Project description:A new, morphologically cryptic species of phaneropterine bush-crickets is described from the grasslands of the Romanian Eastern Carpathians. Despite the morphological and acoustic similarities with the recently described Isophya nagyi Szövényi, Puskás & Orci, I. bucovinensissp. n. is characterized by a peculiar male calling song, with faster syllable repetition rate (160-220 syllables per minute, at 22-27°C) and less complex syllable structure (composed of only two elements instead of three observable in I. nagyi). The morphological description of the new species is supplemented with an oscillographic and spectrographic analysis of the male calling song and male-female pair-forming acoustic duet. An acoustic signal-based identification key is provided for all the presently known species of the Isophya camptoxypha species group, including the new species.

High-speed duetting - latency times of the female acoustic response within the bush-cricket genera Leptophyes and Andreiniimon (Orthoptera, Phaneropteridae).

Project description:To find a mate, male and female bush-crickets of the family Phaneropteridae typically engage in duets. The male sings and the female responds. For mutual recognition, the amplitude pattern of the male song and the species-specific timing of the female response have been shown to be very important. In the seven studied species, belonging to the genera Leptophyes and Andreiniimon, these duets are extremely fast and nearly completely in the ultrasonic range. The females produce very short sounds by fast closing movements of the tegmina. They respond with species-specific delays of 20 to 150 ms after the beginning of the male song. The different latency times are probably not important for species recognition, since in sympatric species they are quite similar.

Rapid chromosomal evolution in the bush-cricket Gonatoxia helleri Hemp, 2016 (Orthoptera, Phaneropterinae).

Project description:Gonatoxia helleri Hemp, 2016 is one of the most widespread bush-crickets of the genus Gonatoxia Karsch, 1889 in East Africa. This species with seven large chromosomes (2n? = 7) differs from other representatives of the genus Gonatoxia drastically by its reduced chromosome number, the asymmetrical karyotype including karyomorphs rarely found in tettigoniids, as well as in irregularities in the course of meiosis. To better understand the origin of such an exceptional karyotype, chromosomes of 29 specimens from four populations/localities were studied using classical techniques, such as C-banding, silver impregnation, fluorochrome double staining and fluorescence in situ hybridization (FISH) technique with 18S rDNA and (TTAGG) n telomeric probes. FISH showed many 18S rDNA loci as well as interstitial telomeric sequences, where chromosome morphology varied in these components in terms of quantity and distribution. The 18S rDNA loci coincided with active NORs and C-banding patterns. We suggest that a combination of Robertsonian rearrangements and/or multiple common tandem fusions involving the same chromosomes contributed to the formation of this karyotype/karyomorphs. The results are the first step towards a better understanding of chromosomal reorganization and evolution within the genus Gonatoxia. Low chromosome number, together with the incidence of chromosomal polymorphism that is higher in G. helleri than previously reported in bush-crickets, implies that this species can be a valuable new model for cytogenetic and speciation studies. Our findings suggest that chromosomal translocations lead to diversification and speciation in this species and could be the driving force of adaptive radiation.

Project description:Insects have developed different structures to adhere to surfaces. Most common are smooth and hairy attachment pads, while nubby pads have also been described for representatives of Mantophasmatodea, Phasmida and Plecoptera. Here we report on the unusual combination of nubby and smooth tarsal attachment structures in the !nara cricket Acanthoproctus diadematus. Their three proximal tarsal pads (euplantulae) have a nubby surface, whereas the most distal euplantula is rather smooth with a hexagonal ground pattern resembling that described for the great green bush-cricket Tettigonia viridissima. This is, to our knowledge, the first report on nubby euplantulae in Orthoptera and the co-occurrence of nubby and smooth euplantulae on a single tarsus in a polyneopteran species. When adhering upside down to a horizontal glass plate, A. diadematus attaches its nubby euplantulae less often, compared to situations in which the animal is hanging upright or head down on a vertical plate. We discuss possible reasons for this kind of clinging behaviour, such as morphological constrains, the different role of normal and shear forces in attachment enhancement of the nubby and smooth pads, ease of the detachment process, and adaptations to walking on cylindrical substrates.

Isolated populations of the bush-cricket Pholidoptera frivaldszkyi (Orthoptera, Tettigoniidae) in Russia suggest a disjunct area of the species distribution.

Project description:Phylogenetic analysis and assessment of the species status of mostly isolated populations of Pholidoptera frivaldszkyi in south-western Russia occurring far beyond the accepted area of the species distribution in the Carpathian-Balkan region were performed. Using the mitochondrial DNA cytochrome c oxidase subunit I gene fragment, we found a very low level of genetic diversity in these populations. Phylogeographic reconstruction did not support recent introduction events but rather historical range fragmentation. The grouping of the Russian and Romanian haplotypes in a distinct phylogenetic clade suggests that the pre-glacial range of P. frivaldszkyi had extended towards the Ponto-Caspian region, with considerable gene flow between different refugia. However, post-glacial northward expansion of the species from supposed Caucasus refugia contributed most likely to the current disjunct distribution of this relict-like bush-cricket.

Island mysteries in the spotlight: Barbitistes kaltenbachi and Rhacocleis buchichii, the only bush-cricket species endemic to Croatia (Orthoptera, Tettigoniidae).

Project description:Hvar Saw Bush-cricket Barbitistes kaltenbachi Harz, 1965 (Phaneropterinae: Barbitistini) and Lesina Bush-cricket Rhacocleis buchichii Brunner von Wattenwyl in Herman 1874 (Tettigoniinae: Platycleidini) are flightless orthopterans restricted to a narrow area in the Mediterranean part of Croatia, both originally described from Hvar Island. In this study, all available information on these two interesting species is presented: data on morphology, bioacoustics, distribution, habitat, and a key to identification of the species belonging to genera Barbitistes and Rhacocleis in Croatia. The songs of both B. kaltenbachi and R. buchichii are described here for the first time, with the former one being the second known example of a synchronising and presumably duetting species. Both species were reassessed according to the IUCN Red List criteria, where B. kaltenbachi should be considered an endangered species, while R. buchichii is suggested to be downgraded to a less threatened category. Biogeography and evolution of the species are briefly discussed.

Structural biomechanics determine spectral purity of bush-cricket calls.

Project description:Bush-crickets (Orthoptera: Tettigoniidae) generate sound using tegminal stridulation. Signalling effectiveness is affected by the widely varying acoustic parameters of temporal pattern, frequency and spectral purity (tonality). During stridulation, frequency multiplication occurs as a scraper on one wing scrapes across a file of sclerotized teeth on the other. The frequency with which these tooth-scraper interactions occur, along with radiating wing cell resonant properties, dictates both frequency and tonality in the call. Bush-cricket species produce calls ranging from resonant, tonal calls through to non-resonant, broadband signals. The differences are believed to result from differences in file tooth arrangement and wing radiators, but a systematic test of the structural causes of broadband or tonal calls is lacking. Using phylogenetically controlled structural equation models, we show that parameters of file tooth density and file length are the best-fitting predictors of tonality across 40 bush-cricket species. Features of file morphology constrain the production of spectrally pure signals, but systematic distribution of teeth alone does not explain pure-tone sound production in this family.

Auditory DUM neurons in a bush-cricket: inhibited inhibitors.

Project description:Thoracic ganglia of many hearing insects house the first level of auditory processing. In bush-crickets, the largest population of local auditory neurons in the prothoracic processing centre are dorsal unpaired median (DUM) neurons. It has been suggested that DUM neurons are inhibitory using γ-aminobutyric acid (GABA) as transmitter. Immunohistochemistry reveals a population of about 35-50 GABA-positive somata in the posterior medial cluster of the prothoracic ganglion. Only very few small somata in this cluster remain unstained. At least 10 neurites from 10 neurons can be identified. Intracellularly stained auditory DUM neurons have their soma in the cluster of median GABA positive cells and most of them exhibit GABA-immunoreactivity. Responses of certain DUM neurons show obvious signs of inhibition. Application of picrotoxin (PTX), a chloride-channel blocker in insects, changes the responses of many DUM neurons. They become broader in frequency tuning and broader or narrower in temporal pattern tuning. Furthermore, inhibitory postsynaptic potentials (IPSPs) may be replaced by excitatory postsynaptic potentials. Loss of an IPSP in the rising graded potential after PTX-application leads to a significant reduction of first-spike latency. Therefore, auditory DUM neurons receive effective inhibition and are the best candidates for inhibition in DUM neurons and other auditory interneurons.

Acoustic telemetry reveals cryptic residency of whale sharks.

Project description:Although whale sharks (Rhincodon typus) have been documented to move thousands of kilometres, they are most frequently observed at a few predictable seasonal aggregation sites. The absence of sharks at the surface during visual surveys has led to the assumption that sharks disperse to places unknown during the long 'off-seasons' at most of these locations. Here we compare 2 years of R. typus visual sighting records from Mafia Island in Tanzania to concurrent acoustic telemetry of tagged individuals. Sightings revealed a clear seasonal pattern with a peak between October and February and no sharks observed at other times. By contrast, acoustic telemetry demonstrated year-round residency of R. typus. The sharks use a different habitat in the off-season, swimming deeper and further away from shore, presumably in response to prey distributions. This behavioural change reduces the sharks' visibility, giving the false impression that they have left the area. We demonstrate, for the first time to our knowledge, year-round residency of unprovisioned, individual R. typus at an aggregation site, and highlight the importance of using multiple techniques to study the movement ecology of marine megafauna.
