ABSTRACT: An environmentally benign, fast and convenient protocol has been developed for the Michael addition of 1,3-dicarbonyl compounds to ?-nitroalkenes in good to excellent yields by a grinding method under catalyst- and solvent-free conditions.

Addition of 1,3-dicarbonyl compounds to terminal alkynes catalyzed by a cationic cobalt(iii) complex.

Project description:The Nakamura reaction using a cationic cobalt(iii) complex, [Cp*Co(CH3CN)3][SbF6]2 as the catalyst under neutral and aerobic conditions at 110 °C has been described. In solution, the complex is expected to lose a hemilabile acetonitrile ligand to produce a highly electron-deficient cobalt(iii) center, and the Lewis acidic nature of the cobalt center has been exploited for the enolization of the dicarbonyl compounds. The reaction of 1,3-dicarbonyl compounds with alkynes affords the corresponding alkenyl derivative. However, the reaction of phenylacetylene and its derivatives with β-ketoesters affords corresponding terphenyl compounds. Details of the mechanisms of the reactions have been proposed based on in situ LCMS measurements.

Asymmetric Michael Addition Organocatalyzed by α,β-Dipeptides under Solvent-Free Reaction Conditions.

Project description:The application of six novel α,β-dipeptides as chiral organocatalysts in the asymmetric Michael addition reaction between enolizable aldehydes and N-arylmaleimides or nitroolefins is described. With N-arylmaleimides as substrates, the best results were achieved with dipeptide 2 as a catalyst in the presence of aq. NaOH. Whereas dipeptides 4 and 6 in conjunction with 4-dimethylaminopyridine (DMAP) and thiourea as a hydrogen bond donor proved to be highly efficient organocatalytic systems in the enantioselective reaction between isobutyraldehyde and various nitroolefins.

Project description:Under dinuclear catalysis, the direct conjugate addition of 2(5H)-furanone to nitroalkenes involves the gamma-position of the nucleophile. The synthetically versatile Michael adducts are prepared in good yields, with high levels of diastereo- and enantioselectivity. A model is presented to rationalize the observed stereoselectivity.

Direct asymmetric vinylogous Michael addition of cyclic enones to nitroalkenes via dienamine catalysis.

Project description:In spite of the many catalytic methodologies available for the asymmetric functionalization of carbonyl compounds at their ? and ? positions, little progress has been achieved in the enantioselective carbon-carbon bond formation ? to a carbonyl group. Here, we show that primary amine catalysis provides an efficient way to address this synthetic issue, promoting vinylogous nucleophilicity upon selective activation of unmodified cyclic ?,?-unsaturated ketones. Specifically, we document the development of the unprecedented direct and vinylogous Michael addition of ?-substituted cyclohexenone derivatives to nitroalkenes proceeding under dienamine catalysis. Besides enforcing high levels of diastereo- and enantioselectivity, chiral primary amine catalysts derived from natural cinchona alkaloids ensure complete ?-site selectivity: The resulting, highly functionalized vinylogous Michael adducts, having two stereocenters at the ? and ? positions, are synthesized with very high fidelity. Finally, we describe the extension of the dienamine catalysis-induced vinylogous nucleophilicity to the asymmetric ?-amination of cyclohexene carbaldehyde.

Enantio- and diastereoselective diarylmethylation of 1,3-dicarbonyl compounds.

Project description:A Cu-catalyzed enantio- and diastereoselective diarylmethylation of 1,3-dicarbonyl compounds is described. It is a successful example of constructing all-carbon quaternary stereocenters from 3 °C-H nucleophiles, a challenging topic in synthetic chemistry. In the present work, two contiguous stereocenters are constructed with high levels of stereoselectivity and atom economy. The broad scope of 1,3-dicarbonyl nucleophiles and the tolerance of a wide range of functional groups make this protocol of great importance in the synthesis of chiral diarylmethane compounds.

Reaction Mechanism of Organocatalytic Michael Addition of Nitromethane to Cinnamaldehyde: A Case Study on Catalyst Regeneration and Solvent Effects.

Project description:The Michael addition of nitromethane to cinnamaldehyde has been computationally studied in the absence of a catalyst and the presence of a biotinylated secondary amine by a combined computational and experimental approach. The calculations were performed at the density functional theory (DFT) level with the M06-2X hybrid functional, and a polarizable continuum model has been employed to mimic the effect of two different solvents: dichloromethane (DCM) and water. Contrary to common assumption, the product-derived iminium intermediate was absent in both of the solvents tested. Instead, hydrating the C1-C2 double bond in the enamine intermediate directly yields the tetrahedral intermediate, which is key for forming the product and regenerating the catalyst. Enamine hydration is concerted and found to be rate-limiting in DCM but segregated into two non-rate-limiting steps when the solvent is replaced with water. However, further analysis revealed that the use of water as solvent also raises the energy barriers for other chemical steps, particularly the critical step of C-C bond formation between the iminium intermediate and nucleophile; this consequently lowers both the reaction yield and enantioselectivity of this LUMO-lowering reaction, as experimentally detected. These findings provide a logical explanation to why water often enhances organocatalysis when used as an additive but hampers the reaction progress when employed as a solvent.

Project description:The highly enantioselective Michael addition of 2-hydroxy-1,4-naphthoquinones to nitroalkenes, promoted by binaphthyl-modified chiral bifunctional organocatalysts is described. This reaction afforded the chiral functionalized naphthoquinones in high yields (81-95%) and excellent enantioselectivities (91-98% ee) under low catalyst loading (1 mol %).

Project description:An asymmetric thia-Michael addition of arylthiols to α,β-unsaturated carboxylic acids using a thiourea catalyst that bears arylboronic acid and tertiary amine moieties is reported. Both enantiomers of the Michael adducts can be obtained in high enantioselectivity and good yield merely by changing the solvent. The origin of the chirality switch in the products was examined in each solvent via spectroscopic analyses.

Project description:The Mn(III)-initiated aerobic oxidation of heterocyclic 1,3-dicarbonyl compounds, such as 4-alkyl-1,2-diphenylpyrazolidine-3,5-diones, 1,3-dialkylpyrrolidine-2,4-diones, 3-alkyl-1,5-dimethylbarbituric acids, and 3-butyl-4-hydroxy-2-quinolinone gave excellent to good yields of the corresponding hydroperoxides, which were gradually degraded by exposure to the metal initiator after the reaction to afford the corresponding alcohols. The synthesis of 30 heterocyclic 1,3-dicarbonyl compounds, the corresponding hydroperoxides and the 10 alcohols, their characterization, and the limitations of the procedure are described. In addition, the mechanism of the hydroperoxidation and the redox decomposition of the hydroperoxides are discussed.
