Project description:Deficits in temporal judgment in schizophrenia have been observed in behavioral and electrophysiological studies for years. The functional neuroanatomy of temporal judgment in schizophrenia is, however, poorly understood. Recent neurophysiological research suggests that timing deficits in this population may not be widespread across all timing tasks, but specifically associated with high levels of difficulty. We evaluated differences between individuals with schizophrenia (N=16) and healthy subjects (N=18) during a temporal discrimination task at two levels of difficulty. Subjects were studied with functional magnetic resonance imaging (fMRI) at 3T while discriminating tone durations. Behaviorally, the schizophrenia group performed worse than the control group at both levels of difficulty. Similarly, group differences in patterns of brain activation were observed across both difficulty conditions. In the easy condition, individuals with schizophrenia showed less activation in the supplementary motor area and insula/opercula, regions known to be involved in temporal processing. These group differences increased in the difficult condition. In addition, the striatum was less active in individuals with schizophrenia in the difficult condition. Comparing the difficult to easy conditions revealed robust differences in the bilateral striatum and the insula/opercula, suggesting that the striatum plays a key role in temporal processing deficits in schizophrenia, especially under difficult conditions. These observations suggest that temporal judgment deficits reflect widespread neuroanatomical network involvement in schizophrenia, some of which are not directly related to task difficulty. These findings shed light on disparate findings in the timing literature regarding the role of task difficulty in temporal judgment deficits in schizophrenia.

Project description:Using an fMR-adaptation paradigm for different face morphing levels we have recently demonstrated a narrow neuronal tuning to faces even at the sub-exemplar level which was tightly related to perceptual discrimination (Gilaie-Dotan and Malach, 2007). However, it is unclear whether this relationship is unique to faces or is a general property of object representations including unfamiliar objects, and whether the adaptation tuning is due to physical changes in the stimulus or to changes in perceptual discrimination. Here we compared the same face-morph paradigm for upright and inverted faces, thus modulating familiarity and perceptual discrimination effects while equating all low-level features. We found, as expected, a perceptual "inversion effect", i.e. a significant reduction in inverted face discrimination. Importantly, the fMR-adaptation tuning in the fusiform face area (FFA) changed in accordance with the different perceptual sensitivity both for upright and inverted faces. Additional object selective regions displayed differential tuning widths to the two categories. Our results are compatible with a model by which the ability of human observers to discriminate objects depends on the shape tuning properties of individual neurons.

Project description:Visual crowding refers to the impairment of recognizing peripherally presented objects flanked by distractors. Crowding effects, exhibiting a certain spatial extent between target and flankers, can be reduced by perceptual learning. In this experiment, we investigated the learning-induced reduction of crowding in normally sighted participants and tested if learning on one optotype (Landolt-C) transfers to another (Tumbling-E) or vice versa. Twenty-three normally sighted participants (18-42 years) trained on a crowding task in the right-upper quadrant (target at 6.5 degrees eccentricity) over four sessions. Half of the participants had the four-alternative forced-choice task to discriminate the orientation of a Landolt-C, the other half of participants had the task to discriminate the orientation of a Tumbling-E, each flanked by distractors. In the fifth session, all participants switched to the other untrained optotype, respectively. Learning success was measured as reduction of the spatial extent of crowding. We found an overall significant and comparable learning-induced reduction of crowding in both conditions (Landolt-C and Tumbling-E). However, only in the group who trained on the Landolt-C task did learning effects transfer to the other optotype. The specific target-flanker-constellations may modulate the transfer effects found here. Perceptual learning of a crowding task with optotypes could be a promising tool in rehabilitation programs to help improve peripheral vision (e.g. in patients with central vision loss), but the dependence of possible transfer effects on the optotype and distractors used requires further clarification.

Project description:Perceptual decisions are likely to be based on signals that are provided by populations of neurons in early sensory cortical areas. How these neural responses are combined across neurons and over time to mediate behavior is unknown. To study the link between neural responses and perceptual decisions, we recorded the activity of single units (SU) and multiple units (MU) in the primary visual cortex (V1) of monkeys while they performed a reaction-time visual detection task. We then determined how well the target could be detected from these neural signals. We found that, on average, the detection sensitivities supported by SU and MU in V1 are comparable with the detection sensitivity of the monkey even when considering neural responses during brief temporal intervals (median duration, 137 ms) that ended shortly before the monkey's reaction time. However, we observed systematic differences between the overall shape of the neurometric functions and the monkey's psychometric functions. We also examined the quantitative relationship between SU and MU activity and found that MU responses are consistent with the sum of the responses of multiple SU, most of which have low stimulus selectivity. Finally, we found weak but significant trial-to-trial covariations between V1 activity and behavioral choices, demonstrating for the first time that choice probability can be observed at the earliest stages of cortical sensory processing. Together, these results suggest that the activity of a large population of V1 neurons is combined suboptimally by subsequent processing stages to mediate behavioral performance in visual detection tasks.

Project description:A large body of work has focused on understanding stimulus-driven behavior, sex differences in these processes, and the neural circuits underlying them. Many preclinical mouse models present rewarding or aversive stimuli in isolation, ignoring that ethologically, reward seeking requires the consideration of potential aversive outcomes. In addition, the context (or reinforcement schedule under) in which stimuli are encountered can engender different behavioral responses to the same stimulus. Thus, delineating neural control of behavior requires a dissociation between stimulus valence and stimulus-driven behavior. We developed the Multidimensional Cue Outcome Action Task (MCOAT) to dissociate motivated action from cue learning and valence in mice. First, mice acquire positive and negative reinforcement in the presence of discrete discriminative stimuli. Next, discriminative stimuli are presented concurrently allowing for parsing innate behavioral strategies based on reward seeking and avoidance. Lastly, responding in the face of punishment is assessed, thus examining how positive and negative outcomes are relatively valued. First, we identified sex-specific behavioral strategies, showing that females prioritize avoidance of negative outcomes over seeking positive, while males have the opposite strategy. Next, we show that chemogenetically inhibiting D1 medium spiny neurons (MSNs) in the nucleus accumbens-a population that has been linked to reward-driven behavior-reduces positive and increases negative reinforcement learning rates. Thus, D1 MSNs modulate stimulus processing, rather than motivated responses or the reinforcement process itself. Together, the MCOAT has broad utility for understanding complex behaviors as well as the definition of the discrete information encoded within cellular populations.

Project description:Inhibitory control can be triggered directly via the retrieval of previously acquired stimulus-stop associations from memory. However, a recent study suggests that this item-specific stop learning may be mediated via expectancies of the contingencies in play (Best, Lawrence, Logan, McLaren, & Verbruggen, 2016). This could indicate that stimulus-stop learning also induces strategic proactive changes in performance. We further tested this hypothesis in the present study. In addition to measuring expectancies following task completion, we introduced a between-subjects expectancy manipulation in which one group of participants were informed about the stimulus-stop contingencies and another group did not receive any information about the stimulus-stop contingencies. Moreover, we combined this instruction manipulation with a distractor manipulation that was previously used to examine strategic proactive adjustments. We found that the stop-associated items slowed responding in both conditions. Furthermore, participants in both conditions generated expectancies following task completion that were consistent with the stimulus-stop contingencies. The distractor manipulation was ineffective. However, we found differences in the relationship between the expectancy ratings and task performance: in the instructed condition, the expectancies reliably correlated with the response slowing for the stop-associated items, whereas in the uninstructed condition we found no reliable correlation. These differences between the correlations were reliable, and our conclusions were further supported by Bayesian analyses. We conclude that stimulus-stop associations that are acquired either via task instructions or via task practice have similar effects on behaviour but could differ in how they elicit response slowing.

Project description:To interpret the sensory environment, the brain combines ambiguous sensory measurements with knowledge that reflects context-specific prior experience. But environmental contexts can change abruptly and unpredictably, resulting in uncertainty about the current context. Here we address two questions: how should context-specific prior knowledge optimally guide the interpretation of sensory stimuli in changing environments, and do human decision-making strategies resemble this optimum? We probe these questions with a task in which subjects report the orientation of ambiguous visual stimuli that were drawn from three dynamically switching distributions, representing different environmental contexts. We derive predictions for an ideal Bayesian observer that leverages knowledge about the statistical structure of the task to maximize decision accuracy, including knowledge about the dynamics of the environment. We show that its decisions are biased by the dynamically changing task context. The magnitude of this decision bias depends on the observer's continually evolving belief about the current context. The model therefore not only predicts that decision bias will grow as the context is indicated more reliably, but also as the stability of the environment increases, and as the number of trials since the last context switch grows. Analysis of human choice data validates all three predictions, suggesting that the brain leverages knowledge of the statistical structure of environmental change when interpreting ambiguous sensory signals.

Project description:In perceptual decision making, the selection of an appropriate action depends critically on an organism's ability to use sensory inputs to accumulate evidence for a decision. However, differentiating decision-related processes from effects of "time on task" can be difficult. Here we combine the response signal paradigm, in which the experimenter rather than the subject dictates the time of the response, and independent components analysis (ICA) to search for signatures consistent with time on task and decision making, respectively, throughout the brain. Using this novel approach, we identify two such independent components from BOLD activity related to a random dot motion task: one sensitive to the main effect of stimulus duration, and one to both the main effect of motion coherence and its interaction with duration. Furthermore, we demonstrate that these two components are expressed differently throughout the brain, with activity in occipital regions most reflective of the former, activity within intraparietal sulcus modulated by both factors, and more anterior regions including the anterior insula, pre-SMA, and inferior frontal sulcus driven almost exclusively by the latter. Consistent with these ICA findings, cluster analysis identifies a posterior-to-anterior gradient that differentiates regions sensitive to time on task from regions whose activity is strongly tied to motion coherence. Together, these findings demonstrate that progressively more anterior regions are likely to participate in progressively more proximate decision-related processes.

Project description:Trial-and-error learning is a universal strategy for establishing which actions are beneficial or harmful in new environments. However, learning stimulus-response associations solely via trial-and-error is often suboptimal, as in many settings dependencies among stimuli and responses can be exploited to increase learning efficiency. Previous studies have shown that in settings featuring such dependencies, humans typically engage high-level cognitive processes and employ advanced learning strategies to improve their learning efficiency. Here we analyze in detail the initial learning phase of a sample of human subjects (N = 85) performing a trial-and-error learning task with deterministic feedback and hidden stimulus-response dependencies. Using computational modeling, we find that the standard Q-learning model cannot sufficiently explain human learning strategies in this setting. Instead, newly introduced deterministic response models, which are theoretically optimal and transform stimulus sequences unambiguously into response sequences, provide the best explanation for 50.6% of the subjects. Most of the remaining subjects either show a tendency towards generic optimal learning (21.2%) or at least partially exploit stimulus-response dependencies (22.3%), while a few subjects (5.9%) show no clear preference for any of the employed models. After the initial learning phase, asymptotic learning performance during the subsequent practice phase is best explained by the standard Q-learning model. Our results show that human learning strategies in the presented trial-and-error learning task go beyond merely associating stimuli and responses via incremental reinforcement. Specifically during initial learning, high-level cognitive processes support sophisticated learning strategies that increase learning efficiency while keeping memory demands and computational efforts bounded. The good asymptotic fit of the Q-learning model indicates that these cognitive processes are successively replaced by the formation of stimulus-response associations over the course of learning.

Project description:A large body of literature has examined specificity and transfer of perceptual learning, suggesting a complex picture. Here, we distinguish between transfer over variations in a "task-relevant" feature (e.g., transfer of a learned orientation task to a different reference orientation) and transfer over a "task-irrelevant" feature (e.g., transfer of a learned orientation task to a different retinal location or different spatial frequency), and we focus on the mechanism for the latter. Experimentally, we assessed whether learning a judgment of one feature (such as orientation) using one value of an irrelevant feature (e.g., spatial frequency) transfers to another value of the irrelevant feature. Experiment 1 examined whether learning in eight-alternative orientation identification with one or multiple spatial frequencies transfers to stimuli at five different spatial frequencies. Experiment 2 paralleled Experiment 1, examining whether learning in eight-alternative spatial-frequency identification at one or multiple orientations transfers to stimuli with five different orientations. Training the orientation task with a single spatial frequency transferred widely to all other spatial frequencies, with a tendency to specificity when training with the highest spatial frequency. Training the spatial frequency task fully transferred across all orientations. Computationally, we extended the identification integrated reweighting theory (I-IRT) to account for the transfer data (Dosher, Liu, & Lu, 2023; Liu, Dosher, & Lu, 2023). Just as location-invariant representations in the original IRT explain transfer over retinal locations, incorporating feature-invariant representations effectively accounted for the observed transfer. Taken together, we suggest that feature-invariant representations can account for transfer of learning over a "task-irrelevant" feature.
