Coding of the reach vector in parietal area 5d.

ABSTRACT: Competing models of sensorimotor computation predict different topological constraints in the brain. Some models propose population coding of particular reference frames in anatomically distinct nodes, whereas others require no such dedicated subpopulations and instead predict that regions will simultaneously code in multiple, intermediate, reference frames. Current empirical evidence is conflicting, partly due to difficulties involved in identifying underlying reference frames. Here, we independently varied the locations of hand, gaze, and target over many positions while recording from the dorsal aspect of parietal area 5. We find that the target is represented in a predominantly hand-centered reference frame here, contrasting with the relative code seen in dorsal premotor cortex and the mostly gaze-centered reference frame in the parietal reach region. This supports the hypothesis that different nodes of the sensorimotor circuit contain distinct and systematic representations, and this constrains the types of computational model that are neurobiologically relevant.

Parietal reach region encodes reach depth using retinal disparity and vergence angle signals.

Project description:Performing a visually guided reach requires the ability to perceive the egocentric distance of a target in three-dimensional space. Previous studies have shown that the parietal reach region (PRR) encodes the two-dimensional location of frontoparallel targets in an eye-centered reference frame. To investigate how a reach target is represented in three dimensions, we recorded the spiking activity of PRR neurons from two rhesus macaques trained to fixate and perform memory reaches to targets at different depths. Reach and fixation targets were configured to explore whether neural activity directly reflects egocentric distance as the amplitude of the required motor command, which is the absolute depth of the target, or rather the relative depth of the target with reference to fixation depth. We show that planning activity in PRR represents the depth of the reach target as a function of disparity and fixation depth, the spatial parameters important for encoding the depth of a reach goal in an eye centered reference frame. The strength of modulation by disparity is maintained across fixation depth. Fixation depth gain modulates disparity tuning while preserving the location of peak tuning features in PRR neurons. The results show that individual PRR neurons code depth with respect to the fixation point, that is, in eye centered coordinates. However, because the activity is gain modulated by vergence angle, the absolute depth can be decoded from the population activity.

Decoding Movement Goals from the Fronto-Parietal Reach Network.

Project description:During reach planning, fronto-parietal brain areas need to transform sensory information into a motor code. It is debated whether these areas maintain a sensory representation of the visual cue or a motor representation of the upcoming movement goal. Here, we present results from a delayed pro-/anti-reach task which allowed for dissociating the position of the visual cue from the reach goal. In this task, the visual cue was combined with a context rule (pro vs. anti) to infer the movement goal. Different levels of movement goal specification during the delay were obtained by presenting the context rule either before the delay together with the visual cue (specified movement goal) or after the delay (underspecified movement goal). By applying functional magnetic resonance imaging (fMRI) multivoxel pattern analysis (MVPA), we demonstrate movement goal encoding in the left dorsal premotor cortex (PMd) and bilateral superior parietal lobule (SPL) when the reach goal is specified. This suggests that fronto-parietal reach regions (PRRs) maintain a prospective motor code during reach planning. When the reach goal is underspecified, only area PMd but not SPL represents the visual cue position indicating an incomplete state of sensorimotor integration. Moreover, this result suggests a potential role of PMd in movement goal selection.

Extrastriate area MST and parietal area VIP similarly represent forward headings.

Project description:Many studies have documented the involvement of medial superior temporal extrastriate area (MST) in the perception of heading based on optic flow information. Furthermore, both heading perception and the responses of MST neurons are relatively stable in the presence of eye movements that distort the retinal flow information on which perception is based. Area VIP in the posterior parietal cortex also contains a robust representation of optic flow cues for heading. However, the studies in the two areas were frequently conducted using different stimuli, making quantitative comparison difficult. To remedy this, we studied MST using a family of random dot heading stimuli that we have previously used in the study of VIP. These stimuli simulate observer translation through a three-dimensional cloud of points, and a range of forward headings was presented both with and without horizontal smooth pursuit eye movements. We found that MST neurons, like VIP neurons, respond robustly to these stimuli and partially compensate for the presence of pursuit. Quantitative comparison of the responses revealed no substantial difference between the heading responses of MST and VIP neurons or in their degree of pursuit tolerance.

Human posterior parietal cortex plans where to reach and what to avoid.

Project description:In this time-resolved functional magnetic resonance imaging (fMRI) study, we aimed to trace the neuronal correlates of covert planning processes that precede visually guided motor behavior. Specifically, we asked whether human posterior parietal cortex has prospective planning activity that can be distinguished from activity related to retrospective visual memory and attention. Although various electrophysiological studies in monkeys have demonstrated such motor planning at the level of parietal neurons, comparatively little support is provided by recent human imaging experiments. Rather, a majority of experiments highlights a role of human posterior parietal cortex in visual working memory and attention. We thus sought to establish a clear separation of visual memory and attention from processes related to the planning of goal-directed motor behaviors. To this end, we compared delayed-response tasks with identical mnemonic and attentional demands but varying degrees of motor planning. Subjects memorized multiple target locations, and in a random subset of trials targets additionally instructed (1) desired goals or (2) undesired goals for upcoming finger reaches. Compared with the memory/attention-only conditions, both latter situations led to a specific increase of preparatory fMRI activity in posterior parietal and dorsal premotor cortex. Thus, posterior parietal cortex has prospective plans for upcoming behaviors while considering both types of targets relevant for action: those to be acquired and those to be avoided.

Local field potentials in the parietal reach region reveal mechanisms of bimanual coordination.

Project description:Primates use their arms in complex ways that frequently require coordination between the two arms. Yet the planning of bimanual movements has not been well-studied. We recorded spikes and local field potentials (LFP) from the parietal reach region (PRR) in both hemispheres simultaneously while monkeys planned and executed unimanual and bimanual reaches. From analyses of interhemispheric LFP-LFP and spike-LFP coherence, we found that task-specific information is shared across hemispheres in a frequency-specific manner. This shared information could arise from common input or from direct communication. The population average unit activity in PRR, representing PRR output, encodes only planned contralateral arm movements while beta-band LFP power, a putative PRR input, reflects the pattern of planned bimanual movement. A parsimonious interpretation of these data is that PRR integrates information about the movement of the left and right limbs, perhaps in service of bimanual coordination.

Motor planning poststroke: impairment in vector-coded reach plans.

Project description:Healthy individuals appear to use both vector-coded reach plans that encode movements in terms of their desired direction and extent, and target-coded reach plans that encode the desired endpoint position of the effector. We examined whether these vector and target reach-planning codes are differentially affected after stroke. Participants with stroke and healthy controls made blocks of reaches that were grouped by target location (providing target-specific practice) and by movement vector (providing vector-specific practice). Reach accuracy was impaired in the more affected arm after stroke, but not distinguishable for target- versus vector-grouped reaches. Reach velocity and acceleration were not only impaired in both the less and more affected arms poststroke, but also not distinguishable for target- versus vector-grouped reaches. As previously reported in controls, target-grouped reaches yielded isotropic (circular) error distributions and vector-grouped reaches yielded error distributions elongated in the direction of the reach. In stroke, the pattern of variability was similar. However, the more affected arm showed less elongated error ellipses for vector-grouped reaches compared to the less affected arm, particularly in individuals with right-hemispheric stroke. The results suggest greater impairment to the vector-coded movement-planning system after stroke, and have implications for the development of personalized approaches to poststroke rehabilitation: Motor learning may be enhanced by practice that uses the preserved code or, conversely, by retraining the more impaired code to restore function.

Inactivation of the parietal reach region causes optic ataxia, impairing reaches but not saccades.

Project description:Lesions in human posterior parietal cortex can cause optic ataxia (OA), in which reaches but not saccades to visual objects are impaired, suggesting separate visuomotor pathways for the two effectors. In monkeys, one potentially crucial area for reach control is the parietal reach region (PRR), in which neurons respond preferentially during reach planning as compared to saccade planning. However, direct causal evidence linking the monkey PRR to the deficits observed in OA is missing. We thus inactivated part of the macaque PRR, in the medial wall of the intraparietal sulcus, and produced the hallmarks of OA, misreaching for peripheral targets but unimpaired saccades. Furthermore, reach errors were larger for the targets preferred by the neural population local to the injection site. These results demonstrate that PRR is causally involved in reach-specific visuomotor pathways, and reach goal disruption in PRR can be a neural basis of OA.

Distinct contributions of human posterior parietal and dorsal premotor cortex to reach trajectory planning.

Project description:Goal-directed hand movements are usually directed straight at the target, e.g. when swatting a fly. Their paths can also become quite complex, when drawing or avoiding obstacles. Studies on movement planning have largely neglected the latter movement type and the question of whether it is the same neural machinery that is planning such complex hand trajectories as well as straight, vector-like movements. Using time-resolved fMRI during delayed response tasks we examined planning activity in human superior parietal lobule (SPL) and dorsal premotor cortex (PMd). We show that the recruitment of both areas in trajectory planning differs significantly: PMd represented both straight and complex hand trajectories while SPL only those that led straight to the target. This suggests that while posterior parietal cortex only provides representations for simple, straight reaches, the complex and computationally demanding reach planning necessarily involves dorsal premotor cortex. Our findings yield new insights into the organization of cerebro-cortical strategies of forming reach trajectory plans.

The posterior parietal cortex encodes in parallel both goals for double-reach sequences.

Project description:The parietal reach region (PRR) is known to be involved in the preparation of visually guided arm movements to single targets. We explored whether PRR encodes only the target of the next movement or, alternatively, also a subsequent goal in a double-reach sequence. Two monkeys were trained to memorize the locations of two peripheral cues and to prepare for a memory-guided delayed double-reach sequence. On a GO-signal they had to reach in a predefined order to both remembered target locations without breaking eye fixation. The movement goals were arranged such that either the first or the second target was inside the response field of an isolated neuron. We analyzed the neural activity of single cells in PRR during the late memory period between cue offset and the GO-signal. During this memory period, most PRR cells encoded the first as well as the second goal of the planned reaching sequence. The results indicate that the posterior parietal cortex is involved in the spatial planning of more complex action patterns and represents immediate and subsequent movement goals.

Coding of interceptive saccades in parietal cortex of macaque monkeys.

Project description:The oculomotor system can initiate remarkably accurate saccades towards moving targets (interceptive saccades) the processing of which is still under debate. The generation of these saccades requires the oculomotor centers to have information about the motion parameters of the target that then must be extrapolated to bridge the inherent processing delays. We investigated to what degree the information about motion of a saccade target is available in the lateral intra-parietal area (area LIP) of macaque monkeys for generation of accurate interceptive saccades. When a multi-layer neural network was trained based on neural discharges from area LIP around the time of saccades towards stationary targets, it was also able to predict the end points of saccades directed towards moving targets. This prediction, however, lagged behind the actual post-saccadic position of the moving target by ~ 80 ms when the whole neuronal sample of 105 neurons was used. We further found that single neurons differentially code for the motion of the target. Selecting neurons with the strongest representation of target motion reduced this lag to ~ 30 ms which represents the position of the moving target approximately at the onset of the interceptive saccade. We conclude that-similarly to recent findings from the Superior Colliculus (Goffart et al. J Neurophysiol 118(5):2890-2901)-there is a continuum of contributions of individual LIP neurons to the accuracy of interceptive saccades. A contribution of other gaze control centers (like the cerebellum or the frontal eye field) that further increase the saccadic accuracy is, however, likely.
