The biological effects of energetic heavy ions are attracting increasing interest for their applications in cancer therapy and protection against space radiation. The cascade of events leading to cell death or late effects starts from stochastic energy deposition on the nanometer scale and the corresponding lesions in biological molecules, primarily DNA. Project description:Recent advances in native mass spectrometry have enabled its use to probe the structure of and interactions within biomolecular complexes. Surface-induced dissociation, in which inter- and intramolecular interactions are disrupted following an energetic ion-surface collision, is a method that can directly interrogate the topology of protein complexes. However, a quantitative relationship between the ion kinetic energy at the moment of surface collision and the internal energy deposited into the ion has not yet been established for proteins. The factors affecting energy deposition in surface-induced unfolding (SIU) of protein monomers were investigated and a calibration relating laboratory-frame kinetic energy to internal energy developed. Protein monomers were unfolded by SIU and by collision-induced unfolding (CIU). CIU and SIU cause proteins to undergo the same unfolding transitions at different values of laboratory-frame kinetic energy. There is a strong correlation between the SIU and CIU energies, demonstrating that SIU, like CIU, can largely be understood as a thermal process. The change in internal energy in CIU was modeled using a Monte Carlo approach and theory. Computed values of the overall efficiency were found to be approximately 25% and used to rescale the CIU energy axis and relate nominal SIU energies to internal energy. The energy deposition efficiency in SIU increases with mass and kinetic energy from a low of ∼20% to a high of ∼68%, indicating that the effective mass of the surface increases along with the mass of the ion. The effect of ion structure on energy deposition was probed using multiple stages of ion activation. Energy deposition in SIU strongly depends on structure, decreasing as the protein is elongated, due to decreased effective protein-surface collisional cross section and increased transfer to rotational modes.

Project description:In this work, chitosan bead materials were modified by cross-linking with epichlorohydrin (EP) and glutaraldehyde (GA) for the removal of heavy metals in wastewater. Using these cross-linked chitosan materials, the dependence of adsorption of chromate anions on pH was investigated experimentally and theoretically. The experimental results show that the adsorption process of the chromate (Cr) ions greatly depends on the pH of the solution, with the chitosan modified by cross-linking being an efficient adsorbent for chromate. On the other hand, quantum chemistry calculations were conducted to find out the factor determining the pH dependence of the adsorption efficiency of chromate ions on the dimer chitosan molecule, and show results similar to those found in the experiment. Both the experimental and numerical results show that the total charge numbers of the adsorbent and the adsorbate species and their relative molecular geometries are crucial in determining the adsorption efficiency.

Project description:Kaolinite can undergo a controlled morphological modification process into exfoliated nanosilicate sheets (EXK) and silicate nanotubes (KNTs). The modified structures were assessed as potential effective adsorbents for the retention of Cs+ ions. The impact of the modification process on the retention properties was assessed based on conventional and advanced equilibrium studies, considering the related steric and energetic functions. The synthetic KNTs exhibit a retention capacity of 249.7 mg g-1 as compared to EXK (199.8 mg g-1), which is significantly higher than raw kaolinite (73.8 mg g-1). The kinetic modeling demonstrates the high effectiveness of the pseudo-first-order kinetic model (R2 > 0.9) to illustrate the sequestration reactions of Cs+ ions by K, EXK, and KNTs. The enhancement effect of the modification processes can be illustrated based on the statistical investigations. The presence of active and vacant receptors enhanced greatly from 19.4 mg g-1 for KA to 40.8 mg g-1 for EXK and 46.9 mg g-1 for KNTs at 298 K. This validates the significant impact of the modification procedures on the specific surface area, reaction interface, and reacting chemical groups' exposure. This also appeared in the enhancement of the reactivity of their surfaces to be able to uptake 10 Cs+ ions by KNTs and 5 ions by EXK as compared to 4 ions by kaolinite. The thermodynamic and energetic parameters (Gaussian energy < 8.6 kJ mol-1; uptake energy < 40 kJ mol-1) show that the physical processes are dominant, which have spontaneous and exothermic properties. The synthetic EXK and KNT structures validate the high elimination performance of the retention of Cs+ either in the existence of additional anions or cations.

Project description:This work reports that the octahedral hydrated Al3+ and Mg2+ ions operate within electrolytic cells as kosmotropic (long-range order-making) "ice makers" of supercooled water (SCW). 10-5 M solutions of hydrated Al3+ and Mg2+ ions each trigger, near the cathode (-20 ± 5 V), electro-freezing of SCW at -4 °C. The hydrated Al3+ ions do so with 100% efficiency, whereas the Mg2+ ions induce icing with 40% efficiency. In contrast, hydrated Na+ ions, under the same experimental conditions, do not induce icing differently than pure water. As such, our study shows that the role played by Al3+ and Mg2+ ions in water electro-freezing is impacted by two synchronous effects: (1) a geometric effect due to the octahedral packing of the coordinated water molecules around the metallic ions, and (2) the degree of polarization which these two ions induce and thereby acidify the coordinated water molecules, which in turn imparts them with an ice-like structure. Long-duration molecular dynamics (MD) simulations of the Al3+ and Mg2+ indeed reveal the formation of "ice-like" hexagons in the vicinity of these ions. Furthermore, the MD shows that these hexagons and the electric fields of the coordinate water molecules give rise to ultimate icing. As such, the MD simulations provide a rational explanation for the order-making properties of these ions during electro-freezing.

Project description:Over the years, a large number of sorption experiments using the aminopolycarboxylic acid (APCA)-functionalized adsorbents were carried out in batch conditions, but prospective research should also be directed towards column studies to check their industrial/commercial feasibility. In this context, sorption studies of five-component heavy metal ion (HMI) solutions containing Zn2+, Pb2+, Cd2+, Ni2+, and Co2+ in equimolar concentrations were assessed in fixed-bed columns using some APCA-functionalized chitosan-clinoptilolite (CS-CPL) cryogel sorbents in comparison to unmodified composite materials. The overall sorption tendency of the APCA-functionalized composite sorbents followed the sequence Co2+ < Zn2+ < Cd2+ ≤ Pb2+ < Ni2+, meaning that Co2+ ions had the lowest affinity for the sorbent’s functional groups, whereas the Ni2+ ions were strongly and preferentially adsorbed. To get more insights into the application of the composite microbeads into continuous flow set-up, the kinetic data were described by Thomas and Yoon−Nelson models. A maximum theoretical HMI sorption capacity of 145.55 mg/g and a 50% breakthrough time of 121.5 min were estimated for the column containing CSEDTA-CPL cryogel sorbents; both values were much higher than those obtained for the column filled with pristine CS-CPL sorbents. In addition, desorption of HMIs from the composite microbeads in dynamic conditions was successfully achieved using 0.1 M HCl aqueous solution. Moreover, a theoretical analysis of APCA structures attached to composite adsorbents and their spatial structures within the complex combinations with transition metals was systematically performed. Starting from the most stable conformer of EDTA, coordinative combinations with HMIs can be obtained with an energy consumption of only 1 kcal/mole, which is enough to shift the spatial structure into a favorable conformation for HMI chelation.

Project description:Fundamental understanding of the interactions of nanoscale materials with molecules of interest is essential for the development of electronic devices, such as sensors. In particular, structures and molecular interaction properties of engineered graphenes are still largely unexplored, despite these materials' great potential to be used as molecular sensors. As an example of end user application, the detection of phosphorus in the form of phosphate in a soil environment is important for soil fertility and plant growth. However, due to the lack of an affordable technology, it is currently hard to measure the amount of phosphate directly in the soil; therefore, suitable sensor technologies need to be developed for phosphate sensors. In this work, pristine graphene and several modified graphene materials (oxygenated graphene, graphene with vacancies, and curved graphene) were studied as candidates for phosphate sensor materials using density functional theory (DFT) calculations. Our calculations showed that both pristine graphene and functionalized graphene were able to adsorb phosphate species strongly. In addition, these graphene nanomaterials showed selectivity of adsorption of phosphate with respect to nitrate, with stronger adsorption energies for phosphate. Furthermore, our calculations showed significant changes in electrical conductivities of pristine graphene and functionalized graphenes after phosphate species adsorption, in particular, on graphene with oxygen (hydroxyl and epoxide) functional groups. Experimental measurements of electrical resistivity of graphene before and after adsorption of dihydrogen phosphate showed an increase in resistivity upon adsorption of phosphate, consistent with the theoretical predictions. Our results recommend graphene and functionalized graphene-based nanomaterials as good candidates for the development of phosphate sensors.

Project description:Herein, we show a comprehensive experimental, theoretical, and computational study aimed at designing macromolecules able to adsorb a cargo at the nanoscale. Specifically, we focus on the adsorption properties of star diblock copolymers, i.e., macromolecules made by a number f of H-T diblock copolymer arms tethered on a central core; the H monomeric heads, which are closer to the tethering point, are attractive toward a specific target, while the T monomeric tails are neutral to the cargo. Experimentally, we exploited the adaptability of poly(2-oxazoline)s (POxs) to realize block copolymer-coated nanoparticles with a proper functionalization able to interact with heavy metals and show or exhibit a thermoresponsive behavior in aqueous solution. We here present the synthesis and analysis of the properties of a high molecular mass block copolymer featured by (i) a polar side chain, capable of exploiting electrostatic and hydrophilic interaction with a predetermined cargo, and (ii) a thermoresponsive scaffold, able to change the interaction with the media by tuning the temperature. Afterward, the obtained polymers were grafted onto iron oxide nanoparticles and the thermoresponsive properties were investigated. Through isothermal titration calorimetry, we then analyzed the adsorption properties of the synthesized superparamagnetic nanoparticles for heavy metal ions in aqueous solution. Additionally, we use a combination of scaling theories and simulations to link equilibrium properties of the system to a prediction of the loading properties as a function of size ratio and effective interactions between the considered species. The comparison between experimental results on adsorption and theoretical prediction validates the whole design process.

Project description:PurposeSide effects related to radiation exposures are based primarily on the assumption that the detrimental effects of radiation occur in directly irradiated cells. However, several studies have reported over the years of radiation-induced non-targeted/ abscopal effects in vivo that challenge this paradigm. There is evidence that Cyclooxygenase-2 (COX2) plays an important role in modulating non-targeted effects, including DNA damages in vitro and mutagenesis in vivo. While most reports on radiation-induced non-targeted response utilize x-rays, there is little information available for heavy ions.Methods and materialsAdult female transgenic gpt delta mice were exposed to an equitoxic dose of either carbon or argon particles using the Heavy Ion Medical Accelerator in Chiba (HIMAC) at the National Institute of Radiological Sciences (NIRS) in Japan. The mice were stratified into 4 groups of 5 animals each: Control; animals irradiated under full shielding (Sham-irradiated); animals receiving whole body irradiation (WBIR); and animals receiving partial body irradiation (PBIR) to the lower abdomen with a 1 x 1 cm2 field. The doses used in the carbon ion group (4.5 Gy) and in argon particle group (1.5 Gy) have a relative biological effectiveness equivalent to a 5 Gy dose of x-rays. 24 hours after irradiation, breast tissues in and out of the irradiated field were harvested for analysis. Induction of COX2, 8-hydroxydeoxyguanosine (8-OHdG), phosphorylated histone H2AX (γ-H2AX), and apoptosis-related cysteine protease-3 (Caspase-3) antibodies were examined in the four categories of breast tissues using immunohistochemical techniques. Analysis was performed by measuring the intensity of more than 20 individual microscopic fields and comparing the relative fold difference.ResultsIn the carbon ion group, the relative fold increase in COX2 expression was 1.01 in sham-irradiated group (p > 0.05), 3.07 in PBIR (p < 0.05) and 2.50 in WBIR (p < 0.05), respectively, when compared with controls. The relative fold increase in 8-OHdG expression was 1.29 in sham-irradiated (p > 0.05), 11.31 in PBIR (p < 0.05) and 11.79 in WBIR (p < 0.05), respectively, when compared with controls. A similar increase in γ-H2AX expression was found in that, compared to controls, the increase was 1.41 fold in sham-irradiated (p > 0.05), 8.41 in PBIR (p < 0.05) and 10.59 in WBIR (p < 0.05). Results for the argon particle therapy group showed a similar magnitude of changes in the various biological endpoints examined. There was no statistical significance observed in Caspase-3 expression among the 4 groups.ConclusionsOur data show that both carbon and argon ions induced non-targeted, out of field induction of COX2 and DNA damages in breast tissues. These effects may pose new challenges to evaluate the risks associated with radiation exposure and understanding radiation-induced side effects.

Project description:Electrochemical immunosensors (EIs) integrate biorecognition molecules (e.g., antibodies) with redox enzymes (e.g., horseradish peroxidase) to combine the advantages of immunoassays (high sensitivity and selectivity) with those of electrochemical biosensors (quantitative electrical signal). However, the complex network of mass-transfer, catalysis, and electrochemical reaction steps that produce the electrical signal makes the design and optimization of EI systems challenging. This paper presents an integrated experimental and modeling framework to address this challenge. The framework includes (1) a mechanistic mathematical model that describes the rate of key mass-transfer and reaction steps; (2) a statistical-design-of-experiments study to optimize operating conditions and validate the mechanistic model; and (3) a novel dimensional analysis to assess the degree to which individual mass-transfer and reaction steps limit the EI's signal amplitude and sensitivity. The validated mechanistic model was able to predict the effect of four independent variables (working electrode overpotential, pH, and concentrations of catechol and hydrogen peroxide) on the EI's signal magnitude. The model was then used to calculate dimensionless groups, including Damkohler numbers, novel current-control coefficients, and sensitivity-control coefficients that indicated the extent to which the individual mass-transfer or reaction steps limited the EI's signal amplitude and sensitivity.

Project description:The presence of dyes and heavy metals in water sources as pollutants is harmful to human and animal health. Therefore, this study aimed to evaluate the efficacy of zinc ferrite (ZnFe2O4) nanoparticles (ZF-NPs) due to their outstanding properties including cost-effectiveness, availability, and applicability for removal of auramine O (AO), methylene blue (MB), and Cd (II). The effect of the main operating parameters such as AO concentration, MB concentration, Cd (II) concentration, adsorbent amount, solution pH, and sonication time was optimized by the response surface methodology (RSM). Optimal conditions were obtained at adsorbent amount of 0.25 g, pH = 6, sonication time of 15 min, and concentration of 15 mg L-1, and more than 91.56% were removed from all three analytes. The adsorption of AO, MB, and Cd (II) onto ZF-NPs followed pseudo-second-order kinetics and the equilibrium data fitted well with Langmuir isotherm. The maximum adsorption capacities of ZF-NPs for AO, MB and Cd (II) were as high as 201.29 mg g-1, 256.76 mg g-1 and 152.48 mg g-1, respectively. Also, the reuse of the adsorbent was investigated, and it was found that the adsorbent can be used for up to five cycles. Based on the results of interference studies, it was found that different ions do not have a significant effect on the removal of AO, MB, and Cd (II) in optimal conditions. The ZF-NPs was investigated successfully to remove AO, MB, and Cd (II) from environmental water samples. The results of this study showed that ZF-NPs can be used as a suitable adsorbent to remove AO, MB, and Cd (II) from aqueous solution.
