N'-(3-Methyl-quinoxalin-2-yl)-N'-phenyl-benzohydrazide.

ABSTRACT: In the crystal structure of the title compound, C(22)H(18)N(4)O, the quinoxaline system makes dihedral angles of 86.59?(7) and 63.37?(9)° with the benzohydrazide and phenyl rings, respectively. The benzohydrazide ring makes a dihedral angle of 72.46?(10)° with the phenyl ring. The crystal structure is stabilized by inter-molecular N-H?O hydrogen bonds, C-H?O contacts and C-H?? inter-actions.

The mean plane through the fused-ring system is almost perpendicular to the terminal phenyl ring, as indicated by the dihedral angle of 70.05?(8)°. In the crystal, the main and solvent mol-ecules are linked by N-H?O hydrogen bonds, forming a layer parallel to (010).

Convenient Synthesis and Anticancer Activity of Methyl 2-[3-(3-Phenyl-quinoxalin-2-ylsulfanyl)propanamido]alkanoates and N-Alkyl 3-((3-Phenyl-quinoxalin-2-yl)sulfanyl)propanamides.

Project description:A series of methyl 2-[3-(3-phenyl-quinoxalin-2-ylsulfanyl)propanamido]alkanoates and their corresponding hydrazides and N-alkyl 3-((3-phenylquinoxalin-2-yl)sulfanyl)propanamides were prepared on the basis of the chemoselective Michael reaction of acrylic acid with the parent substrate 3-phenylquinoxaline-2(1H)-thione. The parent thione was produced by a convenient novel thiation method from the corresponding 3-phenylquinoxalin-2(1H)-one. The chemical structures of the newly synthesized compounds were confirmed by elemental analyses, 1H and 13C NMR. The antiproliferative activity of the synthesized compounds was tested against human HCT-116 and MCF-7 cell lines. Out of 25 screened derivatives, 10 active compounds exhibited IC50's in the range 1.9-7.52 ?g/mL on the HCT-116, and 17 active compounds exhibited IC50's in the range 2.3-6.62 ?g/mL on the MCF-7 cell lines compared to the reference drug doxorubicin (IC50 3.23 ?g/mL). The structure-activity relationship of the tested compounds was studied through their binding affinity to the human thymidylate synthase allosteric site in silico using molecular docking and proved the quinoxaline ring as a suitable scaffold carrying a peptidomimetic side chain in position 3.

Project description:The mol-ecule of the title compound, C42H32N6O, is built up from one pyrazole ring linked to three phenyl rings and to an approximately planar [maximum deviation = 0.0455?(15)?Å] quinoxaline system connected to a phenyl-benzohydrazide group. The pyrazole ring assumes an envelope conformation, the C atom attached to the quinoxalin-3-yl ring system being the flap atom. The dihedral angle between the two phenyl rings of the phenyl-benzohydrazide group is of 58.27?(9)°. The mean plane through the pyrazole ring is nearly perpendicular to the quinoxaline ring system and to the phenyl ring attached to the opposite side, forming dihedral angles of 82.58?(7) and 87.29?(9)°, respectively. An intra-molecular C-H?O hydrogen bond is present. In the crystal, mol-ecules are linked by pairs of N-H?N hydrogen bonds, forming inversion dimers, which are further connected by C-H?N hydrogen bonds into chains parallel to the b axis.

Project description:In the title compound, C(22)H(18)N(4)O(2)S, the seven-membered ring generated by an intra-molecular N-H?O hydrogen bond adopts an envelope conformation in both of the two independent mol-ecules in the asymmetric unit. In the crystal, mol-ecules are linked into C(9) chains along [100] by N-H?O hydrogen bonds. The mol-ecules are also weakly linked by C-H?O and C-H?N inter-actions, forming dimers with edge-connected R(2) (2)(9) rings. The dimers are inter-linked by further weak C-H?N hydrogen bonds into chains along [010].

Project description:The quinoxaline system in the title hydrate, C(15)H(13)N(3)·H(2)O, is roughly planar, the r.m.s. deviation for the 18 non-H atoms being 0.188 Å; this conformation features a short intra-molecular C-H⋯N(pyrazine) inter-action. In the crystal, the amine H atom forms an N-H⋯O hydrogen bond to the water mol-ecule, which in turn forms two O-H⋯N hydrogen bonds to the pyrazine N atoms of different organic mol-ecules. These inter-actions lead to supra-molecular arrays in the bc plane that are two mol-ecules thick; additional π-π inter-actions stabilize the layers [ring centroid-centroid distance = 3.5923 (7) Å]. The layers stack along the a-axis direction via C-H⋯π contacts.

Project description:In the title mol-ecule, C(24)H(18)N(2)O(3)S, the 13-atom ring system comprising the quinoxaline and fused five-membered ring exhibits an r.m.s. deviation from coplanarity of 0.039?Å, with a maximum deviation of 0.0710?(10)?Å for the PhCO-bearing C atom of the five-membered ring. The 10-membered C(8)N(2) quinoxaline ring system has an r.m.s. deviation from coplanarity of 0.022?Å, with a maximum deviation of 0.0403?(9)?Å for the C atom involved in the C=C bond in the five-membered ring. The three atoms of the five-membered ring fused to the quinoxaline ring system show deviations of up to 0.118?(2)?Å for the PhCO-bearing C atom. C-N bond distances in the quinoxaline ring system of the title mol-ecule deviate from those in unsubstituted quinoxaline. In particular, the two C-N distances to the N atom involved in the five-membered ring are essentially equal, with values of 1.3786?(17) and 1.3773?(16)?Å, unlike the difference of nearly 0.06?Å in quinoxaline.
