Coccinia intermedia - a new Cucurbitaceae species from West Africa.

ABSTRACT: Nuclear and plastid sequences from two individuals of a suspected new species of Coccinia from West Africa were added to an available molecular phylogeny for the remaining 27 species of the genus. Phylogenetic analyses of these data indicate the new species' monophyletic status and closest relatives. Based on four fertile collections, we here describe and illustrate Coccinia intermedia Holstein. We also provide a key to the Coccinia species of West Africa and map their distributions.

Project description:This monograph deals with all 95 names described in the Cucurbitaceae genus Coccinia and recognizes 25 species. Taxonomic novelties are Cocciniaadoensisvar.aurantiaca (C.Jeffrey) Holstein, stat. nov., Cocciniasessilifoliavar.variifolia (A.Meeuse) Holstein, stat. nov., and Cocciniaadoensisvar.jeffreyana Holstein, var. nov. For the 25 species 3157 collections were examined, of which 2024 were georeferenced to produce distribution maps. All species are distributed in sub-Saharan Africa with one species, Cocciniagrandis, extending from Senegal in West Africa east to Indonesia and being naturalized on Pacific Islands, in Australia, the Caribbean, and South America. Coccinia species are dioecious creepers or climbers with simple or bifid tendrils that occupy a range of habitats from arid scrubland, woodlands to lowland rainforest and mist forest. The corolla of Coccinia species is sympetalous, usually pale yellow to orange, and 1 to 4.5 cm long. Pollination is by bees foraging for pollen or nectar. After pollination, the developing ovary often exhibits longitudinal mottling, which usually disappears during maturation. All species produce berries with a pericarp in reddish colors (orange-red through to scarlet red), hence the generic name. The globose to cylindrical fruits contain numerous grayish-beige flat to lenticular seeds. Chromosome numbers are 2n = 20, 24, and 22 + XX/XY. Many Coccinia species are used for food, either as roasted tubers, greens as spinach, or the fruits as vegetables. Medicinal value is established in Cocciniagrandis, of which leaves and sap are used against diabetes.

A new genus and species of pennatulacean octocoral from equatorial West Africa (Cnidaria, Anthozoa, Virgulariidae).

Project description:A new genus and species of sea pen or virgulariid pennatulacean from the Gulf of Guinea in the tropical eastern Atlantic is described, and a key to the genera of the Virgulariidae is included. The new genus and species described here adds to the previously described five other genera of the family. It is distinguished by unique sclerite and polyp leaf characters from the superficially-similar genus Virgularia, which lacks conspicuous sclerites in the polyp leaves and coenenchyme (other than minute oval bodies that are generally <0.01 mm in length).

Cryptomyrus: a new genus of Mormyridae (Teleostei, Osteoglossomorpha) with two new species from Gabon, West-Central Africa.

Project description:We use mitochondrial and nuclear sequence data to show that three weakly electric mormyrid fish specimens collected at three widely separated localities in Gabon, Africa over a 13-year period represent an unrecognized lineage within the subfamily Mormyrinae and determine its phylogenetic position with respect to other taxa. We describe these three specimens as a new genus containing two new species. Cryptomyrus, new genus, is readily distinguished from all other mormyrid genera by a combination of features of squamation, morphometrics, and dental attributes. Cryptomyrus ogoouensis, new species, is differentiated from its single congener, Cryptomyrus ona, new species, by the possession of an anal-fin origin located well in advance of the dorsal fin, a narrow caudal peduncle and caudal-fin lobes nearly as long as the peduncle. In Cryptomyrus ona, the anal-fin origin is located only slightly in advance of the dorsal fin, the caudal peduncle is deep and the caudal-fin lobes considerably shorter than the peduncle. Continued discovery of new taxa within the "Lower Guinea Clade" of Mormyridae highlights the incompleteness of our knowledge of fish diversity in West-Central Africa. We present a revised key to the mormyrid genera of Lower Guinea.

Zehneria grandibracteata (Cucurbitaceae), an overlooked new species from western Kenyan forests.

Project description:Zehneria grandibracteata, a new species of Cucurbitaceae from western Kenya, is described here, based on morphological and molecular data. It has long been misidentified as the widely-distributed species Z. scabra. However, it differs by its ovate leafy probract at the base of the inflorescences, subglabrous condition of the entire plant, shorter receptacle-tube and filaments, as well as denser and sessile inflorescences. Furthermore, the molecular phylogenetic analysis of Zehneria, based on nrITS sequences, further supports the argument that Z. grandibracteata should be segregated from Z. scabra.

Rhamnella intermedia (Rhamnaceae), a new evergreen species from southwest Guangxi.

Project description:Rhamnella intermedia, a new evergreen species from southwest Guangxi, is described and illustrated in this study. This species is similar to R. brachycarpa by the size and ratio of length to width of dried fruit and seeds, by which it differs from R. rubrinervis and R. tonkinensis. However, it differs from R. brachycarpa by rarely mucronate seed apices, larger ratio of length to width of leaves, leaf apices acuminate to long acuminate, shorter leaf petioles, and longer fruiting pedicels. Principal component analysis based on phenotypic traits further recognised three separated groups. Rhamnella rubrinervis and R. tonkinensis were clustered into one group; the other two groups represented R. brachycarpa and two Guangxi populations, respectively. Furthermore, phylogenetic analysis of nuclear ITS sequence variations highly supported that the two Guangxi populations represented an independent evolutionary lineage and were closest to R. rubrinervis. Four fixed nucleotide sites were found and were different from R. rubrinervis. However, besides the differentiated traits in seeds and fruit, densely pilose young branches also separated them from R. rubrinervis. In addition, during our field investigations, none of the three closely related species were found at locations where this new species was distributed. Therefore, this new species, based on the two Guangxi populations, is named R. intermedia. The key to four closely related species is also presented.

The genus Lactarius s. str. (Basidiomycota, Russulales) in Togo (West Africa): phylogeny and a new species described.

Project description:Lactarius s. str. represents a monophyletic group of about 40 species in tropical Africa, although the delimitation of the genus from Lactifluus is still in progress. Recent molecular phylogenetic and taxonomic revisions have led to numerous changes in names of tropical species formerly referred to Lactarius. To better circumscribe the genus Lactarius in Togo, we combined morphological data with sequence analyses and phylogeny inference of rDNA ITS sequences. Morphological and molecular data were generated from specimens sampled in various native woodlands and riverside forests; Lactarioid- and Russula sequences from public GenBank NCBI, and UNITE are included for phylogenetic analysis. The Maximum likelihood phylogeny tree inferred from aligned sequences supports the phylogenetic position of the studied samples from Togo within the subgenera Piperites, and Plinthogali. Lactarius s. str. includes about 13 species described from West Africa, of which eight were not previously known from Togo, including one new species: Lactarius subbaliophaeus identifiable by the presence of winged basidiospores, a pallisadic pileipellis with a uprapellis composed of cylindrical cells, inconspicuous pleurocystidia, and fusiform or tortuous, often tapering apex marginal cells. It can also be recognised by a transparent white latex that turns pinkish and then blackish, and a bluish reaction of the flesh context with FeSO4. These features mentioned do not match any of the morpho-anatomically most similar species, notably L. baliophaeus and L. griseogalus.

Molecular characterization of β-thalassemia intermedia in the West Bank, Palestine.

Project description:BackgroundWe aimed to investigate the molecular basis of β-Thalassemia intermedia (TI) in the West Bank region and its management practices.MethodsThis was a case series multi-center study and included 51 cases of TI. DNA sequencing was used to analyze β-globin gene mutations. Common α-globin gene mutations were screened by Gap-PCR (-α3.7, -α4.2, --MED, αααanti3.7) or DNA sequencing (α2-IVS II 5 nt del). XmnI -158 C > T polymorphisms of Gγ-globin gene was determined by RFLP-PCR.ResultsSeven β-globin gene mutations were observed, namely IVS-I -6 C > T, IVS-I-110 G > A, IVS-II-1 G > A, IVS-I-1 G > A, Codon 37 Trp > Stop, beta - 101 and IVS-II-848 C > A. Ten genotypes were observed. Homozygosity for IVS-I-6 accounted for the majority of TI cases with a frequency of 74.5%. The second common β-globin gene genotype was homozygote IVS-I-110 G > A (5.8%) and homozygote IVS-II-1 G > A (5.8%). The remaining seven genotypes were each detected in about 2% of patients. α-Thalassemia mutations were seen in five patients (9.8%), and included (-α3.7, αααanti3.7 and α2-IVSII-5 nt del). XmnI polymorphism was observed in four patients (7.8%), three homozygotes and one heterozygote.ConclusionsHomozygosity for the mild β-globin gene IVS-I-6 allele was the major contributing factor for the TI phenotype among the study subjects. The role of XmnI SNP and α-thalassemia mutations in ameliorating the TI phenotype was observed in few patients for each factor. The beta - 101 C > T mutation was diagnosed in one patient in homozygote state for the first time in Palestine.

Phylogenetic and microscopic studies in the genus Lactifluus (Basidiomycota, Russulales) in West Africa, including the description of four new species.

Project description:Despite the crucial ecological role of lactarioid taxa (Lactifluus, Lactarius) as common ectomycorrhiza formers in tropical African seasonal forests, their current diversity is not yet adequately assessed. During the last few years, numerous lactarioid specimens have been sampled in various ecosystems from Togo (West Africa). We generated 48 ITS sequences and aligned them against lactarioid taxa from other tropical African ecozones (Guineo-Congolean evergreen forests, Zambezian miombo). A Maximum Likelihood phylogenetic tree was inferred from a dataset of 109 sequences. The phylogenetic placement of the specimens, combined with morpho-anatomical data, supported the description of four new species from Togo within the monophyletic genus Lactifluus: within subgen. Lactifluus (L. flavellus), subgen. Russulopsis (L. longibasidius and L. pectinatus), and subgen. Edules (L. melleus). This demonstrates that the current species richness of the genus is considerably higher than hitherto estimated for African species and, in addition, a need to redefine the subgenera and sections within it.

Small RNA Solexa sequencing of 4 Plant Species in the Cucurbitaceae family

Project description:Total RNA was isolated from different tissues (leaf, stem and flesh, rind and placenta of the fruits) using TRIzol reagent and small RNA libraries were generated from four cucurbit species: bottle gourd (Lagenaria siceraria (accession Grif 1617 collection from India)), Cucurbita moschata (accession Grif 14244 Early Butternut) Cucurbita pepo (accession NSL98075 Table King), and watermelon (Citrullus lanatus var. lanatus) (PI 438676 Charleston Grey) by pooling equimolar amounts of total RNA from the aforesaid tissues. Construction of small RNA libraries from size fractionated RNA was carried out as described previously. In brief, small RNA fractions of 18–28 nt were isolated from 15% denaturing polyacrylamide gels and sequentially ligated to 5′ and 3′ RNA adapters. Small RNAs ligated with adapters were converted to DNA by RT-PCR following Solexa protocol. The final PCR product was gel purified and sequenced by Genome Analyser II (Illumina).
