ABSTRACT: We conducted a serologic investigation of porcine reproductive and respiratory syndrome virus (PRRSV) in hybrid wild boar herds in China during 2008-2009. PRRSV isolates with novel genetic markers were recovered. Experimental infection of pigs indicated that hybrid wild boars are involved in the epidemiology of PRRSV.

NADC30-Like Porcine Reproductive and Respiratory Syndrome in China.

Project description:NADC30-like porcine reproductive and respiratory syndrome virus (PRRSV) has widely spread in China and become locally dominant virus strain in some provinces. Although they are not pathogenic as highly pathogenic PRRSV (HP-RRRSV) that outbreaks since 2006, NADC30-like PRRSVs distinguished themselves by high incidence of recombination with other virus strains which lead to change of virulence. The outbreaks of NADC30-like PRRSV in the vaccinated pig herds suggested that current commercial PRRSV vaccines cannot provide complete protection to the infection. In this review, we have described in detail the current situation of NADC30 PRRSV including epidemiology, genomic characterization, pathogenicity, and efficacy of current commercial vaccines in China.

Virological and Histopathological Findings in Boars Naturally Infected With Porcine Reproductive and Respiratory Syndrome Virus Type 1.

Project description:Major geographical transmission of porcine reproductive and respiratory syndrome virus (PRRSV) occurs via semen when a boar stud is infected. This happened in Denmark in 2019, providing an opportunity to compare previous experimental PRRSV boar studies with natural PRRSV-1 infection in boars. The aim of this study was to investigate the association between the presence of PRRSV RNA in serum, semen, testicles, and epididymis of boars naturally infected with PRRSV and to describe the histological lesions in the testes and epididymis combined with direct visualisation of PRRSV-infected cells by immunohistochemical staining (IHC). The exact timing of infection of each boar was not determined, but based on serology the boars were divided into two groups: acute and late infections. All boars included were sampled the same day. In this study, 35 boars and 10 healthy boars from another PRRSV-negative boar stud were included as histological controls. PRRSV RNA was found most often in serum (51%) and least frequently in semen (22%) and was more often detected in the reproductive tract in the acute phase of infection (p < 0.0001; RR: 2.58). Mononuclear cells and multinuclear giant cells were present in the adluminal compartment of the testis and epididymis in PRRSV-infected boars, but not in control boars (p < 0.05), which supports the hypothesis that macrophages are involved in the venereal spread of the virus.

Emergence of Different Recombinant Porcine Reproductive and Respiratory Syndrome Viruses, China.

Project description:Epidemiological investigations were conducted on recently emerging porcine reproductive and respiratory syndrome virus (PRRSV) strains in Shandong province in 2014-2015. The proportion of the NADC30 strain identified by ORF7 sequence alignment has been gradually increasing. Three emerging PRRSV strains were successfully isolated, and the complete genomic sequences were determined. Our results indicate the importance of recombinant strains in Shandong province, China. There was a varied degree of recombination of two or three strains (classical, HP-PRRSV and/or NADC30). Moreover, the recombination strains affected the pathogenicity of newly emerged strains.

A new recombined porcine reproductive and respiratory syndrome virus virulent strain in China.

Project description:Porcine reproductive and respiratory syndrome (PRRS) is one of the most important swine diseases worldwide. In the present study, a new virulent strain of PRRS virus (PRRSV), GDsg, was isolated in Guangdong province, China, and caused high fever, high morbidity, and high mortality in sows and piglets. The genome of this new strain was 15,413 nucleotides (nt) long, and comparative analysis revealed that GDsg shared 82.4% to 94% identity with type 2 PRRSV strains, but only 61.5% identity with type 1 PRRSV Lelystad virus strain. Phylogenetic analysis indicated that type 2 PRRSV isolates include five subgenotypes (I, II, III, IV, and V), which are represented by NADC30, VR-2332, GM2, CH-1a, and HuN4, respectively. Moreover, GDsg belongs to a newly emerging type 2 PRRSV subgenotype III. More interestingly, the newly isolated GDsg strain has multiple discontinuous nt deletions, 131 (19 + 18 + 94) at position 1404-1540 and a 107 nt insertion in the NSP2 region. Most importantly, the GDsg strain was identified as a virus recombined between low pathogenic field strain QYYZ and vaccine strain JXA1-P80. In conclusion, a new independent subgenotype and recombinant PRRSV strain has emerged in China and could be a new threat to the swine industry of China.

Outbreaks of highly pathogenic porcine reproductive and respiratory syndrome in Jiangxi province, China.

Project description:In 2007, herds of pigs in Jiangxi Province, China experienced outbreaks of a severe form of suspected porcine reproductive and respiratory syndrome (PRRS) characterized by high fever, high morbidity and mortality in animals of different ages. 152 swine sera and 42 tissues (consisting of liver, lung, lymph node and kidney) from five herds of pigs were collected. Pigs were diagnosed as infected with a highly pathogenic form of the PRRS virus (PRRSV) based on ELISA and reverse transcriptase polymerase chain reaction (RT-PCR) results. Serological surveys indicated that 67-100% of the examined pig herds in Jiangxi Province were seropositive. 42 tissue samples were used to detect classical swine fever virus, porcine circovirus type 2 and PRRSV. Results indicated that only PRRSV was detected in 42 samples. 12 PRRSV amplified products of five herds, which consisted of two or three samples randomly selected from each herd, were used for sequencing. Subsequent nucleotide sequencing showed that the NSP2 gene had 99-99.7% nucleotide and 99.2-100% derived amino acid sequence identities among 12 tissues with that of the PRRS-JXA1 strain, deletions of 29 amino acids corresponded to positions 534-562 of the NSP2 gene sequence. These results revealed that the diseased pigs were all caused by fatal PRRSV variant. Compared with the same period in 2006, the number of positive cases from Jiangxi Province remained unchanged. These findings demonstrated that the highly pathogenic Northern American type PRRSV was still spreading in Jiangxi Province, China in 2007.

Porcine teschovirus in wild boars in Hungary.

Project description:The genus Teschovirus, family Picornaviridae, currently includes 12 serotypes (PTV 1 to 12) isolated from swine. PTVs have been well studied in domestic pigs, but knowledge about PTVs in wild boars is deficient. Here, we report the first complete PTV genome sequence from 7 (70 %) of 10 fecal samples of wild boar piglets (Sus scrofa) by RT-PCR and pyrosequencing. Analysis of the wild boar PTV strain WB2C-TV/2011/HUN (JQ429405) showed considerable difference, especially in VP1 (66-74 % amino acid identity) compared to the available PTVs. PTV is present in wild boars, and WB2C-TV/2011/HUN represents a novel PTV genotype, provisionally named PTV-13.

Porcine kobuvirus in wild boars (Sus scrofa).

Project description:Fecal samples (N = 10) from 6- to 8-week-old wild boar piglets (Sus scrofa), collected from an animal park in Hungary in April 2011, were analyzed using viral metagenomics and complete genome sequencing. Kobuvirus (genus Kobuvirus, family Picornaviridae) was detected in all (100 %) specimens, with the closest nucleotide (89 %) and amino acid (94 %) sequence identity of the strain wild boar/WB1-HUN/2011/HUN (JX177612) to the prototype porcine kobuvirus S-1-HUN (EU787450). This study suggests that genetically highly similar (practically the same geno-/serotype) porcine kobuvirus circulate in wild boars, the wildlife counterparts of domestic pigs. Wild boars could be an important host and reservoir for kobuvirus.
