Project description:Fourier transforms, integer and fractional, are ubiquitous mathematical tools in basic and applied science. Certainly, since the ordinary Fourier transform is merely a particular case of a continuous set of fractional Fourier domains, every property and application of the ordinary Fourier transform becomes a special case of the fractional Fourier transform. Despite the great practical importance of the discrete Fourier transform, implementation of fractional orders of the corresponding discrete operation has been elusive. Here we report classical and quantum optical realizations of the discrete fractional Fourier transform. In the context of classical optics, we implement discrete fractional Fourier transforms of exemplary wave functions and experimentally demonstrate the shift theorem. Moreover, we apply this approach in the quantum realm to Fourier transform separable and path-entangled biphoton wave functions. The proposed approach is versatile and could find applications in various fields where Fourier transforms are essential tools.

Project description:The MRI image is obtained in the spatial domain from the given Fourier coefficients in the frequency domain. It is costly to obtain the high resolution image because it requires higher frequency Fourier data while the lower frequency Fourier data is less costly and effective if the image is smooth. However, the Gibbs ringing, if existent, prevails with the lower frequency Fourier data. We propose an efficient and accurate local reconstruction method with the lower frequency Fourier data that yields sharp image profile near the local edge. The proposed method utilizes only the small number of image data in the local area. Thus the method is efficient. Furthermore the method is accurate because it minimizes the global effects on the reconstruction near the weak edges shown in many other global methods for which all the image data is used for the reconstruction. To utilize the Fourier method locally based on the local non-periodic data, the proposed method is based on the Fourier continuation method. This work is an extension of our previous 1D Fourier domain decomposition method to 2D Fourier data. The proposed method first divides the MRI image in the spatial domain into many subdomains and applies the Fourier continuation method for the smooth periodic extension of the subdomain of interest. Then the proposed method reconstructs the local image based on L2 minimization regularized by the L1 norm of edge sparsity to sharpen the image near edges. Our numerical results suggest that the proposed method should be utilized in dimension-by-dimension manner instead of in a global manner for both the quality of the reconstruction and computational efficiency. The numerical results show that the proposed method is effective when the local reconstruction is sought and that the solution is free of Gibbs oscillations.

Project description:Active illumination microscopy (AIM) is a method of redistributing dynamic range in a scanning microscope using real-time feedback to control illumination power on a sub-pixel time scale. We describe and demonstrate a fully integrated instrument that performs both feedback and image reconstruction. The image is reconstructed on a logarithmic scale to accommodate the dynamic range benefits of AIM in a single output channel. A theoretical and computational analysis of the influence of noise on active illumination feedback is presented, along with imaging examples illustrating the benefits of AIM. While AIM is applicable to any type of scanning microscope, we apply it here specifically to two-photon microscopy.

Project description:Fourier-transformed alternating current voltammetry (FTacV) is a technique utilizing a combination of a periodic (frequently sinusoidal) oscillation superimposed onto a staircase or linear potential ramp. The advanced utilization of a large amplitude sine wave induces substantial nonlinear current responses. Subsequent filter processing (via Fourier-transformation, band selection, followed by inverse Fourier-transformation) generates a series of harmonics in which rapid electron transfer processes may be separated from non-Faradaic and competing electron transfer processes with slower kinetics. Thus, FTacV enables the isolation of current associated with redox processes under experimental conditions that would not generate meaningful data using direct current voltammetry (dcV). In this study, the enhanced experimental sensitivity and selectivity of FTacV versus dcV are illustrated in measurements that (i) separate the Faradaic current from background current contributions, (ii) use a low (5 μM) concentration of analyte (exemplified with ferrocene), and (iii) enable discrimination of the reversible [Ru(NH3)6]3+/2+ electron-transfer process from the irreversible reduction of oxygen under a standard atmosphere, negating the requirement for inert gas conditions. The simple, homebuilt check-cell described ensures that modern instruments can be checked for their ability to perform valid FTacV experiments. Detailed analysis methods and open-source data sets that accompany this work are intended to facilitate other researchers in the integration of FTacV into their everyday electrochemical methodological toolkit.

Project description:The role of splicing dysregulation in cancer is underscored by splicing factor mutations; however, its impact in the absence of such rare mutations is poorly understood. To reveal complex patient subtypes and putative regulators of pathogenic splicing in Acute Myeloid Leukemia (AML), we developed a new approach called OncoSplice. Among diverse new subtypes, OncoSplice identified a biphasic poor prognosis signature that partially phenocopies U2AF1-mutant splicing, impacting thousands of genes in over 40% of adult and pediatric AML cases. U2AF1-like splicing co-opted a healthy circadian splicing program, was stable over time and induced a leukemia stem cell (LSC) program. Pharmacological inhibition of the implicated U2AF1-like splicing regulator, PRMT5, rescued leukemia mis-splicing and inhibited leukemic cell growth. Genetic deletion of IRAK4, a common target of U2AF1-like and PRMT5 treated cells, blocked leukemia development in xenograft models and induced differentiation. These analyses reveal a new prognostic alternative-splicing mechanism in malignancy, independent of splicing-factor mutations.

Project description:Exact data of an electric circuit (EC) model of RLC (resistor, inductor, capacitor) elements representing rational immittance of LTI (linear, time invariant) systems are numerically Fourier transformed to demonstrate within error bounds applicability of the Hilbert integral tranform (HT) and Kramers-Kronig (KK) integral tranform (KKT) method. Immittance spectroscopy (IS) data are validated for their HT (KKT) compliance using non-equispaced fast Fourier transformation (NFFT) computations. Failing of HT (KKT) testing may not only stem from non-compliance with causality, stability and linearity which are readily distinguished using anti HT (KKT) relations. It could also indicate violation of uniform boundedness to be overcome either by using singly or multiply subtracted KK transform (SSKK or MSKK) or by seeking KKT of the same set of data at a complementary immit- tance level. Experimental IS data of a fuel cell (FC) are also numerically HT (KKT) validated by NFFT assessing whether LTI principles are met. Figures of merit are suggested to measure success in numerical validation of IS data.

Project description:Background and purposeFall accidents are a social challenge in Korea and elsewhere. Most previous studies have focused on the effects of reduced visual acuity due to myopia on falls and body balance. The objective of this study was to investigate whether uncorrected hyperopia was a major risk factor for falls and to establish whether the risk of falls was absolutely correlated with visual acuity.MethodsFifty-one young subjects with a mean age of 22.75 ± 2.13 years were enrolled in this study. To induce hyperopic and myopic refractive errors, spherical lenses of ±1.0-6.0 D (1.0 D stepwise) were used. Under each induced condition, fall risk index and sway power were assessed via Fourier transformation of postural sway using a TETRAX system.ResultsThe fall risk index for eyes-closed was significantly greater than that of eyes-open with full correction (t = -5.876, p < 0.05). The fall risk index increased significantly from hyperopia induced with -4.0 D lenses (with visual acuity of 0.69 ± 0.32) compared to eyes-open with full correction (F = 3.213, p < 0.05). However, there was no significant change in the induced myopia conditions, despite a drastic decline in decimal visual acuity. Sway power increased significantly in the low-to-medium frequency band derived from the peripheral vestibular system when hyperopia was induced. A significant difference was detected in hyperopia induced with -6.0 D lenses compared to eyes-open with full correction (F = 4.981, p = 0.017).ConclusionAn uncorrected hyperopia rather than myopia may increase the risk of falls, although eyes may show normal visual acuity due to the inherent accommodation mechanism. Our findings suggest that the corrected state of refractive errors is more important than the level of visual acuity as the criteria for appropriate visual input, which contributes to stable posture. Therefore, clinicians should consider the refractive condition, especially the characteristics of hyperopia, when analyzing body balance, and appropriate correction of uncorrected hyperopia to prevent falls.

Project description:To optimally assess oscillatory phenomena within physiological variables, spectral domain transforms are used. A discrete Fourier transform (DFT) is one of the most common methods used to attain this spectral change. In traumatic brain injury (TBI), a DFT is used to derive more complicated methods of physiological assessment, particularly that of cerebrovascular reactivity (CVR). However, a practical application of a DFT will introduce various errors that need to be considered. This study will evaluate the pulse amplitude DFT derivation of intracranial pressure (AMP) to highlight how slight differences in DFT methodologies can impact calculations. Utilizing a high-frequency prospectively maintained data set of TBI patients with recorded arterial and intracranial blood pressure, various cerebral physiological aspects of interest were assessed using the DFT windowing methods of rectangular, Hanning, and Chebyshev. These included AMP, CVR indices (including the pressure reactivity and pulse amplitude index), and the optimal cerebral perfusion pressure (with all methods of CVR). The results of the different DFT-derived windowing methods were compared using the Wilcoxon signed-ranked test and histogram plots between individual patients and over the whole 100-patient cohort. The results for this analysis demonstrate that, overall and for grand average values, there were limited differences between the different DFT windowing techniques. However, there were individual patient outliers to whom the different methods resulted in noticeably different overall values. From this information, for derived indices utilizing a DFT in the assessment of AMP, there are limited differences within the resulting calculations for larger aggregates of data. However, when the amplitude of spectrally resolved response is important and needs to be robust in smaller moments in time, it is recommended to use a window that has amplitude accuracy (such as Chebyshev or flat-top).

Project description:There are numerous statistical models used to identify individuals at high risk of cancer due to inherited mutations. Mendelian models predict future risk of cancer by using family history with estimated cancer penetrances (age- and sex-specific risk of cancer given the genotype of the mutations) and mutation prevalences. However, there is often residual risk heterogeneity across families even after accounting for the mutations in the model, due to environmental or unobserved genetic risk factors. We aim to improve Mendelian risk prediction by incorporating a frailty model that contains a family-specific frailty vector, impacting the cancer hazard function, to account for this heterogeneity. We use a discrete uniform population frailty distribution and implement a marginalized approach that averages each family's risk predictions over the family's frailty distribution. We apply the proposed approach to improve breast cancer prediction in BRCAPRO, a Mendelian model that accounts for inherited mutations in the BRCA1 and BRCA2 genes to predict breast and ovarian cancer. We evaluate the proposed model's performance in simulations and real data from the Cancer Genetics Network and show improvements in model calibration and discrimination. We also discuss alternative approaches for incorporating frailties and their strengths and limitations.

Project description:BackgroundThe detection of wheezes as an exacerbation sign is important in certain respiratory diseases. However, few highly accurate clinical methods are available for automatic detection of wheezes in children. This study aimed to develop a wheeze detection algorithm for practical implementation in children.MethodsA wheeze recognition algorithm was developed based on wheezes features following the Computerized Respiratory Sound Analysis guidelines. Wheezes can be detected by auscultation with a stethoscope and using an automatic computerized lung sound analysis. Lung sounds were recorded for 30 s in 214 children aged 2 months to 12 years and 11 months in a pediatric consultation room. Files containing recorded lung sounds were assessed by two specialist physicians and divided into two groups: 65 were designated as "wheeze" files, and 149 were designated as "no-wheeze" files. All lung sound judgments were agreed between two specialist physicians. We compared wheeze recognition between the specialist physicians and using the wheeze recognition algorithm and calculated the sensitivity, specificity, positive predictive value, and negative predictive value for all recorded sound files to evaluate the influence of age on the wheeze detection sensitivity.ResultsThe detection of wheezes was not influenced by age. In all files, wheezes were differentiated from noise using the wheeze recognition algorithm. The sensitivity, specificity, positive predictive value, and negative predictive value of the wheeze recognition algorithm were 100%, 95.7%, 90.3%, and 100%, respectively.ConclusionsThe wheeze recognition algorithm could identify wheezes in sound files and therefore may be useful in the practical implementation of respiratory illness management at home using properly developed devices.
