Revision of the Lispe longicollis-group (Diptera, Muscidae).

ABSTRACT: The Lispe longicollis species-group is revised. Lispe ethiopicasp. n. is described. The following 3 new synonyms are established: Lispe assimilis Wiedemann, 1824 (syn: cyrtoneurina Stein, 1900 and modesta Stein, 1913); Lispe manicata Wiedemann, 1830 (syn: forficata Kurahashi & Shinonaga, 2009). Female of Lispe microptera Seguy, 1937 is described for the first time. Identification key for known Eurasian and African species is given.

Revision of the Merodon serrulatus group (Diptera, Syrphidae).

Project description:The phytophagous hoverfly genus Merodon Meigen, 1803 (Diptera, Syrphidae), which comprises more than 160 species distributed in Palaearctic and Afrotropical regions, can be differentiated into multiple groups of species that harbor high levels of hidden diversity. In this work, the serrulatus species group of Merodon is revised, providing an illustrated key to species, a detailed discussion on the taxonomic characters and a morphological diagnosis, including also the first data about the preimaginal morphology of this species group. The study includes characteristics of the 13 species of the M. serrulatus group, along with the available distributional data. Moreover, descriptions are provided for seven new species, namely M. defectus Vujić, Likov & Radenković sp. nov., M. disjunctus Vujić, Likov & Radenković sp. nov., M. medium Vujić, Likov & Radenković sp. nov., M. nigrocapillatus Vujić, Likov & Radenković sp. nov., M. nigropunctum Vujić, Likov & Radenković sp. nov., M. opacus Vujić, Likov & Radenković sp. nov., and M. trianguloculus Vujić, Likov & Radenković sp. nov. In addition, the taxa M. serrulatus (Wiedemann in Meigen, 1822), M. bequaerti Hurkmans, 1993, M. hirsutus Sack, 1913, M. kawamurae Matsumura, 1916, M. sacki (Paramonov, 1936) and M. sophron Hurkmans, 1993 are redefined and redescribed. Following a detailed study of the type material sourced from different entomological collections, the status of all available taxa related to M. serrulatus is revised and a new synonymy is proposed: M. tener Sack, 1913 syn. nov. (junior synonym of M. serrulatus). The identity of M. trizonus (Szilády, 1940) could not be assessed as the type specimens are lost. Thus, the name M. trizonus is considered as nomen dubium. The monophyly and composition of this species group are assessed through Maximum Parsimony and Maximum Likelihood analyses of the mitochondrial COI and nuclear 28S rRNA gene sequences.

Revision of the Exechiaparva group (Diptera: Mycetophilidae).

Project description:BackgroundExechia is a diverse genus of small fungus gnats, widespread in the Holarctic Region, while the fauna is largely unknown elsewhere, such as in the Afrotropical and Oriental Region. Members of Exechia can be arranged into several species groups, based on homologies in the male and female terminalia. The Exechiaparva group is delimited, based on male terminalia possessing a pair of gonocoxal lobes on the apicoventral gonocoxal margin. Eight previously-described species can be placed in this group, of which six are from the Holarctic Region, while one is recorded each from the Oriental and the Afrotropical Regions.New informationThe Exechiaparva group was reviewed and found to include 33 species, of which 24 were described as new to science and six were re-described. Identification keys to 32 species for males and nine species for females are provided together with illustrations and photos of male and female terminalia. Species delimitations were based on morphological examination of 94 male and female specimens, as well as DNA barcodes obtained from 124 specimens. Molecular and morphological species delimitations were mostly congruent, except in two cases where two species were delimited within a single Barcode Index Number (BIN). We found that each species is only known from a single zoogeographical region and that several species complexes are largely congruent with zoogeographical divisions, indicating that intercontinental barriers may have a strong impact on the species diversity of the group.

New records of Muscidae (Diptera) from Mediterranean countries.

Project description:New records are provided for Muscidae from four different Mediterranean countries, with new distribution records for species in ten different genera. Seven species are newly recorded for Croatia, four species for Montenegro and one species for Bosnia & Herzegovina. In this paper we give the first confirmation of an aquatic larval stage for Lispocephalabrachialis (Rondani, 1877), Lispocephalaspuria (Zetterstedt, 1838) and Lispocephalamikii (Strobl, 1893). A first record of the species Coenosialyneborgi Pont, 1972 since its original description is also provided.

Molecular Identification and Geometric Morphometric Analysis of Haematoboscaaberrans (Diptera: Muscidae).

Project description:The genus Haematobosca Bezzi, 1907 (Diptera: Muscidae) contains haematophagous flies of veterinary importance. A new fly species of this genus was recognised from northern Thailand based on morphological characters and described as Haematobosca aberrans Pont, Duvallet & Changbunjong, 2020. In the present study, the mitochondrial cytochrome c oxidase I (COI) gene was used to confirm the morphological identification of H. aberrans. In addition, landmark-based geometric morphometrics was used to determine sexual dimorphism. The molecular analysis was conducted with 10 COI sequences. The results showed that all sequences were 100% identical. The sequence was not highly similar to reference sequences from GenBank and did not match any identified species from Barcode of Life Data Systems (BOLD). Phylogenetic analysis clearly differentiated this species from other species within the subfamily Stomoxyinae. For geometric morphometric analysis, a total of 16 wing pictures were analysed using the landmark-based approach. The results showed significant differences in wing shape between males and females, with a cross-validated classification score of 100%. The allometric analysis showed that wing shape has no correlation with size. Therefore, the COI gene is effective in species identification of H. aberrans, and geometric morphometrics is also effective in determining sexual dimorphism.

Project description:The taxonomic reasons for regarding Lispe draperi Séguy, 1933, sp. rev., as a valid species instead of a synonym of Lispe tentaculata (De Geer, 1776) and for treating Lispe quaerens Villeneuve, 1936, syn. n., as a junior synonym of Lispe sericipalpis Stein, 1904 are given. A revised key for the Palaearctic members of the Lispe tentaculata species-group is given. Data on ecology, distribution and feeding preferences are provided.

Revision of the Palaearctic species of the Merodon desuturinus group (Diptera, Syrphidae).

Project description:This revision of material belonging to the Merodon desuturinus group from the Palaearctic Region resulted in the delimitation of four species: Merodon cabanerensis Marcos-García, Vujić & Mengual, 2007; Merodon desuturinus Vujić, Šimić & Radenković, 1995; Merodon neolydicus Vujić, nom. n.; and Merodon murorum Fabricius, 1794. Merodon murorum is redescribed. A neotype for Merodon auripilus Meigen, 1830 is designated, which is a new junior synonym of Merodon murorum. The related Afrotropical species Merodon cuthbertsoni Curran, 1939 is re-evaluated and compared to its sibling Palaearctic taxon Merodon desuturinus. An identification key for the Merodon desuturinus group is provided.

High chromosomal variation in wild horn fly Haematobiairritans (Linnaeus) (Diptera, Muscidae) populations.

Project description:The horn fly, Haematobiairritans is an obligate haematophagous cosmopolitan insect pest. The first reports of attacks on livestock by Haematobiairritans in Argentina and Uruguay occurred in 1991, and since 1993 it is considered an economically important pest. Knowledge on the genetic characteristics of the horn fly increases our understanding of the phenotypes resistant to insecticides that repeatedly develop in these insects. The karyotype of Haematobiairritans, as previously described using flies from an inbred colony, shows a chromosome complement of 2n=10 without heterochromosomes (sex chromosomes). In this study, we analyze for the first time the chromosome structure and variation of four wild populations of Haematobiairritans recently established in the Southern Cone of South America, collected in Argentina and Uruguay. In these wild type populations, we confirmed and characterized the previously published "standard" karyotype of 2n=10 without sex chromosomes; however, surprisingly a supernumerary element, called B-chromosome, was found in about half of mitotic preparations. The existence of statistically significant karyotypic diversity was demonstrated through the application of orcein staining, C-banding and H-banding. This study represents the first discovery and characterization of horn fly karyotypes with 2n=11 (2n=10+B). All spermatocytes analyzed showed 5 chromosome bivalents, and therefore, 2n=10 without an extra chromosome. Study of mitotic divisions showed that some chromosomal rearrangements affecting karyotype structure are maintained as polymorphisms, and multiple correspondence analyses demonstrated that genetic variation was not associated with geographic distribution. Because it was never observed during male meiosis, we hypothesize that B-chromosome is preferentially transmitted by females and that it might be related to sex determination.

The complete mitochondrial genome of Graphomya rufitibia (Diptera: Muscidae).

Project description:Graphomya rufitibia (Diptera: Muscidae) was distributed worldwide. In this study, the complete mitogenome of G. rufitibia was sequenced and annotated, and the full-length was a 15,374 bp fragment, consisting of A (40.64%), G (8.96%), T (36.80%), and C (13.59%). The mitogenome is composed of 13 protein-coding genes, 22 transfer RNA genes, two ribosomal RNA genes, and a non-coding AT-rich region. Most PCGs used the canonical putative codon to start and stop, 21 tRNAs are folded into the classic clover-leaf structure, with the one exception of tRNA-Ser (AGN). A total length of 11,182 bp encoding 3727 amino acids, and account for 72.7% of the whole mitogenome. Phylogenetic analyses showed that G. rufitibia belongs to family Mydaeinae.

Behavioral response of house flies (Diptera: Muscidae) to 3 bittering agents.

Project description:House flies, Musca domestica L., are pests at livestock facilities. House fly numbers are controlled mainly by manure management and chemical controls, like use of toxic baits. Some toxic baits contain denatonium benzoate as a bittering agent, to avoid poisoning of children. House fly behavioral response to denatonium benzoate was examined, using 2 different diluents, sucrose solution or orange juice. Response of flies to quinine and sucrose octaacetate, 2 other bittering agents, was also examined in sucrose. Sucrose is the sweetener in commercially available toxic house fly baits. The proboscis extension response of both male and female flies was significantly reduced by addition of 1,000 ppm of denatonium benzoate to sucrose. Response in males was also reduced at 100 ppm. Consumption of sucrose by both male and female flies was reduced by addition of 100, 500, and 1,000 ppm of denatonium benzoate, but consumption by males was also reduced at 10 ppm. Addition of sucrose octaacetate had no detectable effect on consumption of sucrose at 100, 500, or 1,000 ppm. In contrast, addition of quinine reduced consumption by both male and female flies in all tested concentrations. In orange juice, 10 ppm of denatonium benzoate is often avoided by children. With orange juice, fly proboscis extension response patterns were the same for both sexes, with a reduction at 1,000 ppm of denatonium benzoate. Orange juice consumption by flies was reduced significantly at 1,000 ppm of denatonium benzoate for females but not at any of the concentrations tested for males.
