Five new species of Triotemnus (Coleoptera, Curculionidae, Scolytinae) from Morocco and Yemen.

ABSTRACT: Fivenew species of the genus Triotemnus from Morocco and Yemen are described. Triotemnus is a new genus of Scolytinae for the Yemen region. External morphology of the new species and all morphologically related species of the genus were studied. While the new species from Morocco are morphologically similar to the known species from the corresponding region, all three newly described species from Yemen, mainly two of them living in Socotra, are morphologically very different from all other known species of the genus. Geographical distribution and the probability of endemicity are discussed.

Rapid Molecular Identification of Scolytinae (Coleoptera: Curculionidae).

Project description:Routine identification of bark and ambrosia beetles is done using morphology. For people lacking the necessary taxonomic knowledge, proper identification of a novel specimen can be challenging and time consuming. This study compares the usefulness of four genetic markers (28S, EF-1a, ITS2, and COI) and five primer pairs (D2F1/D3R2, eflafor1/eflarev1, ets149/efa754, ITS2F/ITS2R, and LCO1490/HCO2198) to identify Scolytinae beetles, and outlines a molecular identification strategy, with results possible in two days. Markers COI and EF-1a were selected based on the ability of the respective primers to amplify DNA from multiple genera (Coptoborus, Xyleborus, Hypothenemus, Theoborus, and Araptus) and the ability of the resulting sequences to provide accurate and unambiguous matches in GenBank. BLASTn analysis of EF-1a sequences (both primer pairs) correctly identified four out of the five genera and COI sequences identified at least one sample of every genus tested and was the only primer pair to correctly identify Araptus specimens. Further, 28S sequences successfully identified Coptoborus, Xyleborus, and Theoborus but not Hypothenemus or Araptus. The low number of EF-1a (1), 28S (7), and ITS2 (0) sequences from Araptus individuals present in GenBank compared with COI (137) is likely the reason that only the latter marker was capable of identifying members of this genus. ITS2 sequences were insufficient to identify any of the samples tested. This study also determined the minimum quantity of DNA that could be used for molecular identification. Primers D2F1 and D3R2, which had the highest rate of amplification in all genera tested, could yield an informative sequence with as little as 0.00048 ng of DNA, however, at least 0.0024 ng was needed for reliable amplification.

Project description:Following the recent reclassification of the Palaeotropic xyleborine genera (Hulcr and Cognato in press), additional species are transferred to correct genera or synonymized based on analysis of their morphological characters. Project description:Ten species of ambrosia beetles of the tribe Xyleborini, Amasabeesoni (Eggers, 1930), Amasaopalescens (Schedl, 1937), Amasacylindrotomica (Schedl, 1939), Arixyleborushirsutulus Schedl, 1969, Beaveriumlatus (Eggers, 1923), Cnestusprotensus (Eggers, 1930), Coptodryasquadricostata (Schedl, 1942), Cryptoxyleborusconfusus Browne, 1950, Cryptoxyleboruspercuneolus (Schedl, 1951) and Cyclorhipidionvigilans (Schedl, 1939), are recorded here for the first time in Thailand. Diagnostic characters, illustrations, distribution and biological data are provided for each species. Xylosandrusramulorum (Schedl, 1957), stat. res. is removed from synonymy with Amasacylindrotomica and reinstated as a valid species.

Revision of the genus Phloeoditica Schedl - with description of two new genera and two new species in Phloeosinini (Coleoptera, Curculionidae, Scolytinae).

Project description:The genus Phloeoditica Schedl currently includes four species from Southeast Asia. These species vary substantially in important morphological characters and indicate the existence of multiple genera for these species. A revision based on morphological and in part molecular data resulted in the transfer of Phloeoditica setosa to Pseudoxylechinus the erection of a new genus Asiophilus for Phloeoditica phloeosinoides and a new species Asiophilus macropunctatus from Vietnam. Another new genus with affinities to Phloeoditica is described based on the new species Microditica uniseriata from Thailand. The new genera are included in a revised key to the tribe Phloeosinini.

Resurrection of Dryotomicus Wood and description of two new species from the Amazon River Basin (Coleoptera, Curculionidae, Scolytinae, Phloeotribini).

Project description:A cladistic analysis based on 20 morphological characters was conducted for 11 species representing two valid and two synonymized Phloeotribini genera. One hundred-eighty most-parsimonious trees were recovered and the Dryotomicus Wood species were monophyletic in a mostly unresolved strict-consensus tree. The unusual antennal morphology, with the length of the first two funicular segments equal to the last three segments and a scape which is twice the length of the funicle, distinguish Dryotomicus from the other Phloeotribini genera. Hence this genus is resurrected because of monophyly and diagnostic characters. Dryotomicus oenophilissp. n. and Dryotomicus woodrexsp. n. are described from Guyana and Peru, respectively. In the male specimen of Dryotomicus oenophilis, the frons has one median and two large lateral carinae and in the male specimen of Dryotomicus woodrex, the frons has three smaller median tubercles arranged transversely. Phloeotribus puberulus Chapuis and Phloeotribus tuberculatus (Eggers) were monophyletic with the new Dryotomicus species and thus are transferred to this genus. Keys to the Phloeotribini genera and Dryotomicus species are given.

Core Mycobiome and Their Ecological Relevance in the Gut of Five Ips Bark Beetles (Coleoptera: Curculionidae: Scolytinae).

Project description:Bark beetles are destructive forest pests considering their remarkable contribution to forest depletion. Their association with fungi is useful against the challenges of survival on the noxious and nutritionally limited substrate, i.e., conifer tissues. Fungal symbionts help the beetles in nutrient acquisition and detoxification of toxic tree secondary metabolites. Although gut is the prime location for food digestion and detoxification, limited information is available on gut-mycobiome of bark beetles. The present study screened the gut-mycobiont from six bark beetles (five Ips and one non-Ips) from Scolytinae subfamily using high-throughput sequencing and explored their putative role in symbiosis with the host insect. Results revealed the predominance of four fungal classes- Sordariomycetes, Saccharomycetes, Eurothiomycetes, and Dothidomycetes in all bark beetles. Apart from these, Agaricomycetes, Leothiomycetes, Incertae sedis Basidiomycota, Tremellomycetes, Lecanoromycetes, and Microbotryomycetes were also documented in different beetles. Five Ips bark beetles share a consortium of core fungal communities in their gut tissues consisting of 47 operational taxonomic units (OTUs) belonging to 19 fungal genera. The majority of these core fungal genera belong to the phylum Ascomycota. LEfSe analysis revealed a set of species-specific fungal biomarkers in bark beetles. The present study identified the gut mycobiont assemblage in bark beetles and their putative ecological relevance. An enriched understanding of bark beetle-fungal symbiosis is not only filling the existing knowledge gap in the field but may also unleash an unforeseen potential for future bark beetle management.

A taxonomic monograph of Nearctic Scolytus Geoffroy (Coleoptera, Curculionidae, Scolytinae).

Project description:The Nearctic bark beetle genus Scolytus Geoffroy was revised based in part on a molecular and morphological phylogeny. Monophyly of the native species was tested using mitochondrial (COI) and nuclear (28S, CAD, ArgK) genes and 43 morphological characters in parsimony and Bayesian phylogenetic analyses. Parsimony analyses of molecular and combined datasets provided mixed results while Bayesian analysis recovered most nodes with posterior probabilities >90%. Native hardwood- and conifer-feeding Scolytus species were recovered as paraphyletic. Native Nearctic species were recovered as paraphyletic with hardwood-feeding species sister to Palearctic hardwood-feeding species rather than to native conifer-feeding species. The Nearctic conifer-feeding species were monophyletic. Twenty-five species were recognized. Four new synonyms were discovered: Scolytuspraeceps LeConte, 1868 (= Scolytusabietis Blackman, 1934; = Scolytusopacus Blackman, 1934), Scolytusreflexus Blackman, 1934 (= Scolytusvirgatus Bright, 1972; = Scolytuswickhami Blackman, 1934). Two species were reinstated: Scolytusfiskei Blackman, 1934 and Scolytussilvaticus Bright, 1972. A diagnosis, description, distribution, host records and images were provided for each species and a key is presented to all species.
