Project description:Full plastome sequences for land plants have become readily accessible thanks to the development of Next Generation Sequencing (NGS) techniques and powerful bioinformatic tools. Despite this vast amount of genomic data, some lineages remain understudied. Full plastome sequences from the highly diverse (>1,500 spp.) subfamily Tillandsioideae (Bromeliaceae, Poales) have been published for only three (i.e., Guzmania, Tillandsia, and Vriesea) out of 22 currently recognized genera. Here, we focus on core Tillandsioideae, a clade within subfamily Tillandsioideae, and explore the contribution of individual plastid markers and data categories to inform deep divergences of a plastome phylogeny. We generated 37 high quality plastome assemblies and performed a comparative analysis in terms of plastome structure, size, gene content and order, GC content, as well as number and type of repeat motifs. Using the obtained phylogenetic context, we reconstructed the evolution of these plastome attributes and assessed if significant shifts on the evolutionary traits' rates have occurred in the evolution of the core Tillandsioideae. Our results agree with previously published phylogenetic hypotheses based on plastid data, providing stronger statistical support for some recalcitrant nodes. However, phylogenetic discordance with previously published nuclear marker-based hypotheses was found. Several plastid markers that have been consistently used to address phylogenetic relationships within Tillandsioideae were highly informative for the retrieved plastome phylogeny and further loci are here identified as promising additional markers for future studies. New lineage-specific plastome rearrangements were found to support recently adopted taxonomic groups, including large inversions, as well as expansions and contractions of the inverted repeats. Evolutionary trait rate shifts associated with changes in size and GC content of the plastome regions were found across the phylogeny of core Tillandsioideae.

Project description:Guzmania panamensis (Bromeliaceae), a new species so far endemic to Veraguas province, Western Panama, is described and illustrated. The new species is recognized due to its peduncle much longer than the leaves and its red floral bracts, shorter than the yellow flowers. The new species is compared to morphologically similar species, namely Guzmania monostachia, Guzmania berteroniana, Guzmania elvallensis, and Guzmania skotakii.

Project description:BackgroundDNA barcoding has been successfully established in animals as a tool for organismal identification and taxonomic clarification. Slower nucleotide substitution rates in plant genomes have made the selection of a DNA barcode for land plants a much more difficult task. The Plant Working Group of the Consortium for the Barcode of Life (CBOL) recommended the two-marker combination rbcL/matK as a pragmatic solution to a complex trade-off between universality, sequence quality, discrimination, and cost.Methodology/principal findingsIt is expected that a system based on any one, or a small number of plastid genes will fail within certain taxonomic groups with low amounts of plastid variation, while performing well in others. We tested the effectiveness of the proposed CBOL Plant Working Group barcoding markers for land plants in identifying 46 bromeliad species, a group rich in endemic species from the endangered Brazilian Atlantic Rainforest. Although we obtained high quality sequences with the suggested primers, species discrimination in our data set was only 43.48%. Addition of a third marker, trnH-psbA, did not show significant improvement. This species identification failure in Bromeliaceaecould also be seen in the analysis of the GenBank's matK data set. Bromeliaceae's sequence divergence was almost three times lower than the observed for Asteraceae and Orchidaceae. This low variation rate also resulted in poorly resolved tree topologies. Among the three Bromeliaceae subfamilies sampled, Tillandsioideae was the only one recovered as a monophyletic group with high bootstrap value (98.6%). Species paraphyly was a common feature in our sampling.Conclusions/significanceOur results show that although DNA barcoding is an important tool for biodiversity assessment, it tends to fail in taxonomy complicated and recently diverged plant groups, such as Bromeliaceae. Additional research might be needed to develop markers capable to discriminate species in these complex botanical groups.

Project description:Based on morphological evidence, we propose to raise Tillandsia mauryana forma secundifolia to species level with the name Viridantha secundifolia (Ehlers) Hern.-Cárdenas, Espejo & López-Ferr. Viridantha secundifolia can be readily distinguished by the falciform rosettes, the broadly oblong to square, 1-1.2 × 0.8-1.1 cm leaf sheaths and by the 1.8-2 × 0.7-1.2 cm floral bracts. Additionally, we describe and illustrate Viridantha uniflora Hern.-Cárdenas, Espejo & López-Ferr., from the state of Oaxaca, Mexico. The new species is morphologically similar to Viridantha boqueronensis, but differs by the nearly square leaf sheaths, 1.3-1.5 × 0.4-0.5 cm spikes and by the presence of only one flower per spike. A key to the taxa, morphological descriptions, list of specimens examined, illustrations and a distribution map of the described taxa are included.

Project description:Genome size is known to vary widely across plants. Yet, the evolutionary drivers and consequences of genome size variation across organisms are far from understood. We investigated genome size variation and evolution in two major subfamilies of the Neotropical family Bromeliaceae by determining new genome size values for 83 species, testing phylogenetic signal in genome size variation, and assessing the fit to different evolutionary models. For a subset of epiphytic bromeliad species, we also evaluated the relationship of genome size with thermal traits and relative growth rate (RGR), respectively. Genome size variation in Bromelioideae appears to be evolutionary conserved, while genome size among Tillandsioideae varies considerably, not just due to polyploidy but arguably also due to environmental factors. The subfamilies show fundamental differences in genome size and RGR: Bromelioideae have, on average, lower genome sizes than Tillandsioideae and at the same time exhibit higher RGR. We attribute this to different resource use strategies in the subfamilies. Analyses among subfamilies, however, revealed unexpected positive relationships between RGR and genome size, which might be explained by the nutrient regime during cultivation. Future research should test whether there is indeed a trade-off between genome size and growth efficiency as a function of nutrient supply.

Project description:Pollinators can be a limited resource and natural selection should favour differences in phenotypic characteristics to reduce competition among plants. Bats are important pollinators of many Neotropical plants, including the Bromeliaceae; however, the pre-pollination mechanisms for isolation among sympatric bat-pollinated bromeliads are unknown. Here, we studied the mechanisms for reproductive segregation between Pitcairnia recurvata, Pseudalcantarea viridiflora, Werauhia noctiflorens and W. nutans. The study was conducted at Los Tuxtlas Biosphere Reserve, in Veracruz, Mexico We carried out ex situ and in situ manual pollination treatments to determine the breeding system by assessing fruiting and seedling success and sampled bat visitors using mist-nets and infrared cameras. We determined the nocturnal nectar production pattern, estimating the energetic content of this reward. All four bromeliads are self-compatible, but only P. recurvata appears to require pollinators, because the physical separation between anthers and stigma prevents self-pollination, it is xenogamous and presents a strictly nocturnal anthesis. The bats Anoura geoffroyi, Glossophaga soricina and Hylonycteris underwoodi are probable pollinators of three of the studied bromeliads. We did not record any animal visiting the fourth species. The flowering season of each species is staggered throughout the year, with minimal overlap, and the floral morphology segregates the locations on the body of the bat where the pollen is deposited. The most abundant nectar per flower is provided by P. viridiflora, but P. recurvata offers the best reward per hectare, considering the density of flowering plants. Staggered flowering, different pollen deposition sites on the body of the pollinator and differences in the reward offered may have evolved to reduce the competitive costs of sharing pollinators while providing a constant supply of food to maintain a stable nectarivorous bat community.

Project description:Triblidiaceae (Rhytismatales) currently consists of two genera: Triblidium and Huangshania. Triblidium is the type genus and is characterised by melanized apothecia that occur scattered or in small clusters on the substratum, cleistohymenial (opening in the mesohymenial phase), inamyloid thin-walled asci and hyaline muriform ascospores. Before this study, only the type species, Triblidium caliciiforme, had DNA sequences in the NCBI GenBank. In this study, six specimens of Triblidium were collected from China and France and new ITS, mtSSU, LSU and RPB2 sequences were generated. Our molecular phylogenetic analysis and morphological study demonstrated three new species of Triblidium, which are formally described here: T. hubeiense, T. rostriforme and T. yunnanense. Additionally, our results indicated that Huangshania that was considered to be distinct from Triblidium because of its elongated, transversely-septate ascospores, is congeneric with Triblidium. Therefore, we have placed Huangshania in synonymy under Triblidium, rendering Triblidiaceae a monotypic family.
