ABSTRACT: A stable and highly selective fluorescent probe has been designed and synthesized for the rapid detection of fluoride ions (F(-)) in aqueous solution and living cells. The design was based on the high reactivity of F(-) toward a silyl group.

Two-Step Sensing, Colorimetric and Ratiometric Fluorescent Probe for Rapid Detection of Bisulfite in Aqueous Solutions and in Living Cells.

Project description:Bisulfite and sulfite (HSO3 -/SO3 2-) are not only widely used toxic chemicals but also active anions with important biological functions. Hence, the development of new detection methods for HSO3 -/SO3 2- is important for environmental security and human health. In this paper, we report a symmetrical hemicyanine for the detection of HSO3 -/SO3 2-, SHC, which is constructed with p-diphthalaldehyde with trimethylbenzoindolium via condensation. The red fluorescent probe can fast respond to HSO3 -/SO3 2- (<30 s) to give cyan fluorescence, and its sensing process is twice nucleophilic additions, which were observed from fluorescence response, initial ratiometric change, and subsequent turn-on increment; especially in a low-concentration level, the ratiometric fluorescence measurement can eliminate environmental interference. This probe can achieve a quantitative detection of HSO3 -/SO3 2- in a wide concentration range. Furthermore, the probe SHC is a mitochondria-specific probe for ratiometric fluorescent detection of HSO3 -/SO3 2- in living cells.

A novel fluorescent off-on probe for the sensitive and selective detection of fluoride ions.

Project description:A highly sensitive and selective fluorescent probe for fluoride ions has been developed by incorporating the dimethylphosphinothionyl group as a recognition moiety into the fluorophore of coumarin. The detection mechanism is based on the fluoride ion-triggered cleavage of the dimethylphosphinothionyl group, followed by the release of coumarin, which leads to a large fluorescence enhancement at 455 nm (λ ex = 385 nm). Under the optimized conditions, the fluorescence enhancement of the probe is directly proportional to the concentration of fluoride ions in the range of 0-30 μM with a detection limit of 0.29 μM, which is much lower than the maximum content of fluoride ions guided by WHO. Notably, satisfying results have been obtained by utilizing the probe to determine fluoride ions in real-water samples and commercially available toothpaste samples. The proposed probe is rather simple and may be useful in the detection of fluoride ions in more real samples.

Rapid, photoinduced electron transfer-modulated, turn-on fluorescent probe for detection and cellular imaging of biologically significant thiols.

Project description:There is a very limited number of existing probes whose fluorescence is turned on in the presence of the class of biological thiols made up of glutathione, cysteine, and homocysteine. The extant probes for this class of biological thiols commonly have poor aqueous solubility and long analyte response times, and they demand a very high probe/thiol ratio for decreased time of significant reporter signal generation; knowledge regarding their selectivity with respect to other sulfur-based analytes is unclear. Described here is a previously unreported photoinduced electron-transfer-quenched probe (HMBQ-Nap 1) that offers highly selective and rapid in vitro detection of this class of biologically important thiols at low concentrations and low probe/thiol ratio, and importantly, very rapid imaging of these biological thiols in human cells.

A novel HBI-based ratiometric fluorescent probe for rapid detection of trifluoroborate.

Project description:A 2-(2'-hydroxyphenyl)benzimidazole (HBI)-based ratiometric fluorescent probe, known as BTEP, was synthesized using 5-bromosalicylaldehyde as the raw material via Sonogashira coupling and condensation reaction. This probe was designed for rapid detection of boron trifluoride solutions and gases. The N and O coordination atoms in the probe undergo a boron difluoride addition with BF3, which affects the process of excited state intramolecular proton transfer (ESIPT) leading to a blue shift of fluorescence emission. Obvious changes in the fluorescence signal can be observed within 60 seconds. The introduction of an acetylene trimethylsilane fragment increases the conjugate plane and is beneficial to improving the selectivity of the probe. The I408/I479 fluorescence ratio of the probe displays a linear relationship with the concentration of BF3 in the range of 5-50 μM, with a detection of limit as low as 69.5 nM. Furthermore, the probe demonstrates specific and selective recognition of BF3 among eight common interference substances. Test strips prepared using BTEP have the capability of real-time naked-eye detection of trace BF3 gas.

Rapid cancer detection by topically spraying a γ-glutamyltranspeptidase-activated fluorescent probe.

Project description:The ability of the unaided human eye to detect small cancer foci or accurate borders between cancer and normal tissue during surgery or endoscopy is limited. Fluorescent probes are useful for enhancing visualization of small tumors but are typically limited by either high background signal or the requirement for administration hours to days before use. We synthesized a rapidly activatable, cancer-selective fluorescence imaging probe, γ-glutamyl hydroxymethyl rhodamine green (gGlu-HMRG), with intramolecular spirocyclic caging for complete quenching. Activation occurs by rapid one-step cleavage of glutamate with γ-glutamyltranspeptidase (GGT), which is not expressed in normal tissue, but is overexpressed on the cell membrane of various cancer cells, thus leading to complete uncaging and dequenching of the fluorescence probe. In vitro activation of gGlu-HMRG was evident in 11 human ovarian cancer cell lines tested. In vivo in mouse models of disseminated human peritoneal ovarian cancer, activation of gGlu-HMRG occurred within 1 min of topically spraying the tumor, creating high signal contrast between the tumor and the background. The gGlu-HMRG probe is practical for clinical application during surgical or endoscopic procedures because of its rapid and strong activation upon contact with GGT on the surface of cancer cells.

A Novel NIR Fluorescent Probe for Highly Selective Detection of Nitroreductase and Hypoxic-Tumor-Cell Imaging.

Project description:Nitroreductase as a potential biomarker for aggressive tumors has received extensive attention. In this work, a novel NIR fluorescent probe for nitroreductase detection was synthesized. The probe Py-SiRh-NTR displayed excellent sensitivity and selectivity. Most importantly, the confocal fluorescence imaging demonstrated that HepG-2 cells treated with Py-SiRh-NTR under hypoxic conditions showed obvious enhanced fluorescence, which means that the NTR was overexpressed under hypoxic conditions. Moreover, the probe showed great promise that could help us to study related anticancer mechanisms research.

Coumarin Probe for Selective Detection of Fluoride Ions in Aqueous Solution and Its Bioimaging in Live Cells.

Project description:We have synthesized novel coumarin-based fluorescent chemosensors for detection of fluoride ions in aqueous solution. The detection mechanism relied on a fluoride-mediated desilylation triggering fluorogenic reaction and a strong interaction between fluoride and the silicon center. In this work, the hydroxyl-decorated coumarins containing oxysilyl moiety have been synthesized through the aldehyde-functionalized coumarins. The optical responses toward fluoride, as well as aqueous stability studies of both aldehyde and hydroxyl functionalized coumarins, have been investigated. Due to the highest fluorescence enhancement upon the addition of fluoride and good stability in aqueous solution, the hydroxyl-decorated coumarin connected with the bulky tert-butyldiphenyloxysilyl group (-OSitBuPh?) has been selected for further investigation of its potential as a fluoride sensor. This hydroxyl-decorated coumarin can selectively sense fluoride ions in aqueous media (contain 0.8% MeCN) with desirable response times (40 min). The limit of detection of this compound was determined as 0.043 ppm, satisfying the standard fluoride level (0.7 ppm) in drinking water recommended by U.S. Department of Health and Human Services. The application of this silyl-capped coumarin derivative for fluoride analysis in collected water samples displayed satisfactory analytical accuracy (<5% error). Finally, this compound was successfully employed in fluorescence bioimaging of fluoride ions in human liver cancer cells, indicating its excellent cell permeability, ability to retain inside the living cells, and good stability under physiological conditions.

Highly Selective Fluorescent Probe for the Detection of Copper (II) and Its Application in Live Cell Imaging.

Project description:The development of fluorescent methods for the detection of metal ions is of great importance due to their diverse environmental and biological roles. Herein, a rhodamine 6G-based off-on fluorescent probe (L1) with a t-butyl pyrrole moiety as the recognition site was designed and synthesized. Photophysical studies show that L1 exhibits excellent sensitivity and selectivity towards Cu2+ to other metal ions in neutral acetonitrile aqueous media. Mechanism studies suggest that the recognition process may associate with a Cu2+ promoted hydrolysis reaction of L1. Furthermore, L1 has been successfully applied in fluorescence imaging of Cu2+ ion in living cells.

A "Turn-On" fluorescent probe for sensitive and selective detection of fluoride ions based on aggregation-induced emission.

Project description:Based on the fluorophore of 2-(2'-hydroxyphenyl)benzothiazole (HBT) with aggregation-induced emission (AIE) properties, a highly selective and sensitive fluorescent probe PBT towards F- was investigated. "Turn-On" fluorescence type signaling was realized by employing fluoride-selective cleavage of the latent thiophosphinated probe in mixed aqueous media. The probe is designed in such a way that the excited state intramolecular proton transfer (ESIPT) of the HBT moiety becomes blocked. The chemodosimetric approach of F- to the probe results in the recovery of the ESIPT by removal of a free AIE-active HBT moiety through a subsequent hydrolysis process. The F- detection limit of the probe was 3.8 nM in the dynamic range of 0.5 μM to 10 μM. In addition, the proposed probe has been used to detect F- in water samples and toothpaste samples with satisfying results.

A curcumin-based AIEE-active fluorescent probe for Cu<sup>2+</sup> detection in aqueous solution.

Project description:Curcuminoids have been extensively investigated as metal ion probes, but the intrinsic aggregation-caused-quenching (ACQ) characteristic of curcumin would hinder their applications in aqueous solution. Fortunately, tetraphenylethylene (TPE) could endow the compounds with aggregation-induced emission (AIE)/aggregation-induced enhanced emission (AIEE) characteristics to eliminate the ACQ effect. According to this strategy, a series of TPE-modified curcumin derivatives L1-4 were prepared and studied for their AIEE properties. Among the four TPE-curcumin analogues, only L1 particles have been successfully used as an on-off fluorescence probe for detecting Cu2+ in aqueous solution. The fluorescence titration experiment determined its detection limit of 1.49 × 10-7 mol L-1, and the binding ratio between L1 and Cu2+ was estimated as 2 : 1, which was in agreement with the results of high resolution mass spectrum and Job's plot. In addition, the binding constant was evaluated as 6.77 × 102 M-1 using a Benesi-Hildebrand plot. Finally, the obtained L1-based indicator paper showed significant fluorescence response to Cu2+ aqueous solution. This TPE-modified strategy improves the detection capability of curcumin probe in aqueous solution and provides a feasible way to obtain other probes with ACQ characteristics.
