Complete Genome Sequence of a Natural Recombinant H9N2 Influenza Virus from Wild Birds in Republic of Korea.

ABSTRACT: We report the complete genome sequence of a natural recombinant H9N2 avian influenza virus (AIV) that was isolated from a wild bird in the Republic of Korea in 2005. The genomic sequence and phylogenetic analyses showed that this virus contains six genes of North American lineage AIV and two genes of Eurasian lineage AIV. These data are beneficial for investigating the ecology and epidemiology of AIVs.

Project description:In 2007, we isolated a natural recombinant H9N2 avian influenza virus (AIV) from the fecal droppings of a white-fronted goose (Anser albifrons) in South Korea. Phylogenetic analyses of the complete genome sequence showed that polymerase acidic (PA) and neuraminidase (NA) genes belonged to the Eurasian lineage AIV, but polymerase basic 2 (PB2), PB1, hemagglutinin (HA), nucleoprotein (NP), matrix (M), and nonstructural (NS) genes belonged to the North-American lineage AIV. These data are beneficial for understanding the ecology and epidemiology of AIVs.

Complete Genome Sequence of an Avian H1N1 Influenza Virus Strain Isolated from Migratory Birds in the Republic of Korea.

Project description:Here, we report the complete genome sequence of an H1N1 avian influenza virus (AIV), which was isolated from the feces of migratory birds in the Republic of Korea during the winters of 2014 and 2015. Full-genome sequencing and phylogenetic analysis revealed that all genome segments belonged to the Eurasian lineage.

Complete genome sequence of a novel avian paramyxovirus isolated from wild birds in South Korea.

Project description:A novel avian paramyxovirus (APMV), Cheonsu1510, was isolated from wild bird feces in South Korea and serologically and genetically characterized. In hemagglutination inhibition tests, antiserum against Cheonsu1510 showed low reactivity with other APMVs and vice versa. The complete genome of Cheonsu1510 comprised 15,408 nucleotides, contained six open reading frames (3'-N-P-M-F-HN-L-5'), and showed low sequence identity to other APMVs (< 63%) and a unique genomic composition. Phylogenetic analysis revealed that Cheonsu1510 was related to but distinct from APMV-1, -9, and -15. These results suggest that Cheonsu1510 represents a new APMV serotype, APMV-17.

H9N2 influenza viruses from birds used in falconry.

Project description:H9N2 avian influenza viruses continue to spread in poultry and wild birds throughout Eurasia.To characterize H9N2 influenza viruses from pheasants, quail, and white-bellied bustards (WBBs) used to train falcons in the United Arab Emirates (UAE).Four H9N2 viruses were isolated from pheasants, quail, and WBB used for falconry in the UAE, and antigenic, molecular, phylogenetic analysis, and invivo characterization of H9N2 viruses were performed.The pheasant and WBB isolates were antigenically and molecularly clearly related and along with the quail isolates contained multiple "avian-human" substitutions. The release of smuggled H9N2-infected birds for falconry may contribute to the spread of these viruses to wild birds, domestic poultry, and humans.

Characterization of recombinant H9N2 influenza viruses isolated from wild ducks in China.

Project description:Wild birds are considered to be the natural reservoirs for avian influenza A viruses (AIV). During active influenza surveillance in Poyang Lake of southeast China, we isolated and characterized 11 H9N2 viruses from two species of wild ducks. Phylogenetic analysis showed that the 11 isolates were almost identical with 99.3-100% nucleotide homology in their entire genome, and they all closely related in whole eight genes (95.6-99.4% homology) to human H9N2 isolates (HK/33982/2009) and clustered in the same sublineage. The isolates belonged to triple reassortant H9N2 genotype viruses containing Ck/Bei-like NA genes, Y439-like PA genes and six other G1-like genes. We also found that the subtype of virus replicated efficiently in the lungs and tracheas of BALB/c mice and caused mortality in 20-40% of infected groups after 3-6 days, which indicates that the subtype of virus is capable of establishing lethal mammalian infections. However, whether or not the virus has features transmittable from wild ducks to humans is not known. This study showed H9N2 subtype avian influenza virus for the first time in wild birds, and suggests that wild birds may carry the virus along migratory routes, highlighting the need for continued surveillance of wild birds.

Infectivity and transmissibility of H9N2 avian influenza virus in chickens and wild terrestrial birds.

Project description:Genetic changes in avian influenza viruses influence their infectivity, virulence and transmission. Recently we identified a novel genotype of H9N2 viruses in widespread circulation in poultry in Pakistan that contained polymerases (PB2, PB1 and PA) and non-structural (NS) gene segments identical to highly pathogenic H7N3 viruses. Here, we investigated the potential of these viruses to cause disease and assessed the transmission capability of the virus within and between poultry and wild terrestrial avian species. Groups of broilers, layers, jungle fowl, quail, sparrows or crows were infected with a representative strain (A/chicken/UDL-01/08) of this H9N2 virus and then mixed with naïve birds of the same breed or species, or different species to examine transmission. With the exception of crows, all directly inoculated and contact birds showed clinical signs, varying in severity with quail showing the most pronounced clinical signs. Virus shedding was detected in all infected birds, with quail showing the greatest levels of virus secretion, but only very low levels of virus were found in directly infected crow samples. Efficient virus intra-species transmission was observed within each group with the exception of crows in which no evidence of transmission was seen. Interspecies transmission was examined between chickens and sparrows and vice versa and efficient transmission was seen in either direction. These results highlight the ease of spread of this group of H9N2 viruses between domesticated poultry and sparrows and show that sparrows need to be considered as a high risk species for transmitting H9N2 viruses between premises.

Detection of novel reassortant H9N2 avian influenza viruses in wild birds in Jiangxi Province, China.

Project description:The five avian influenza A/H9N2 viruses isolated from wild birds in Jiangxi, China in 2015 are novel reassortants which most likely evolved from multiple lineages. They shared a high similarity with isolates from poultry, suggesting a frequent contact and continuous viral circulation at the bird-poultry interface. Given the continuous reassortment of H9N2 viruses, it will of substantial importance to implement routine surveillance in wild birds to successfully control avian influenza viruses and better the early warning system of the emerging reassortants with pandemic potential.

Phylogenetic analyses indicated that this virus was phylogenetically related to the Y280 lineage. Sequence analysis showed 1 amino acid deletion in HA1 and 3 amino acid deletions in the NA stalk region.

Characterization of H7 influenza A virus in wild and domestic birds in Korea.

Project description:During surveillance programs in Korea between January 2006 and March 2011, 31 H7 avian influenza viruses were isolated from wild birds and domestic ducks and genetically characterized using large-scale sequence data. All Korean H7 viruses belonged to the Eurasian lineage, which showed substantial genetic diversity, in particular in the wild birds. The Korean H7 viruses from poultry were closely related to those of wild birds. Interestingly, two viruses originating in domestic ducks in our study had the same gene constellations in all segment genes as viruses originating in wild birds. The Korean H7 isolates contained avian-type receptors (Q226 and G228), no NA stalk deletion (positions 69-73), no C-terminal deletion (positions 218-230) in NS1, and no substitutions in PB2-627, PB1-368, and M2-31, compared with H7N9 viruses. In pathogenicity experiments, none of the Korean H7 isolates tested induced clinical signs in domestic ducks or mice. Furthermore, while they replicated poorly, with low titers (10?·??¹·³ EID??/50 µl) in domestic ducks, all five viruses replicated well (up to 7-10 dpi, 10?·???·³EID??/50 µl) in the lungs of mice, without prior adaptation. Our results suggest that domestic Korean viruses were transferred directly from wild birds through at least two independent introductions. Our data did not indicate that wild birds carried poultry viruses between Korea and China, but rather, that wild-type H7 viruses were introduced several times into different poultry populations in eastern Asia.
