Gd3+-Doping Effect on Upconversion Emission of NaYF4: Yb3+, Er3+/Tm3+ Microparticles.

Project description:β-NaYF4 microcrystals co-doped with Yb3+, Er3+/Tm3+, and Gd3+ ions were synthesized via a hydrothermal method using rare-earth chlorides as the precursors. The SEM and XRD data show that the doped β-NaYF4 form uniform hexagonal prisms with an approximate size of 600-800 nm. The partial substitution of Y by Gd results in size reduction of microcrystals. Upconversion luminescence spectra of microcrystals upon 980 nm excitation contain characteristic intra-configurational ff bands of Er3+/Tm3+ ions. An addition of Gd3+ ions leads to a significant enhancement of upconversion luminescence intensity with maxima at 5 mol % of dopant.

Tuning the upconversion photoluminescence lifetimes of NaYF4:Yb3+, Er3+ through lanthanide Gd3+ doping.

Project description:The multiplexing capacity of conventional fluorescence materials are significantly limited by spectral overlap and background interference, mainly due to their short-lived fluorescence lifetimes. Here, we adopt a novel Gd3+ doping strategy in NaYF4 host materials, realized tuning of upconversion photoluminescence (UCPL) lifetimes at selective emissions. Time-correlated single-photon counting (TCSPC), was applied to measure the photoluminescence lifetimes accurately. We demonstrated the large dynamic range of lifetimes of upconversion nanoparticles with good upconversion quantum yields, mainly owing to the dominance of high efficient energy transfer upconversion mechanism. The exceptional tunable properties of upconversion materials allow great potential for them to be utilized in biotechnology and life sciences.

Simultaneous morphology manipulation and upconversion luminescence enhancement of ?-NaYF4:Yb3+/Er3+ microcrystals by simply tuning the KF dosage.

Project description:A strategy has been adopted for simultaneous morphology manipulation and upconversion luminescence enhancement of ?-NaYF4:Yb(3+)/Er(3+) microcrystals by simply tuning the KF dosage. X-ray power diffraction (XRD), field emission scanning electron microscopy (FE-SEM), transmission electron microscopy (TEM), X-ray photoelectron spectroscopy (XPS) and photoluminescence spectra (PL) were used to characterize the samples. The influence of molar ratio of KF to Y(3+) on the crystal phase and morphology has been systematically investigated and discussed. It is found that the molar ratio of KF to Y(3+) can strongly control the morphology of the as-synthesized ?-NaYF4 samples because of the different capping effect of F(-) ions on the different crystal faces. The possible formation mechanism has been proposed on the basis of a series of time-dependent experiments. More importantly, the upconversion luminescence of ?-NaYF4:Yb(3+)/Er(3+) was greatly enhanced by increasing the molar ratio of KF to RE(3+) (RE = Y, Yb, Er), which is attributed to the distortion of local crystal field symmetry around lanthanide ions through K(+) ions doping. This synthetic methodology is expected to provide a new strategy for simultaneous morphology control and remarkable upconversion luminescence enhancement of yttrium fluorides, which may be applicable for other rare earth fluorides.

Time-resolved universal temperature measurements using NaYF4:Er3+,Yb3+ upconverting nanoparticles in an electrospray jet.

Project description:Hexagonal upconverting nanoparticles (UCNPs) of NaYF4:Er3+,Yb3+ (ca. 300 nm) have been widely used to measure the temperature at the nanoscale using luminescence ratio thermometry. However, several factors limit their applications. For example, changes in the peak shape, mainly is the S-band emission, hinders their ability to be used as a universal temperature sensor. Herein, we introduce a universal calibration protocol for NaYF4:Er3+,Yb3+ upconverting nanoparticles that is robust to environmental changes and gives a precise temperature measurement. We used this new procedure to calculate the temperature profile inside a Taylor cone generated with an electrospray jet. Inside the Taylor cone the fluid velocity increases toward the tip of the cone. A constant acquisition length leads to a decrease in excitation and acquisition time. This decrease in excitation time causes a peak shape change that corrupts the temperature measurement if the entire peak shape is integrated in the calibration. Our universal calibration circumvents this problem and can be used for time-resolved applications. The temperature at the end of the Taylor cone increases due to the creation of a whispering gallery mode cavity with 980 nm excitation. We use time-resolved energy balance equations to support our optical temperature measurements inside the Taylor cone. We believe that the findings of this paper provide a foundation for time-resolved temperature measurements using NaYF4:Er3+,Yb3+ upconverting nanoparticles and can be used to understand temperature-dependent reactions such as protein unfolding inside microjet/microdroplets and microfluidic systems.

Effective Shielding of NaYF4:Yb3+,Er3+ Upconverting Nanoparticles in Aqueous Environments Using Layer-by-Layer Assembly.

Project description:Aqueous solutions are the basis for most biomedical assays, but they quench the upconversion luminescence significantly. Surface modifications of upconverting nanoparticles are vital for shielding the obtained luminescence. Modifications also provide new possibilities for further use by introducing attaching sites for biomolecule conjugation. We demonstrate the use of a layer-by-layer surface modification method combining varying lengths of negatively charged polyelectrolytes with positive neodymium ions in coating the upconverting NaYF4:Yb3+,Er3+ nanoparticles. We confirmed the formation of the bilayers and investigated the surface properties with Fourier transform infrared and reflectance spectroscopy, thermal analysis, and ζ-potential measurements. The effect of the coating on the upconversion luminescence properties was characterized, and the bilayers with the highest improvement in emission intensity were identified. In addition, studies for the nanoparticle and surface stability were carried out in aqueous environments. It was observed that the bilayers were able to shield the materials' luminescence from quenching also in the presence of phosphate buffer that is currently considered the most disruptive environment for the nanoparticles.

Multifunctional nanoclusters of NaYF4:Yb3+,Er3+ upconversion nanoparticle and gold nanorod for simultaneous imaging and targeted chemotherapy of bladder cancer.

Project description:This paper reports successful synthesis of multifunctional nanoclusters of upconversion nanoparticle (UCNP) and gold nanorod (AuNR) through a PEGylation process. UCNPs emit visible luminescence under near-infrared excitation, producing high-contrast images with no background fluorescence. When coupled with AuNRs, the resulting UCNP-AuNR multifunctional nanoclusters are capable of simultaneous detection and treatment of bladder cancer. These UCNP-AuNR nanoclusters are further functionalized with antibodies to epidermal growth factor receptor (EGFR) to target bladder cancer cells known to overexpress EGFRs. This paper demonstrates, for the first time, efficient targeting of bladder cancer cells with UCNP-AuNR nanoclusters. In addition to high-contrast imaging and consequently high sensitivity detection of bladder cancer cells, highly selective optoporation-assisted chemotherapy was accomplished using a dosage of chemotherapy agent significantly lower than any previous reports, within a clinically relevant incubation time window. These results are highly relevant to the eventual human application in which the nanoclusters and chemotherapy drugs will be directly instilled in bladder via urinary catheter.

Synthesis, Optical Properties, and Sensing Applications of LaF3:Yb3+/Er3+/Ho3+/Tm3+ Upconversion Nanoparticles.

Project description:Herein, we successfully synthesized a series of LaF3:Yb3+/Er3+/Ho3+/Tm3+ upconversion nanoparticles (UCNPs) and LaF3:Yb3+0.20, Er3+0.02@LaF3:Yb3+0.20 core/shell UCNPs by modifying the amount of NaOH and the reaction time. Hexagonal LaF3 nanocrystals with uniform particle sizes and bright UC emissions were obtained. The crystal structures of the lanthanide-doped LaF3 UCNPs were investigated using wide-angle X-ray diffraction. The morphologies and particle sizes of the nanocrystals were determined using transmission electron microscopy. The photoluminescence (PL) spectra of the LaF3 nanocrystals could be tuned by altering the doping ratio of Er3+, Ho3+, and Tm3+. In addition, the PL intensities increased after coating the UCNP cores with an active shell. The fluorescence intensities of the UCNPs synthesized via a one-hour reaction with the addition of 2.5 or 5 mmol NaOH increased by up to 17 times compared with the sample prepared without the addition of NaOH. By modifying the doping ratio of Yb/Tm, UV-emissive LaF3 nanocrystals were obtained. After surface modification by ligand exchange, the hydrophobic LaF3:Yb3+0.20, Er3+0.02@LaF3:Yb3+0.20 core/shell UCNPs became water-dispersible. These colloid UCNPs could be utilized as a fluorescent probe for the detection of Hg2+ ions under 980 nm near-infrared irradiation.

Local Overheating of Biotissue Labeled With Upconversion Nanoparticles Under Yb3+ Resonance Excitation.

Project description:Local overheating of biotissue is a critical step for biomedical applications, such as photothermal therapy, enhancement of vascular permeability, remote control of drug release, and so on. Overheating of biological tissue when exposed to light is usually realized by utilizing the materials with a high-absorption cross section (gold, silica, carbon nanoparticles, etc.). Here, we demonstrate core/shell NaYF4:Yb3+, Tm3+/NaYF4 upconversion nanoparticles (UCNPs) commonly used for bioimaging as promising near-infrared (NIR) absorbers for local overheating of biotissue. We assume that achievable temperature of tissue labeled with nanoparticles is high enough because of Yb3+ resonance absorption of NIR radiation, whereas the use of auxiliary light-absorbing materials or shells is optional for photothermal therapy. For this purpose, a computational model of tissue heating based on the energy balance equations was developed and verified with the experimentally obtained thermal-graphic maps of a mouse in response to the 975-nm laser irradiation. Labeling of biotissue with UCNPs was found to increase the local temperature up to 2°C compared to that of the non-labeled area under the laser intensity lower than 1 W/cm2. The cellular response to the UCNP-initiated hyperthermia at subcritical ablation temperatures (lower than 42°C) was demonstrated by measuring the heat shock protein overexpression. This indicates that the absorption cross section of Yb3+ in UCNPs is relatively large, and microscopic temperature of nanoparticles exceeds the integral tissue temperature. In summary, a new approach based on the use of UCNP without any additional NIR absorbers was used to demonstrate a simple approach in the development of photoluminescent probes for simultaneous bioimaging and local hyperthermia.

The Upconversion Luminescence of Er3+/Yb3+/Nd3+ Triply-Doped β-NaYF₄ Nanocrystals under 808-nm Excitation.

Project description:In this paper, Nd3+-Yb3+-Er3+-doped β-NaYF₄ nanocrystals with different Nd3+ concentrations are synthesized, and the luminescence properties of the upconversion nanoparticles (UCNPs) have been studied under 808-nm excitation for sensitive biological applications. The upconversion luminescence spectra of NaYF₄ nanoparticles with different dopants under 808-nm excitation proves that the Nd3+ ion can absorb the photons effectively, and the Yb3+ ion can play the role of an energy-transfer bridging ion between the Nd3+ ion and Er3+ ion. To investigate the effect of the Nd3+ ion, the decay curves of the ⁴S3/2 → ⁴I15/2 transition at 540 nm are measured and analyzed. The NaYF₄: 20% Yb3+, 2% Er3+, 0.5% Nd3+ nanocrystals have the highest emission intensity among all samples under 808-nm excitation. The UC (upconversion) mechanism under 808-nm excitation is discussed in terms of the experimental results.

Triple-doped KMnF3:Yb3+/Er3+/Tm3+ nanocubes: four-color upconversion emissions with strong red and near-infrared bands.

Project description:Triple-doped (Yb(3+)/Er(3+)/Tm(3+)) KMnF3 nanocubes with uniform sizes of 250 nm were synthesized by a facile hydrothermal route using the oleic acid as the capping agent. It was found that these nanocubes can simultaneously exhibited four-color (blue, green, red and NIR) upconversion emissions under a single 980 nm near-infrared (NIR) laser excitation, which should have potential multicolor in vivo imaging applications. Specifically, the red (660 nm) and NIR (800 nm) peaks, known as two "optical windows" for imaging biological tissues, were strong. The spectral and pump analyses indicated the two-photon processes were responsible for the both red and NIR emissions.
