Antibacterial activity of the alkaloid-enriched extract from Prosopis juliflora pods and its influence on in vitro ruminal digestion.

ABSTRACT: The purpose of this study was to assess the in vitro antimicrobial activity of alkaloid-enriched extracts from Prosopis juliflora (Fabaceae) pods in order to evaluate them as feed additives for ruminants. As only the basic chloroformic extract (BCE), whose main constituents were juliprosopine (juliflorine), prosoflorine and juliprosine, showed Gram-positive antibacterial activity against Micrococcus luteus (MIC = 25 ?g/mL), Staphylococcus aureus (MIC = 50 ?g/mL) and Streptococcus mutans (MIC = 50 ?g/mL), its influence on ruminal digestion was evaluated using a semi-automated in vitro gas production technique, with monensin as the positive control. Results showed that BCE has decreased gas production as efficiently as monensin after 36 h of fermentation, revealing its positive influence on gas production during ruminal digestion. Since P. juliflora is a very affordable plant, this study points out this alkaloid enriched extract from the pods of Prosopis juliflora as a potential feed additive to decrease gas production during ruminal digestion.

Project description:BackgroundThe Ancient system of medicine showed the limelight on the use of herbal remedies and was found to possess minimal side effects and acceptable therapeutic outcomes. In this context, Prosopis juliflora gained importance in managing chronic diseases such as cancer, dermatological diseases, and chronic inflammatory disorders. Hence, P. juliflora was selected for further investigation associated with diabetes and inflammation.AimThe present study aimed to evaluate the anti-diabetic activity in chemically induced experimental rats and explore the nature of phytocomponents that may produce this activity.MethodsExperimentally, diabetes was induced by a single administration of streptozotocin at 50 mg/kg intraperitoneally in Wistar rats. The animals were treated orally with P. juliflora at low and high doses (200 and 400 mg/kg) for 10 days. Blood collected from the retro-orbital plexus was analyzed for parameters like blood glucose levels, insulin, adiponectin, Keap1 and Nrf2. PPAR-γ, AMPK and GLUT 2 levels were analyzed in the pancreatic tissue. Besides, at the end of the experiment, animals were sacrificed, and the pancreatic tissue sections were subjected for histopathological, morphometrical and immune histochemical exploration. The phytochemical composition of the plant was investigated by GC-MS.ResultsThe administration of P. juliflora higher dose showed a significant decrease (**p< 0.001) in blood glucose levels with a rise in adiponectin, PPARγ, Keap1, Nrf2, Glut 2, and AMPK significantly (**p< 0.001). The inflammatory cytokine TNFα was also estimated and was found to be lowered significantly (**p< 0.001) in test drug-treated animals. Furthermore, in the pancreatic tissue, the number of Islets, the area, and the number of β-cells were improved significantly with the sub-chronic treatment of P. juliflora extract. The structure and function of β-cells were also revamped.ConclusionThe study results demonstrated a significant effect of P. juliflora on glycemic status, inflammatory condition, and the architecture of pancreatic tissue. In the identification and isolation process by GC MS, it was noticed that P. juliflora contained few phytochemical constituents from which it might be considered a promising drug for type 2 diabetes mellitus.

Project description:This study aimed to assess community perceptions towards invasion of Prosopis juliflora, utilization, and its control options in Afar region, Northern Ethiopia. Using purposive sampling and stratified random methods, 20 members of key informants and 154 households from four sites of Awash Fentale and Amibara Districts were selected. For data analysis, we used Kruskal Wallis non-parametric tests of K independent samples. About 30% of respondents in Amibara and 29% in Awash Fentale reported that Prosopis juliflora was largely introduced into their landscape by livestock. It showed that 29% of the respondents in Awash Fentale and 41% in Amibara responded that Prosopis juliflora largely invaded and affected rangelands. Morevover, about 1% of respondents in Awash Fentale and 14% in Amibara argued that Prosopis juliflora hindered movements of livestock. In addition, 30% of respondents in Amibara and 29% in Awash Fentale believe that Prosopis juliflora was largely dispersed by livestock. It showed that 20% of households in Awash Fentale and 41% in Amibara have the notion that Prosopis juliflora majorly impacted rangelands. Whereas 1.3% of respondents in Awash Fentale and 14% in Amibara argued that Prosopis juliflora have hampered the movement of livestock. Thus, the afromentioned findings are implications for management of rangelands. With regard to the control of Prosopis juliflora invasions, 12% of respondents in Awash Fentale and 33% in Amibara District tried control its expansion by fire. About 10% of respondents in Awash Fentale and 9% in Amibara district managed Prosopis juliflora expansion by its utilization, whereas, in Awash Fentale (11%) and Amibara (8%) households indicated that invasion of Prosopis juliflora could be controlled by mechanical methods. It is advisable to do some managerial work to reverse these impacts as perceived by local communities in the study area to avert the aggressive proliferation of Prosopis juliflora in the region.
