Trichome-derived O-acyl sugars are a first meal for caterpillars that tags them for predation.

ABSTRACT: Plant glandular trichomes exude secondary metabolites with defensive functions, but these epidermal protuberances are surprisingly the first meal of Lepidopteran herbivores on Nicotiana attenuata. O-acyl sugars, the most abundant metabolite of glandular trichomes, impart a distinct volatile profile to the body and frass of larvae that feed on them. The headspace composition of Manduca sexta larvae is dominated by the branched chain aliphatic acids hydrolyzed from ingested O-acyl sugars, which waxes and wanes rapidly with trichome ingestion. In native habitats a ground-hunting predator, the omnivorous ant Pogonomyrmex rugosus, but not the big-eyed bug Geocoris spp., use these volatile aliphatic acids to locate their prey.

Parasitised caterpillars suffer reduced predation: potential implications for intra-guild predation.

Project description:Intra-guild predation (IGP) is an important phenomenon structuring ecological communities and affects the success of biological control. Here we show that parasitism by the koinobiont wasp Cotesia vestalis is associated with behavioural changes in its larval host (diamondback moth, Plutella xylostella) that reduce risk of IGP. Compared with unparasitised caterpillars, parasitised P. xylostella moved less frequently to new feeding patches on plants and were less likely to fall from the plant. Wolf spiders killed significantly fewer parasitised larvae. Reflecting their reduced movement and capacity to select plant tissue of optimal quality, parasitised caterpillars fed at a lower rate and exhibited delayed development suggesting a trade-off between IGP avoidance and nutrient intake by the host. This change in behaviour to reduce risk may cascade to the first trophic level and help explain the stability of IGP systems.

Out in the open: behavior's effect on predation risk and thermoregulation by aposematic caterpillars.

Project description:Warning coloration should be under strong stabilizing selection but often displays considerable intraspecific variation. Opposing selection on color by predators and temperature is one potential explanation for this seeming paradox. Despite the importance of behavior for both predator avoidance and thermoregulation, its role in mediating selection by predators and temperature on warning coloration has received little attention. Wood tiger moth caterpillars, Arctia plantaginis, have aposematic coloration, an orange patch on the black body. The size of the orange patch varies considerably: individuals with larger patches are safer from predators, but having a small patch is beneficial in cool environments. We investigated microhabitat preference by these caterpillars and how it interacted with their coloration. We expected caterpillar behavior to reflect a balance between spending time exposed to maximize basking and spending time concealed to avoid detection by predators. Instead, we found that caterpillars preferred exposed locations regardless of their coloration. Whether caterpillars were exposed or concealed had a strong effect on both temperature and predation risk, but caterpillars in exposed locations were both much warmer and less likely to be attacked by a bird predator (great tits, Parus major). This shared optimum may explain why we observed so little variation in caterpillar behavior and demonstrates the important effects of behavior on multiple functions of coloration.

Predation by avian insectivores on caterpillars is linked to leaf damage on oak (Quercus robur).

Project description:Birds that are foraging in tree canopies can cause a substantial decrease in arthropod numbers. Trees may benefit from avian insectivores attacking insect herbivores. In a field study, we tested whether the intensity of bird predation on caterpillars is linked quantitatively to leaf damage caused by insect herbivores, a hypothesized relationship that previously was poorly investigated. Artificial caterpillars were placed in the lower part of oak trees (Quercus robur) in urban and suburban sites across the city of Gothenburg, Sweden. Two days later, we recorded the survival: the pooled predation rate was 11.5% (5.7% day-1). Mean predation rate per tree was 10.4%. Mean leaf damage, i.e. leaf area eaten by insect herbivores, per tree was 5.7% but there was large variation between trees. We found a significant negative relationship between survival probability of caterpillars and leaf damage in an analysis using a mixed model logistic regression. This suggests that caterpillars are at high risk of bird attacks in trees with a high degree of leaf damage and avian insectivores may increase the foraging effort in the foliage of such oak trees. Our findings concerning the quantitative relationship between the predator-prey interactions and plant damage suggested tentatively that the survival probability of caterpillars decreases rapidly at 15-20% leaf damage in lower part of oak canopies. Furthermore, our findings add credence to the idea of using artificial caterpillars as a means to obtain standardized comparisons of predation rates in various habitats.

Identification of a BAHD acetyltransferase that produces protective acyl sugars in tomato trichomes.

Project description:Glandular secreting trichomes on the surface of tomato plants and many of its relatives in the Solanaceae produce a mixture of O-acyl sugars that contribute to insect resistance. The majority of acyl sucroses produced by the cultivated tomato (Solanum lycopersicum) contain three or four short chain aliphatic acyl esters, and tetra-acyl sucroses have an acetyl group as one of the acyl chains. We previously reported overlapping S. lycopersicum × Solanum pennellii introgression lines (ILs) that fail to accumulate high levels of acetylated tetra-acyl sucroses. A survey of the annotated genes in this region of cultivated tomato chromosome 1 revealed three candidate acyltransferases that were tested for function using virus-induced gene silencing. A member of the BAHD family of acyltransferases (Solyc01g105580, SlAT2) was shown to encode an acetyl-CoA-dependent acyltransferase enzyme capable of acyl sucrose acetylation in vitro. RNAi suppression of SlAT2 in transgenic S. lycopersicum cv. M82 resulted in reduced acyl sugar acetylation, whereas expression of the functional S. lycopersicum allele of SlAT2 in the triacyl sucrose producing IL1-3 restored the ability of the IL to synthesize acetylated tetra-acyl sugars. Transgenic plants with the SlAT2 promoter driving GFP expression showed fluorescence in tips cells of long, slender trichomes that is consistent with acyl sugar acetylation occurring in these cells.

Unifying Scheme for the Biosynthesis of Acyl-Branched Sugars: Extended Substrate Scope of Thiamine-Dependent Enzymes.

Project description:Thiamine diphosphate (ThDP) dependent enzymes are useful catalysts for asymmetric C-C bond formation through benzoin-type condensation reactions that result in α-hydroxy ketones. A wide range of aldehydes and ketones can be used as acceptor substrates; however, the donor substrate range is mostly limited to achiral α-keto acids and simple aldehydes. By using a unifying retro-biosynthetic approach towards acyl-branched sugars, we identified a subclass of (myco)bacterial ThDP-dependent enzymes with a greatly extended donor substrate range, namely functionalized chiral α-keto acids with a chain length from C4 to C8 . Highly enantioenriched acyloin products were obtained in good to high yields and several reactions were performed on a preparative scale. The newly introduced functionalized α-keto acids, accessible by known aldolase-catalyzed transformations, substantially broaden the donor substrate range of ThDP-dependent enzymes, thus enabling a more general use of these already valuable catalysts.

Project description:ObjectiveTo examine if first-year roommates made similar meal plan decisions.MethodsResidence information for 1186 first-year students (N = 593 roommate pairs) and 559 floormates was obtained for the 2015-2016 academic year. Linear generalized estimating equations (GEEs) were used to examine if the number of meals students used over the semester was higher if their roommate used their meal plan more frequently. A logistic GEE examined joint meal plan usage between students and roommates for each month of the semester. To determine if residence, rather than roommate, explained the results, a simulation was conducted by randomly assigning the floormates to a same-sex roommate.ResultsThe number of meals students used in spring was higher if the students' roommate had used more meals in spring, even after controlling for the number of meals students used in fall (Female: β = 0.07, 99% CI = 0.00, 0.13; Male: β = 0.10, 99% CI = 0.02, 0.18). Students were more likely to use a meal with their roommate if they were on the same meal plan (Female: OR = 1.61, 99% CI = 1.27, 2.04; Male: OR = 1.57, 99% CI = 1.09, 2.25), and less likely after the first month of being roommates (Female: OR = 0.57-0.25; Male: OR = 0.50-0.22; p < 0.001). The simulation analysis indicated these findings were not due to shared residence.DiscussionStudents' meal plan choices were associated with their roommates' meal plan choices. Roommates' joint meal plan usage was highest at the start of the year. Strategic roommate pairings may result in students using their meal plan more. Further research should determine the extent of roommate influence on students' diet.

Radicalization and Radical Catalysis of Biomass Sugars: Insights from First-principles Studies.

Project description:Ab initio and density functional calculations are conducted to investigate the radicalization processes and radical catalysis of biomass sugars. Structural alterations due to radicalization generally focus on the radicalized sites, and radicalization affects H-bonds in D-fructofuranose more than in D-glucopyranose, potentially with outcome of new H-bonds. Performances of different functionals and basis sets are evaluated for all radicalization processes, and enthalpy changes and Gibbs free energies for these processes are presented with high accuracy, which can be referenced for subsequent experimental and theoretical studies. It shows that radicalization can be utilized for direct transformation of biomass sugars, and for each sugar, C rather than O sites are always preferred for radicalization, thus suggesting the possibility to activate C-H bonds of biomass sugars. Radical catalysis is further combined with Brønsted acids, and it clearly states that functionalization fundamentally regulates the catalytic effects of biomass sugars. In presence of explicit water molecules, functionalization significantly affects the activation barriers and reaction energies of protonation rather than dehydration steps. Tertiary butyl and phenyl groups with large steric hindrances or hydroxyl and amino groups resulting in high stabilities for protonation products drive the protonation steps to occur facilely at ambient conditions.

Expression and characterization of a protease-resistant β-d-fructofuranosidase BbFFase9 gene suitable for preparing invert sugars from soybean meal.

Project description:A novel gene (BbFFase9), with an ORF of 1557 bp that encodes β-d-fructofuranosidase from Bifidobacteriaceae bacterium, was cloned and expressed in Escherichia coli. The recombinant protein (BbFFase9) was successfully purified and showed a single band with a molecular mass of 66.2 kDa. This was confirmed as a β-d-fructofuranosidase and exhibited a high specific activity of 209.2 U/mg. Although BbFFase9 was a soluble protein, it exhibited excellent tolerance to proteases such as pepsin, trypsin, acidic protease, neutral protease and Flavourzyme®, indicating its potential applicability in different fields. BbFFase9 exhibited typical invertase activity, and highly catalyzed the hydrolysis of the α1↔2β glycosidic linkage in molecules containing fructosyl moieties but with no detectable fructosyltransferase activity. It was optimally active at pH 6.5 and 50 °C and stable between pH 6.0 and 9.0 at a temperature of up to 45 °C for 30 min BbFFase9 could also effectively hydrolyze galacto-oligosaccharides, which are a flatulence factor in soybean meal, thus releasing new types of product such as melibiose and mannotriose, or degrading them into invert sugars, the sweeter fructose and glucose. This study is the first to report the application of this type of β-d-fructofuranosidase.

Ribosomally derived lipopeptides containing distinct fatty acyl moieties.

Project description:Lipopeptides represent a large group of microbial natural products that include important antibacterial and antifungal drugs and some of the most-powerful known biosurfactants. The vast majority of lipopeptides comprise cyclic peptide backbones N-terminally equipped with various fatty acyl moieties. The known compounds of this type are biosynthesized by nonribosomal peptide synthetases, giant enzyme complexes that assemble their products in a non-gene-encoded manner. Here, we report the genome-guided discovery of ribosomally derived, fatty-acylated lipopeptides, termed selidamides. Heterologous reconstitution of three pathways, two from cyanobacteria and one from an arctic, ocean-derived alphaproteobacterium, allowed structural characterization of the probable natural products and suggest that selidamides are widespread over various bacterial phyla. The identified representatives feature cyclic peptide moieties and fatty acyl units attached to (hydroxy)ornithine or lysine side chains by maturases of the GCN5-related N-acetyltransferase superfamily. In contrast to nonribosomal lipopeptides that are usually produced as congener mixtures, the three selidamides are selectively fatty acylated with C10, C12, or C16 fatty acids, respectively. These results highlight the ability of ribosomal pathways to emulate products with diverse, nonribosomal-like features and add to the biocatalytic toolbox for peptide drug improvement and targeted discovery.

Aminopolyols from Carbohydrates: Amination of Sugars and Sugar-Derived Tetrahydrofurans with Transaminases.

Project description:Carbohydrates are the major component of biomass and have unique potential as a sustainable source of building blocks for chemicals, materials, and biofuels because of their low cost, ready availability, and stereochemical diversity. With a view to upgrading carbohydrates to access valuable nitrogen-containing sugar-like compounds such as aminopolyols, biocatalytic aminations using transaminase enzymes (TAms) have been investigated as a sustainable alternative to traditional synthetic strategies. Demonstrated here is the reaction of TAms with sugar-derived tetrahydrofuran (THF) aldehydes, obtained from the regioselective dehydration of biomass-derived sugars, to provide access to cyclic aminodiols in high yields. In a preliminary study we have also established the direct transamination of sugars to give acyclic aminopolyols. Notably, the reaction of the ketose d-fructose proceeds with complete stereoselectivity to yield valuable aminosugars in high purity.
