ABSTRACT: The reaction of enamines and bromoquinones in the presence of copper(II) acetate and potassium carbonate results in a regiospecific synthesis of indolequinones. Copper-mediated synthesis of drug-like bicyclopentanes.

Project description:Multicomponent reactions are relied on in both academic and industrial synthetic organic chemistry owing to their step- and atom-economy advantages over traditional synthetic sequences1. Recently, bicyclo[1.1.1]pentane (BCP) motifs have become valuable as pharmaceutical bioisosteres of benzene rings, and in particular 1,3-disubstituted BCP moieties have become widely adopted in medicinal chemistry as para-phenyl ring replacements2. These structures are often generated from [1.1.1]propellane via opening of the internal C-C bond through the addition of either radicals or metal-based nucleophiles3-13. The resulting propellane-addition adducts are then transformed to the requisite polysubstituted BCP compounds via a range of synthetic sequences that traditionally involve multiple chemical steps. Although this approach has been effective so far, a multicomponent reaction that enables single-step access to complex and diverse polysubstituted drug-like BCP products would be more time efficient compared to current stepwise approaches. Here we report a one-step three-component radical coupling of [1.1.1]propellane to afford diverse functionalized bicyclopentanes using various radical precursors and heteroatom nucleophiles via a metallaphotoredox catalysis protocol. This copper-mediated reaction operates on short timescales (five minutes to one hour) across multiple (more than ten) nucleophile classes and can accommodate a diverse array of radical precursors, including those that generate alkyl, α-acyl, trifluoromethyl and sulfonyl radicals. This method has been used to rapidly prepare BCP analogues of known pharmaceuticals, one of which is substantially more metabolically stable than its commercial progenitor.

Synthesis of Oxazolidinone and Tosyl Enamines by Tertiary Amine Catalysis.

Project description:A procedure for the synthesis of oxazolidinone and tosyl enamines is reported. Alkynoyl oxazolidinones and tosyl imides undergo reaction to form enamines in the presence of catalytic amounts of tertiary amines. The data suggest that an amide anion is formed during the reaction, which undergoes conjugate addition to form the final product.

Copper(II)-Mediated Iodination of 1-Nitroso-2-naphthol.

Project description:The 3-Iodo-1-nitrosonaphthalene-2-ol (I-NON) was obtained by the copper(II)-mediated iodination of 1-nitroso-2-naphthol (NON). The suitable reactants and optimized reaction conditions, providing 94% NMR yield of I-NON, included the usage of Cu(OAc)2·H2O and 1:2:8 CuII/NON/I2 molar ratio between the reactants. The obtained I-NON was characterized by elemental analyses (C, H, N), high-resolution ESI+-MS, 1H and 13C{1H} NMR, FTIR, UV-vis spectroscopy, TGA, and X-ray crystallography (XRD). The copper(II) complexes bearing deprotonated I-NON were prepared as follows: cis-[Cu(I-NON-H)(I-NON)](I3) (1) was obtained by the reaction between Cu(NON-H)2 and I2 in CHCl3/MeOH, while trans-[Cu(I-NON-H)2] (2) was synthesized from I-NON and Cu(OAc)2 in MeOH. Crystals of trans-[Cu(I-NON-H)2(THF)2] (3) and trans-[Cu(I-NON-H)2(Py)2] (4) were precipitated from solutions of 2 in CHCl3/THF and Py/CHCl3/MeOH mixtures, respectively. The structures of 1 and 3-4 were additionally verified by X-ray crystallography. The characteristic feature of the structures of 1 and 3 is the presence of intermolecular halogen bonds with the involvement of the iodine center of the metal-bound deprotonated I-NON. The nature of the I···I and I···O contacts in the structures of 1 and 3, correspondingly, were studied theoretically at the DFT (PBE0-D3BJ) level using the QTAIM, ESP, ELF, NBO, and IGM methods.

Continuous Flow Copper Laser Ablation Synthesis of Copper(I and II) Oxide Nanoparticles in Water.

Project description:Copper(I) oxide (Cu2O) nanoparticles (NPs) are selectively prepared in high yields under continuous flow in a vortex fluidic device (VFD), involving irradiation of a copper rod using a pulsed laser operating at 1064 nm and 600 mJ. The plasma plume generated inside a glass tube (20 mm O.D.), which is rapidly rotating (7.5 k rpm), reacts with the enclosed air in the microfluidic platform, with then high mass transfer of material into the dynamic thin film of water passing up the tube. The average size of the generated Cu2ONPs is 14 nm, and they are converted to copper(II) oxide (CuO) nanoparticles with an average diameter of 11 nm by heating the as-prepared solution of Cu2ONPs in air at 50 °C for 10 h.

Mechanism of alcohol oxidation mediated by copper(II) and nitroxyl radicals.

Project description:2,2'-Bipyridine-ligated copper complexes, in combination with TEMPO (2,2,6,6-tetramethylpiperidine-N-oxyl), are highly effective catalysts for aerobic alcohol oxidation. Considerable uncertainty and debate exist over the mechanism of alcohol oxidation mediated by Cu(II) and TEMPO. Here, we report experimental and density functional theory (DFT) computational studies that distinguish among numerous previously proposed mechanistic pathways. Oxidation of various classes of radical-probe substrates shows that long-lived radicals are not formed in the reaction. DFT computational studies support this conclusion. A bimolecular pathway involving hydrogen-atom-transfer from a Cu(II)-alkoxide to a nitroxyl radical is higher in energy than hydrogen transfer from a Cu(II)-alkoxide to a coordinated nitroxyl species. The data presented here reconcile a collection of diverse and seemingly contradictory experimental and computational data reported previously in the literature. The resulting Oppenauer-like reaction pathway further explains experimental trends in the relative reactivity of different classes of alcohols (benzylic versus aliphatic and primary versus secondary), as well as the different reactivity observed between TEMPO and bicyclic nitroxyls, such as ABNO (ABNO = 9-azabicyclo[3.3.1]nonane N-oxyl).

Removal of Copper (II) by Biochar Mediated by Dissolved Organic Matter.

Project description:The effects of humic acid (HA) and fulvic acid (FA) on Cu2+ adsorption on biochar were investigated, with mechanisms confirmed by excitation-emission matrix spectroscopy, Fourier transform infrared spectroscopy, and scanning electron microscopy. HA loading enhanced Cu2+ adsorption on biochar, with the maximum enhancement of 55.0% occurring at an HA loading of 100?mg-C/L. The adsorbed HA introduced many additional functional groups to biochar, thus enhancing Cu2+ adsorption, which decreased at HA concentrations >100?mg-C/L due to self-association of HA at high loading concentrations. In contrast, FA loading caused no enhancement on Cu2+ adsorption on biochar. FA was adsorbed through H-bonding with the functional groups of biochar, which set up a competition with Cu2+ for adsorption on biochar. The functional groups occupied by adsorbed FA were offset by the newly introduced functional groups of FA, thus there was no net increase in the amount of Cu2+ adsorption upon FA loading. These findings imply that, because of the enhanced adsorption of HA-loaded biochar, the amount of Cu2+ immobilized would increase by 28.2% for mature compost and 31.9% for fresh compost if there exist interaction between biochar and HA compared with the amounts immobilized by non-interactive HA and biochar.

Mechanism for Copper(II)-Mediated Disaggregation of a Porphyrin J-Aggregate.

Project description:J-aggregates of anionic meso-tetrakis(4-sulfonatophenyl)porphyrin form at intermediate pH (2.3-3.1) in the presence of NiSO4 or ZnSO4 (ionic strength, I.S. = 3.2 M). These aggregates convert to monomeric porphyrin units via metallation with copper(II) ions. The kinetics for the disassembly process, as monitored by UV/vis spectroscopy, exhibits zeroth-order behavior. The observed zeroth-order rate constants show a two-term dependence on copper(II) ion concentrations: linear and second order. Also observed is an inverse dependence on hydrogen ion concentration. Activation parameters have been determined for the disassembly process leading to ΔH ≠ = (+163 ± 15) kJ·mol-1 and ΔS ≠ = (+136 ± 11) J·K-1. A mechanism is proposed in which copper(II) cation is in pre-equilibrium with a reactive site at the rim of the J-aggregate. An intermediate copper species is thus formed that eventually leads to the final metallated porphyrin either through an assisted attack of a second metal ion or through a direct insertion of the metal cation into the macrocycle core.

Stereoselective synthesis of ?-hydroxy enamines, aminocyclopropanes, and 1,3-amino alcohols via asymmetric catalysis.

Project description:Tandem methods for the catalytic asymmetric preparation of enantioenriched ?-hydroxy (E)-enamines and aminocyclopropanes are presented. The diastereoselective hydrogenation of enantioenriched (E)-trisubstituted hydroxy enamines to generate 1,2-disubstituted-1,3-amino alcohols is also outlined. These methods are initiated by highly regioselective hydroboration of N-tosyl-substituted ynamides with diethylborane to generate ?-amino alkenyl boranes. In situ boron-to-zinc transmetalation generates ?-amino alkenylzinc reagents. These functionalized vinylzinc intermediates are subsequently added to aldehydes in the presence of a catalyst derived from an enantioenriched amino alcohol (morpholino isoborneol, MIB). The catalyst promotes highly enantioselective C-C bond formation to provide ?-hydroxy enamines in good isolated yields (68-86%) with 54-98% enantioselectivity. The intermediate zinc ?-alkoxy enamines can be subjected to a tandem cyclopropanation to afford aminocyclopropyl carbinols with three continuous stereocenters in a one-pot procedure with good yields (72-82%), enantioselectivities of 76-94%, and >20:1 diastereomeric ratios. Diastereoselective hydrogenation of isolated enantioenriched ?-hydroxy enamines over Pd/C furnished syn-1,2-disubstituted-1,3-amino alcohols in high yields (82-90%) with moderate to excellent diastereoselectivities. These methods were used in an efficient preparation of the enantioenriched precursor to PRC200-SS derivatives, which are potent serotonin-norepinephrine-dopamine reuptake inhibitors.
