Enantioselective hydroformylation of aniline derivatives.

ABSTRACT: We have developed a ligand that reversibly binds to aniline substrates, allowing for the control of regioselectivity and enantioselectivity in hydroformylation. In this paper we address how the electronics of the aniline ring affect both the binding of the substrate to the ligand and the enantioselectivity in this reaction.

Project description:Enantioenriched aldehydes are produced through asymmetric hydroformylation of styrene derivatives using BIBOP-type ligands. The featured example is enantioselective synthesis of 4-methyl-3,4-dihydroisocoumarin, which was prepared in a 95.1:4.9 enantiomeric ratio from asymmetric hydroformylation of ethyl 2-vinylbenzoate followed by in situ lactonization during the reduction process. The conditions are compatible with both electron-rich and electron-poor substituents.

Silicon-oriented regio- and enantioselective rhodium-catalyzed hydroformylation.

Project description:Hydroformylation of 1,2-disubstituted alkenes usually occurs at the ? position of the directing heteroatom such as oxygen atom and nitrogen atom. By contrast, to achieve hydroformylation on the ? position of the heteroatom is a tough task. Herein, we report the asymmetric rhodium-catalyzed hydroformylation of 1,2-disubstituted alkenylsilanes with excellent regioselectivity at the ? position (relative to the silicon heteroatom) and enantioselectivity. In a synthetic sense, we achieve the asymmetric hydroformylation on the ? position of the oxygen atom indirectly by using the silicon group as a surrogate for the hydroxyl. Density functional theory (DFT) calculations are carried out to examine energetics of the whole reaction path for Rh/YanPhos-catalyzed asymmetric hydroformylation and understand its regioselectivity and enantioselectivity. Our computational study suggests that the silicon group can activate the substrate and is critical for the regioselectivity.

Enantioselective Hydroformylation of 1-Alkenes with Commercial Ph-BPE Ligand.

Project description:A rhodium complex, in conjunction with commercially available Ph-BPE ligand, catalyzes the branch-selective asymmetric hydroformylation of 1-alkenes and rapidly generates ?-chiral aldehydes. A wide range of terminal olefins including 1-dodecene were examined, and all delivered high enantioselectivity (up to 98:2 er) as well as good branch:linear ratios (up to 15:1).

Application of a chiral scaffolding ligand in catalytic enantioselective hydroformylation.

Project description:The synthesis of β-amino-aldehydes has been achieved through enantioselective hydroformylation of PMP-protected allylic amines. The reaction is accomplished by using a scalemic scaffolding ligand that covalently and reversibly binds to the substrate. These ligands behave like chiral auxiliaries because they are covalently attached to the substrate during hydroformylation; however, similar to traditional asymmetric ligands, they can be used in catalytic quantities. The directed hydroformylation of disubstituted olefins occurs under mild conditions (35 °C and 50 psi CO/H(2)), and Z-olefins afford excellent enantioselectivities (up to 93% ee).

Radical ?-C-H Cyclobutylation of Aniline Derivatives.

Project description:A catalytic system has been developed for the direct alkylation of ?-C-H bonds of aniline derivatives with strained C-C ?-bonds. This method operates through a photoredox mechanism in which oxidative formation of aminoalkyl radical intermediates enables addition to a bicyclobutane derivative, giving rise to ?-cyclobutyl N-alkylaniline products. This mild system proceeds through a redox- and proton-neutral mechanism and is operational for a range of substituted arylamine derivatives.

Enantioselective hydroformylation of N-vinyl carboxamides, allyl carbamates, and allyl ethers using chiral diazaphospholane ligands.

Project description:Rhodium complexes of diazaphospholane ligands catalyze the asymmetric hydroformylation of N-vinyl carboxamides, allyl ethers, and allyl carbamates; products include 1,2- and 1,3-aminoaldehydes and 1,3-alkoxyaldehydes. Using glass pressure bottles, short reaction times (generally less than 6 h), and low catalyst loading (commonly 0.5 mol %), 20 substrates are successfully converted to chiral aldehydes with useful regioselectivity and high enantioselectivity (up to 99% ee). Chiral Roche aldehyde is obtained with 97% ee from the hydroformylation of allyl silyl ethers. Commonly difficult substrates such as 1,1- and 1,2-disubstituted alkenes undergo effective hydroformylation with 89-97% ee and complete conversion for six examples. Palladium-catalyzed aerobic oxidative amination of allyl benzyl ether followed by enantioselective hydroformylation yields the ?(3)-aminoaldehyde with 74% ee.

The fluorescence of indoles and aniline derivatives.

Project description:1. The variations in the excitation and fluorescence wavelengths and fluorescence intensities of a number of indole and aniline derivatives over a wide range of acidity and alkalinity (36n-sulphuric acid to 10n-potassium hydroxide) have been studied. 2. The changes in fluorescence with pH of the indoles and anilines had many characteristics in common, and the most fluorescent species were found to be the non-ionized or neutral forms showing fluorescence maxima at about lambda 350mmu. 3. In 10n-potassium hydroxide most of the compounds examined, except those containing a tertiary nitrogen atom, showed a bathochromic shift in fluorescence wavelength attributable to an anion due to a negatively charged nitrogen, but in strong acid (3n-sulphuric acid) these compounds were non-fluorescent, except the anisidines and the 5-hydroxyindoles. 4. p-Anisidine but not the o- and m-isomers showed excited-state ionization in acid solution. 5. Of the hydroxyindoles only the 5-hydroxy derivatives showed a fluorescence (lambda(max.) 520-540mmu) in acid solution. It is suggested that this fluorescence is due to a proton-transfer reaction in the excited state, and various arguments for this suggestion are given. 6. Stokes shifts for the various ionic and neutral species of the indoles and anilines have been calculated, and the large shifts found with indole and p-anisidine may be due to solvent-solute interaction.

A structural study of 2,4-di-methyl-aniline derivatives.

Project description:Crystallographic studies of nitro-gen-containing small mol-ecules aid in the elucidation of their structure-activity relationships and modes of aggregation. In this study, two previously synthesized mol-ecules are crystallographically characterized for the first time. Reaction of 2,4-di-methyl-aniline with N-bromo-succinimide affords the ortho-brominated derivative 2-bromo-4,6-di-methyl-aniline (1; C8H10BrN), which sublimates in vacuo to afford crystals featuring hydrogen-bonded chains as well as Type I halogen-halogen inter-actions. Conversely, alkyl-ation of two equivalents of 2,4-di-methyl-aniline with 1,2-di-bromo-ethane affords a separable mixture of N,N'-bis-(2,4-di-methyl-phen-yl)piperazine (2; C20H26N2), which was crystallographically characterized, as well as N,N'-bis-(2,4-di-methyl-phen-yl)ethyl-enedi-amine (3).

Palladium-catalyzed enantioselective alkenylation of alkenylbenzene derivatives.

Project description:A regioselective and enantioselective palladium-catalyzed relay Heck alkenylation of alkenylbenzene derivatives to construct remote stereocenters is disclosed. Various β-substituted styrenes were readily obtained in moderate yields with good to excellent levels of enantioselectivity. This strategy provides rapid access to enantioenriched δ, ε, ζ, and η-alkenyl aryl compounds from simple starting materials. Mechanistic studies suggest that termination of the relay reaction is controlled by affinity of the arene for the Pd complex during migration.

Diastereo- and enantioselective preparation of cyclopropanol derivatives.

Project description:The diastereoselective carbocupration reaction of alkoxy-functionalized cyclopropene derivatives, followed by a subsequent trapping of the resulting cyclopropylmetal species with an electrophilic source of oxygen (oxenoid) afforded various tetrasubstituted cyclopropanol derivatives in high diastereo- and enantiomeric ratios. Similarly, the enantioselective copper-catalyzed carbomagnesiation/oxidation (or amination) sequence on achiral nonfunctionalized cyclopropenes provided the desired cyclopropanol (and cyclopropylamine) derivatives in excellent diastereo- and enantiomeric excesses.
