Synthesis and stability of new spiroaminoborate esters.

ABSTRACT: New spiroaminoborate esters derived from 1,1-diphenylprolinol, ephedrine and dihydroquinine with different alkoxy substituents were prepared as stable crystalline compounds and characterized by spectroscopical analysis and specific rotation. The structure of the spiroborate 4 derived from 1,1-diphenylprolinol and dicyclohexyl-1,1'-diol was confirmed by X-ray analysis.

Project description:We report here the synthesis and selected properties of various silicate ester derivatives (tetraalkoxysilanes) of the taxanes paclitaxel (PTX) and docetaxel (DTX) [i.e., PTX-OSi(OR)3 and DTX-OSi(OR)3]. Both the hydrophobicity and hydrolytic lability of these silicates can be (independently) controlled by choice of the alkyl group (R). The synthesis, structural characterization, hydrolytic reactivity, and in vitro cytotoxicity against the MDA-MB-231 breast cancer cell line of most of these derivatives are described. We envision that the greater hydrophobicity of these silicates (vis-à-vis PTX or DTX itself) should be advantageous from the perspective of preparation of stable aqueous dispersions of amphiphilic block-copolymer-based nanoparticle formulations.

Tricyclic Derivative of Acyclovir and Its Esters in Relation to the Esters of Acyclovir Enzymatic Stability: Enzymatic Stability Study.

Project description:The 3,9-dihydro-3-[(2-hydroxyethoxy)methyl]-6-(4-methoxyphenyl)-9-oxo-5H-imidazo[1,2-a]-purine (6-(4-MeOPh)-TACV) was selected to assess the enzymatic stability of the tricyclic acyclovir derivatives from the imidazo[1,2-a]-purine group. The parent compound and its esters (acetyl, isobutyryl, pivaloyl, nicotinic, ethoxycarbonyl) were subjected to kinetic studies and compared with the stability of analogous acyclovir (ACV) esters. The enzymatic hydrolysis was observed in vitro in a medium of 80% human plasma in the absence and presence of porcine liver esterase (PLE). The tests were carried out at 37 °C. To determine the kinetic parameters (kobs., t0.5) of the observed reaction, the validated HPLC-UV method in the reversed phase was used. The HPLC-MS/MS method was used to identify the degradation products under the tested conditions. In summary, it was found that 6-(4-MeOPh)-TACV esters are more susceptible to esterase metabolism than ACV esters. It was confirmed by HPLC-MS/MS that in the plasma, the main product of their hydrolysis is 6-(4-MeOPh)-TACV and not ACV, which confirms that their antiviral activity observed in vitro does not result from ring degradation.

Synthesis, Optical, and Geometrical Approaches of New Natural Fatty Acids' Esters/Schiff Base Liquid Crystals.

Project description:Schiff base liquid crystals, known as [4-(hexyloxy)phenylimino)methyl]phenyl palmitate (IA), [4-(hexyloxy)phenylimino)methyl]phenyl oleate (IIA) and [4-(hexyloxy)phenylimino)methyl]phenyl linoleate (IIIA), were synthesized from palmitic, oleic, and linoleic natural fatty acids. The prepared compounds have been investigated for their thermal and optical behavior as well as phase formation using differential scanning calorimetry (DSC) and polarized optical microscopy (POM). Molecular structures of all studied compounds were confirmed via elemental analysis, FT-IR, 1H NMR, and 13C NMR. Smectic phase is the observed mesophase for all compounds; however, their type and range depend upon the terminal alkanoate chains attached to the phenyl ring. Computational calculations, Density functional theory (DFT), energy difference of the frontier molecular orbital (FMOs), as well as the thermodynamic parameters of different molecular configurations isomers were discussed. It was found that the mesophase behavior and the geometrical characteristics were affected by the degree of unsaturation of fatty terminal chains. Furthermore, the geometrical structure of the CH=N linkage plays an important role in the thermal stability and optical transition temperature.

Synthesis of Chiral Allylic Esters by Using the New Recyclable Chiral Heterogeneous Oxazoline-Based Catalysts.

Project description:A new class of recyclable supported chiral heterogeneous ligands has been synthesized by the reaction of functionalized mesoporous SBA-15 with aliphatic- and aromatic-substituted chiral amino oxazoline ligands. The obtained chiral heterogeneous ligands were characterized by several techniques such as Fourier transform infrared, X-ray diffraction, thermogravimetric analysis, scanning electron microscopy, energy-dispersive X-ray spectroscopy, and BET-BJH. The application of these new heterogeneous ligands in copper-catalyzed asymmetric allylic oxidation of olefins by using perester showed better yields up to 95% and better enantioselectivities up to 96% compared to the corresponding homogeneous catalysts. These findings can be considered as an important step in the advancement of green chemistry. Investigation of the recyclability of the catalysts confirmed that they were easily recovered and reused eight times without significant losses in reactivity, yield, and enantioselectivity.

A new strategy for the synthesis of chiral ?-alkynyl esters via sequential palladium and copper catalysis.

Project description:A new strategy for the synthesis of chiral ?-alkynyl esters which relies on sequential Pd and Cu catalysis is reported. Terminal alkynes bearing aryl, alkyl, and silyl groups can be employed without prior activation yielding a wide range of important chiral building blocks. The reaction sequence utilizes a robust Pd(II)-catalyzed hydroalkynylation of ynoates with terminal alkynes providing geometrically pure ynenoates which are readily reduced by CuH. In contrast to previous reports, where additions to ynenoates proceed with marginal preference for the 1,6-pathway, this conjugate reduction occurs with high 1,4-selectivity yielding ?-alkynyl esters with excellent levels of enantioselectivity. Importantly, the method tolerates a wide range of functionality, including allylic carbonates and carbamates, and thus allows for rapid elaboration of the ?-alkynyl esters into a variety of chiral, substituted heterocycles.

Synthesis, metabolic stability and antiviral evaluation of various alkoxyalkyl esters of cidofovir and 9-(S)-[3-hydroxy-2-(phosphonomethoxy)propyl]adenine.

Project description:Alkoxyalkyl esters of cidofovir (CDV) are orally active agents which inhibit the replication of a variety of double stranded DNA (dsDNA) viruses including variola, vaccinia, ectromelia, herpes simplex virus, cytomegalovirus, adenovirus and others. One of these compounds, hexadecyloxypropyl-CDV (HDP-CDV, CMX001) is in clinical development for prevention and treatment of poxvirus infection, vaccination complications, and for infections caused by cytomegalovirus, adenovirus, herpesviruses and other dsDNA viruses. This class of lipid analogs is potentially prone to undergo omega oxidation of the alkyl moiety which can lead to a short chain carboxylic acid lacking antiviral activity. To address this issue, we synthesized a series of alkoxyalkyl or alkyl glycerol esters of CDV and (S)-HPMPA having modifications in the structure of the alkyl residue. Antiviral activity was assessed in cells infected with vaccinia, cowpox or ectromelia viruses. Metabolic stability was determined in S9 membrane fractions from rat, guinea pig, monkey and human liver. All compounds had substantial antiviral activity in cells infected with vaccinia, cowpox or ectromelia. Metabolic stability was lowest in monkey liver S9 incubations where rapid disappearance of HDP-CDV and HDP-(S)-HPMPA was noted. Metabolic stability in monkey preparations increased substantially when a ?-1 methyl group (15-methyl-HDP-CDV) or a terminal cyclopropyl residue (14-cyclopropyl-tetradecyloxypropyl-CDV) was present in the alkyl chain. The most stable compound was 1-O-octadecyl-2-O-benzyl-sn-glycero-3-CDV (ODBG-CDV) which was not metabolized extensively by monkey liver S9. In rat, guinea pig or human liver S9 incubations, most of the modified antiviral compounds were considerably more stable.

Implementation of a recently developed dynamic kinetic resolution of ?-substituted-?-keto esters via Ru(II)-catalyzed asymmetric transfer hydrogenation provides the title motif in routinely high diastereo- and enantioselectivity.

Synthesis and stability study of a new major metabolite of ?-hydroxybutyric acid.

Project description:?-Hydroxybutanoic acid (GHB) is used as a date-rape drug, which renders the victims unconscious and defenceless. Intoxications are very difficult to detect for forensic scientists due to rapid metabolism to endogenous levels of GHB. We recently discovered a new major metabolite, 2, of GHB (1) that could potentially extend the analytical detection window for GHB intoxications. Herein we disclose synthetic procedures based on a Koenigs-Knorr glucuronidation approach that provides GHB glucuronide 2 and a deuterium-labelled analogue d 4-2 of high purity suitable for analytical chemistry. In addition, we have assessed the stability of GHB glucuronide 2 by mimicking the natural pH range for urine, which is of importance in the development of new analytical methods. Using NMR we show that GHB glucuronide 2 is highly stable towards aqueous hydrolysis within the pH range normally observed for urine even at elevated temperature.

Modular synthesis of cell-permeating 2-ketoglutarate esters.

Project description:Cell-permeating esters of 2-ketoglutarate (2-KG) have been synthesized through a convergent sequence from two modules in two and three steps, respectively. This route provides access to a full series of mono- and disubstituted 2-KG esters, enabling us to define the effect of regioisomeric masking on metabolite release and antihypoxic activity in cell-based assays. In addition to providing insight into the biological activity of cell permeable 2-KG esters, the straightforward and modular nature of this synthetic route may prove useful for the development of next-generation 2-KG analogues for diagnostic and therapeutic applications.
