P2Mo5O23)]·4H2O polyoxometalates on the surface of Fe3O4/Ag (core-shell) nanoparticles. The transmission electron microscopy/high resolution transmission electron microscopy (HR-TEM) and X-ray powder diffraction (XRD) analyses show that the Fe3O4/Ag/POMs ternary nanocomposites reveal a core-shell-core nanostructure, good dispersibility, and high crystallinity. The vibrating sample magnetometer (VSM) and physical property measurement system (PPMS) demonstrated the good magnetic properties and superparamagnetic behavior of the nanocomposites at 300 K. The UV-vis spectroscopy displayed the broadband absorption of the Fe3O4/Ag/POMs with the maximum surface plasmon resonance of Ag nanostructure around 420 nm. The dye removal capacity of Fe3O4/Ag/POMs was investigated using methylene blue (MB) as a probe. Through adsorption and photocatalysis, the nanocomposites could quickly remove MB with a removal efficiency of 98.7% under the irradiation of visible light at room temperature. The removal efficiency was still as high as 97.5% even after six runs by magnetic separation of photocatalytic adsorbents after processing, indicating the reusability and high stability of the nanocomposites. These Fe3O4/Ag/POMs photocatalytic adsorbents with magnetic properties will hopefully become a functional material for wastewater treatment in the future.

Project description:Nanoparticles with SiO2 coating were synthesized to have a cubic iron core. These were found to have saturation magnetization very close to the highest possible value of any iron-containing nanoparticles and the bulk iron saturation magnetization. The in vitro toxicology studies show that they are highly biocompatible and possess better MRI contrast agent potential than iron oxide NPs.

Project description:Chemical exchange saturation transfer (CEST) MRI is a versatile molecular imaging approach that holds great promise for clinical translation. A number of compounds have been identified as suitable for performing CEST MRI, including paramagnetic CEST (paraCEST) agents and diamagnetic CEST (diaCEST) agents. DiaCEST agents are very attractive because of their excellent biocompatibility and potential for biodegradation, such as glucose, glycogen, glutamate, creatine, nucleic acids, et al. However, the sensitivity of most diaCEST agents is limited because of small chemical shifts (1.0-4.0 ppm) from water. To expand the catalog of diaCEST agents with larger chemical shifts, herein, we have systematically investigated the CEST properties of acyl hydrazides with different substitutions, including aromatic and aliphatic substituents. We have tuned the labile proton chemical shifts from 2.8-5.0 ppm from water while exchange rates varied from ~680 to 2340 s-1 at pH 7.2, which allows strong CEST contrast on scanners down to B0 = 3 T. One acyl hydrazide, adipic acid dihydrazide (ADH), was tested on a mouse model of breast cancer and showed nice contrast in the tumor region. We also prepared a derivative, acyl hydrazone, which showed the furthest shifted labile proton (6.4 ppm from water) and excellent contrast properties. Overall, our study expands the catalog of diaCEST agents and their application in cancer diagnosis.

Project description:Murine RAW 264.7 cells were exposed to spheroidal core-shell Fe(3)O(4)@Au nanoparticles (SCS-NPs, ca. 34 nm) or nanostars (NSTs, ca. 100 nm) in the presence of bovine serum albumin, with variable effects observed after macrophagocytosis. Uptake of SCS-NPs caused macrophages to adopt a rounded, amoeboid form, accompanied by an increase in surface detachment. In contrast, the uptake of multibranched NSTs did not induce gross changes in macrophage shape or adhesion, but correlated instead with cell enlargement and signatures of macrophage activation such as TNF-α and ROS. MTT assays indicate a low cytotoxic response to either SCS-NPs or NSTs despite differences in macrophage behavior. These observations show that differences in NP size and shape are sufficient to produce diverse responses in macrophages following uptake.

Project description:While carbon dots (C-dots) have been extensively investigated pertaining to their fluorescent, phosphorescent, electrochemiluminescent, optoelectronic, and catalytic features, their inherent chemical exchange saturation transfer magnetic resonance imaging (CEST MRI) properties are unknown. By virtue of their hydrophilicity and abundant exchangeable protons of hydroxyl, amine, and amide anchored on the surface, we report here that C-dots can be adapted as effective diamagnetic CEST (diaCEST) MRI contrast agents. As a proof-of-concept demonstration, human glioma cells were labeled with liposomes with or without encapsulated C-dots and implanted in mouse brain. In vivo CEST MRI was able to clearly differentiate labeled cells from non-labeled cells. The present findings may encourage new applications of C-dots for in vivo imaging in deep tissues, which is currently not possible using conventional fluorescent (near-infrared) C-dots.

Project description:Iron oxides are potential electrode materials for lithium-ion batteries because of their high theoretical capacities, low cost, rich resources, and their non-polluting properties. However, iron oxides demonstrate large volume expansion during the lithium intercalation process, resulting in the electrode material being crushed, which always results in poor cycle performance. In this paper, to solve the above problem, iron oxide/carbon nanocomposites with a hollow core-shell structure were designed. Firstly, an Fe2O3@polydopamine nanocomposite was prepared using an Fe2O3 nanocube and dopamine hydrochloride as precursors. Secondly, an Fe3O4@N-doped C composite was obtained by means of further carbonization treatment. Finally, Fe3O4@void@N-Doped C-x composites with core-shell structures with different void sizes were obtained by means of Fe3O4 etching. The effect of the etching time on the void size was studied. The electrochemical properties of the composites when used as lithium-ion battery materials were studied in more detail. The results showed that the sample that was obtained via etching for 5 h using 2 mol L-1 HCl solution at 30 °C demonstrated better electrochemical performance. The discharge capacity of the Fe3O4@void@N-Doped C-5 was able to reach up to 1222 mA g h-1 under 200 mA g-1 after 100 cycles.

Project description:Amphiphilic triblock copolymers containing Fe(III) -catecholate complexes formulated as spherical- or cylindrical-shaped micellar nanoparticles (SMN and CMN, respectively) are described as new T1-weighted agents with high relaxivity, low cytotoxicity, and long-term stability in biological fluids. Relaxivities of both SMN and CMN exceed those of established gadolinium chelates across a wide range of magnetic field strengths. Interestingly, shape-dependent behavior is observed in terms of the particles' interactions with HeLa cells, with CMN exhibiting enhanced uptake and contrast via magnetic resonance imaging (MRI) compared with SMN. These results suggest that control over soft nanoparticle shape will provide an avenue for optimization of particle-based contrast agents as biodiagnostics. The polycatechol nanoparticles are proposed as suitable for preclinical investigations into their viability as gadolinium-free, safe, and effective imaging agents for MRI contrast enhancement.
