ABSTRACT: Three species of land leeches, including a new combination Haemadipsa rjukjuanacomb. n., a new record for Haemadipsa picta Moore, as well as an updated description for Tritetrabdella taiwana (Oka), are reported in this study. Morphological characters and DNA barcode analysis were used to identify these species. In addition, since Haemadipsa rjukjuana had been regarded as a variety of the Japanese land leech Haemadipsa japonica for a century, morphological differences between these two species were also compared.

Molecular detection of Bartonella spp. in terrestrial leeches (Haemadipsa rjukjuana) feeding on human and animal blood in Gageo-do, Republic of Korea.

Project description:BackgroundLeeches can transmit pathogens and are therefore potentially hazardous to human and animal health. However, only a few studies of diseases transmitted by land leeches have been reported. The purpose of the present study was to analyse which pathogens are carried in Haemadipsa rjukjuana, the first recorded sanguivorous land leech in the Republic of Korea (ROK).FindingsA total of 173 H. rjukjuana were collected from Mt. Dock-Sil on Gageo-do Island, ROK during July 2011. Conventional PCR was conducted for analysis of the origin of blood meal, as well as for detection of species of Anaplasma, Bartonella, Borrelia, Ehrlichia, Rickettsia, and Wolbachia in the leech specimens. Bartonella DNA was detected in eight of the specimens studied based on partial ITS sequence analysis. Seven of the DNA samples were closely related to Bartonella grahamii (99.6-100 % similarity), and one sample exhibited a 90.6 % similarity with Bartonella sp. from Taiwan. Sequences of the mitochondrial cytochrome b gene were generated for a total of 35 of the 173 leech internal organ samples. These included sequences of human (n = 10), mouse (n = 8), weasel (n = 6) and bird (n = 11) origin. Of these 35 sequences, 68.5 % were from mammals, including humans, and 31.4 % were from migratory birds that pass through Gageo-do, ROK.ConclusionsAlthough the present study does not provide evidence that leeches indeed transmit Bartonella species to hosts directly, to our knowledge this is the first report on Bartonella DNA being detected from leeches. Therefore, further studies are needed to explore the possibility of zoonotic pathogen transmission by land leeches.

Characterization of the complete mitogenome of a land leech, <i>Haemadipsa crenata</i> Ngamprasertwong (Arhynchobdellida: Haemadipsidae).

Project description:Land leeches of genus Haemadipsa (Family Haemadipsidae) are widely distributed in South East Asia. Haemadipsa crenata Ngamprasertwong is a blood-feeding species firstly reported from Thailand. A complete mitochondrial genome of H. crenata was characterized in this study for further genetic exploration on land leech. The reads were assembled into a circular mitogenome of 14,725 bp in length. The AT content of H. crenata mitogenome is 76.79%. The annotated mitogenome contains 22 tRNAs, 2 rRNAs, and 13 protein-coding genes (PCGs), and the structure of PCG open reading frames was confirmed. Finally, the phylogenetic relationship of H. crenata and other leech species were reconstructed using mitogenomes.

Project description:BackgroundSawyer (1986) included three species in the nasal leech genus LimnatisMoquin-Tandon 1827: Limnatisnilotica (Savigny 1822), LimnatisbacescuiManoleli 1972 and Limnatispaluda (Tennent 1859). The first and last species have mainly been identified in Middle Eastern countries (e.g. Kinzelbach and Rückert 1985). The second species has been identified only in Romania Dobruja (Manoleli 1972). Although Limnatis leeches are well known species of endoparasitic leeches, Limnatisnilotica was recorded only once in Kazakhstan (Lukin 1976).New informationSpecimens of the genus Limnatis from Almaty Province, Kazakhstan are identified as Limnatispaluda. This is the first record of Limnatispaluda from Kazakhstan. Mitochondrial COI and 12S data demonstrated that the present specimens are genetically close to an Israeli specimen identified as Limnatisnilotica. In addition, molecular data suggest that some Limnatis specimens whose DNA sequences have been reported were misidentified. According to the observed phylogenetic relationships, the taxonomic status of the known Limnatis species should be revisited.

Nuclear microsatellite and mitochondrial DNA analyses reveal the regional genetic structure and phylogeographical history of a sanguivorous land leech, <i>Haemadipsa japonica</i>, in Japan.

Project description:Recent molecular studies have indicated that phylogeographical history of Japanese biota is likely shaped by geohistory along with biological events, such as distribution shifts, isolation, and divergence of populations. However, the genetic structure and phylogeographical history of terrestrial Annelida species, including leech species, are poorly understood. Therefore, we aimed to understand the genetic structure and phylogeographical history across the natural range of Haemadipsa japonica, a sanguivorous land leech species endemic to Japan, by using nine polymorphic nuclear microsatellites (nSSR) and cytochrome oxidase subunit one (COI) sequences of mitochondrial DNA (mtDNA). Analyses using nSSR revealed that H. japonica exhibited a stronger regional genetic differentiation among populations (G'ST = 0.77) than other animal species, probably because of the low mobility of land leech. Analyses using mtDNA indicated that H. japonica exhibited two distinct lineages (A and B), which were estimated to have diverged in the middle Pleistocene and probably because of range fragmentation resulting from climatic change and glacial and interglacial cycles. Lineage A was widely distributed across Japan, and lineage B was found in southwestern Japan. Analyses using nSSR revealed that lineage A was roughly divided into two population groups (i.e., northeastern and southwestern Japan); these analyses also revealed a gradual decrease in genetic diversity with increasing latitude in lineage A and a strong genetic drift in populations of northeastern Japan. Combined with the largely unresolved shallow polytomies from the mtDNA phylogeny, these results implied that lineage A may have undergone a rapid northward migration, probably during the Holocene. Then, the regional genetic structure with local unique gene pools may have been formed within each lineage because of the low mobility of this leech species.
