Project description:Whilst conducting surveys of lignicolous microfungi in Yunnan Province, we collected a large number of taxa that resemble Montagnula (Didymosphaeriaceae, Pleosporales). Our phylogenetic study on Montagnula involved analysing sequence data from ribosomal RNA genes (nc18S, nc28S, ITS) and protein-coding genes (rpb2, tef1-α). We present a biphasic approach (morphological and molecular phylogenetic evidence) that supports the recognition of four new species in Montagnula viz., M.lijiangensis, M.menglaensis, M.shangrilana and M.thevetiae. The global diversity of Montagnula is also inferred from metabarcoding data and published records based on field observations. Metabarcoding data from GlobalFungi and field observations provided insights into the global diversity and distribution patterns of Montagnula. Studies conducted in Asia, Australia, Europe, and North America revealed a concentration of Montagnula species, suggesting regional variations in ecological preferences and distribution. Montagnula species were found on various substrates, with sediments yielding a high number of sequences. Poaceae emerged as a significant contributor, indicating a potential association between Montagnula species and grasses. Culture-based investigations from previously published data revealed Montagnula species associations with 105 plant genera (in 45 plant families), across 55 countries, highlighting their wide ecological range and adaptability. This study enhances our understanding of the taxonomy, distribution, and ecological preferences of Montagnula species. It emphasizes their role in the decomposition of organic matter in grasslands and savannah systems and suggests further investigation into their functional roles in ecosystem processes. The global distribution patterns and ecological interactions of Montagnula species underscore the need for continued research and conservation efforts.

Project description:A new species, Echeveria coruana, is described and illustrated from the malpaís near San Andrés Corú, Michoacan, Mexico. The species belongs to series Gibbiflorae and the new taxon was compared with Echeveria purhepecha and Echeveria patriotica, with whom it shares the closest morphological affinities. Additionally, Echeveria yalmanantlaensis an endangered species from Sierra of Manantlán Biosphere Reserve, State of Colima, was also discovered near San Andrés Corú and is reported for the first time from the State of Michoacan. The conservation status of both species was (re)evaluated according to the criteria of the International Union for Conservation of Nature.

Project description:This paper represents the first article in a series on Yunnanese microfungi. We herein provide insights into Magnolia species associated with microfungi. All presented data are reported from the Kunming Botanical Gardens. Final conclusions were derived from the morphological examination of specimens coupled with phylogenetic sequence data to better integrate taxa into appropriate taxonomic ranks and infer their relationships. Shearia formosa, the type species of Shearia, lacks type material, and its phylogenetic position accordingly remains unresolved. A fresh collection of Shearia formosa, obtained from Magnolia denudata and M. soulangeana in China, therefore, designated a neotype for stabilizing the application of the species and/or genus name. Phylogenetic analyses of a combined DNA data matrix containing SSU, LSU, RPB2 and TEF loci of representative Pleosporales revealed that the genera Crassiperidium, Longiostiolum and Shearia are a well-defined monophylum. It is recognized as the family Longiostiolaceae and strongly supported by Bayesian and Maximum Likelihood methods. Its members are characterized by immersed to semi-immersed, globose to subglobose ascomata with a central, periphysate ostiole, a peridium composed of rectangular to polygonal cells, cylindrical to clavate asci, broadly fusiform, hyaline to pale brown ascospores, a coelomycetous asexual morph with pycnidial conidiomata, enteroblastic, annellidic, ampulliform, doliiform or cylindrical conidiogenous cells and cylindrical to fusiform, transverse and sometimes laterally distoseptate conidia without a sheath or with a basal lateral sheath. Nigrograna magnoliae sp. nov. is introduced from Magnolia denudata with both asexual and sexual morphs. We observed the asexual morph of Brunneofusispora sinensis from the culture and therefore amended the generic and species descriptions of Brunneofusispora.

Project description:A culture-based survey of staining fungi on oil-treated timber after outdoor exposure in Australia and the Netherlands uncovered new taxa in Pezizomycotina. Their taxonomic novelty was confirmed by phylogenetic analyses of multi-locus sequences (ITS, nrSSU, nrLSU, mitSSU, RPB1, RPB2, and EF-1?) using multiple reference data sets. These previously unknown taxa are recognised as part of a new order (Superstratomycetales) potentially closely related to Trypetheliales (Dothideomycetes), and as a new species of Cyanodermella, C. oleoligni in Stictidaceae (Ostropales) part of the mostly lichenised class Lecanoromycetes. Within Superstratomycetales a single genus named Superstratomyces with three putative species: S. flavomucosus, S. atroviridis, and S. albomucosus are formally described. Monophyly of each circumscribed Superstratomyces species was highly supported and the intraspecific genetic variation was substantially lower than interspecific differences detected among species based on the ITS, nrLSU, and EF-1? loci. Ribosomal loci for all members of Superstratomyces were noticeably different from all fungal sequences available in GenBank. All strains from this genus grow slowly in culture, have darkly pigmented mycelia and produce pycnidia. The strains of C. oleoligni form green colonies with slimy masses and develop green pycnidia on oatmeal agar. These new taxa could not be classified reliably at the class and lower taxonomic ranks by sequencing from the substrate directly or based solely on culture-dependent morphological investigations. Coupling phenotypic observations with multi-locus sequencing of fungi isolated in culture enabled these taxonomic discoveries. Outdoor situated timber provides a great potential for culturable undescribed fungal taxa, including higher rank lineages as revealed by this study, and therefore, should be further explored.

Project description:Clinanthus inflatus (Amaryllidaceae) and Ismene parviflora are described from Ancash Department in Peru. The flower of C. inflatus is urceolate, and resembles that of Urceolina (Amaryllidaceae tr. Eucharideae), a unique morphology for the genus. Ismene parviflora, with its small, loosely formed, narrowly funnelform-tubular perigone with a ventricose limb, appears to have some affinity to subgen. Pseudostenomesson and may represent an intermediate form between the former and species of subgen. Ismene. Stenomesson rubrum is transferred to Clinanthus as C. ruber on the basis of its narrowly lorate leaf morphology.

Project description:The family Phaeosphaeriaceae is a diverse group of ascomycetous fungi that are commonly found in terrestrial habitats worldwide. In recent years, there has been increasing interest in the biodiversity of Phaeosphaeriaceae in China, particularly in Sichuan Province, which has not been fully explored for its high fungal diversity. In our study, we conducted extensive surveys in Sichuan Province to identify and describe new species of Ascomycota with diverse habitats. Here, we present a taxonomic revision of Phaeosphaeriaceae with taxonomic novelties from terrestrial habitats in Sichuan Province based on a multi-gene phylogenetic approach. Our study focuses on the description of four new species of Phaeosphaeriaceae, representing novel additions to the diversity of this fungal family. Using a combination of morphological and molecular data, we provide detailed descriptions of the new taxa and their placement within the family. Additionally, we discuss the phylogenetic relationships of these new taxa with other members of Phaeosphaeriaceae, providing insights into the correct taxonomic classification of the family. Our taxonomic revision contributes to understanding fungal diversity in China and provides a foundation for future studies investigating the taxonomy and ecological roles of Phaeosphaeriaceae fungi. Furthermore, our multi-gene phylogenetic approach provides increased resolution and accuracy in the delimitation of species boundaries within the family. Our study highlights the importance of continued exploration and taxonomic revision in order to fully understand the diversity and distribution of fungal species in China and beyond. Project description:Novelties are a familiar part of daily life. They are also fundamental to the evolution of biological systems, human society, and technology. By opening new possibilities, one novelty can pave the way for others in a process that Kauffman has called "expanding the adjacent possible". The dynamics of correlated novelties, however, have yet to be quantified empirically or modeled mathematically. Here we propose a simple mathematical model that mimics the process of exploring a physical, biological, or conceptual space that enlarges whenever a novelty occurs. The model, a generalization of Polya's urn, predicts statistical laws for the rate at which novelties happen (Heaps' law) and for the probability distribution on the space explored (Zipf's law), as well as signatures of the process by which one novelty sets the stage for another. We test these predictions on four data sets of human activity: the edit events of Wikipedia pages, the emergence of tags in annotation systems, the sequence of words in texts, and listening to new songs in online music catalogues. By quantifying the dynamics of correlated novelties, our results provide a starting point for a deeper understanding of the adjacent possible and its role in biological, cultural, and technological evolution.

Project description:To account for infants' perceptual and cognitive development, the constructivist model proposes that learning a new object depends on the capability of processing simpler lower-level units, and then integrating these units into more complex higher-level units based on their relationships, such as regular co-occurrence. Here, we demonstrate that the process of associating visual and auditory attributes to build a new multisensory object representation is not only observed in the course of development, but also in the course of infants' in-the-moment information processing. After a brief familiarization session of learning two pairs of novel audiovisual stimuli, 15-month-old infants showed two components in pupil dilations over time: A rapid dilation was observed when processing perceptually novel compared to familiar stimuli, and a slower dilation was observed when processing novel combinations of familiar stimuli. However, in 10-month-old infants, only the effect elicited by novel stimuli was observed. Our results therefore demonstrate that detecting perceptual novelty occurred earlier than detecting association novelty in infants' information processing. These results support the view that infants perceive newly-learned objects by processing their constituent attributes and then integrating these components, as suggested by the constructivist model.

Project description:Treatment of locally advanced rectal cancer is evolving through surgical innovation and paradigm shifts in neoadjuvant treatment. Whereas local recurrence was a significant concern before the systematic implementation of neoadjuvant chemoradiation therapy and surgery according to total mesorectal excision principles, distant relapse remains a major drawback. Hence, efforts in recent years have focused on delivering preoperative chemotherapy regimens to overcome compliance issues with adjuvant administration. In parallel, new surgical techniques, including transanal video-assisted total mesorectal excision and robot-assisted surgery, emerged to face the challenge to navigate in the deep and narrow spaces of the pelvis. Furthermore, patients experiencing a complete response after neoadjuvant treatment might even escape surgery within a close surveillance strategy. This novel "watch and wait" concept has gained interest to improve quality of life in highly selected patients. This review summarizes recent evidence and controversies and provides an overview on timely and innovative aspects in the treatment of locally advanced rectal cancer.

Project description:Gene duplication and alternative splicing are important mechanisms in the production of genomic novelties. Previous work has shown that a gene's family size and the number of splice variants it produces are inversely related, although the underlying reason is not well understood. Here, we report that gene length and expression level together explain this relationship. We found that gene lengths correlate with both gene duplication and alternative splicing: Longer genes are less likely to produce duplicates and more likely to exhibit alternative splicing. We show that gene length is a dynamic property, increasing with evolutionary time--due in part to the insertions of transposable elements--and decreasing following partial gene duplications. However, gene length alone does not account for the relationship between alternative splicing and gene duplication. A gene's expression level appears both to impose a strong constraint on its length and to restrict gene duplications. Furthermore, high gene expression promotes alternative splicing, in particular for long genes, and alternatively, short genes with low expression levels have large gene families. Our analysis of the human and mouse genomes shows that gene length and expression level are primary genic properties that together account for the relationship between gene duplication and alternative splicing and bias the origin of novelties in the genome.
