New Volvovirus Isolates from Acheta domesticus (Japan) and Gryllus assimilis (United States).

ABSTRACT: A novel circular single-stranded DNA (ssDNA) virus, volvovirus, from the house cricket has been described recently. Here, we report the isolation of volvoviruses from Acheta domesticus in Japan and Gryllus assimilis in the United States. These Acheta domesticus volvovirus (AdVVV) isolates have genomes of 2,517 and 2,516 nucleotides (nt) and 4 large open reading frames (ORFs).

The Bacterial Microbiota of Edible Insects <i>Acheta domesticus</i> and <i>Gryllus assimilis</i> Revealed by High Content Analysis.

Project description:In the concept of novel food, insects reared under controlled conditions are considered mini livestock. Mass-reared edible insect production is an economically and ecologically beneficial alternative to conventional meat gain. Regarding food safety, insect origin ingredients must comply with food microbial requirements. House crickets (Acheta domesticus) and Jamaican field crickets (Gryllus assimilis) are preferred insect species that are used commercially as food. In this study, we examined cricket-associated bacterial communities using amplicon-based sequencing of the 16S ribosomal RNA gene region (V3-V4). The high taxonomic richness of the bacterial populations inhabiting both tested cricket species was revealed. According to the analysis of alpha and beta diversity, house crickets and Jamaican field crickets displayed significantly different bacterial communities. Investigation of bacterial amplicon sequence variants (ASVs) diversity revealed cricket species as well as surface and entire body-associated bacterial assemblages. The efficiency of crickets processing and microbial safety were evaluated based on viable bacterial counts and identified bacterial species. Among the microorganisms inhabiting both tested cricket species, the potentially pathogenic bacteria are documented. Some bacteria representing identified genera are inhabitants of the gastrointestinal tract of animals and humans, forming a normal intestinal microflora and performing beneficial probiotic functions. The novel information on the edible insect-associated microbiota will contribute to developing strategies for cricket processing to avoid bacteria-caused risks and reap the benefits.

Comparative Genomic Analysis of Acheta domesticus Densovirus Isolates from Different Outbreaks in Europe, North America, and Japan.

Project description:The first densovirus from a cricket, Acheta domesticus densovirus (AdDNV) (Parvoviridae), was isolated in Europe in 1977 and has been studied previously. We compared seven additional AdDNV genomes isolated from 4 other European outbreaks, 2 major North American outbreaks, and a Japanese outbreak. Phylogenetic analysis suggested that the 2009 Japanese and North American outbreaks were not related.

Project description:Edible insect rearing could provide one alternative for protein production by having a smaller environmental impact than traditional livestock farming due to insects' ability to convert organic side streams. Currently, the insect rearing industry utilizes soybeans as a major source of protein in the feeds. Protein-rich by-products of food industry could be used to replace them in insect feeds, but it is not known if they also meet the insects' nutritional requirements. Our study evaluated the growth performance of two widely used edible cricket species, Acheta domesticus and Gryllus bimaculatus (Orthoptera: Gryllidae), on 18 experimental diets. The experimental diets included commercial chicken feeds and cricket diets, where soybean was partly and completely replaced with by-products from food industry: potato protein, barley mash, barley feed, compressed leftover of turnip rape and mix of broad bean and pea on three levels of protein. We found that the high- and medium-protein turnip rape and barley mash diets produced the highest yield and an increase in all performance variables. Overall, the high- and medium-protein diets produced the highest yield, growth and fastest development. Our results showed that by-products of food industry could be utilized as a part of the cricket feeds and thus advance the goals of circular economy.

Cricket antennae shorten when bending (Acheta domesticus L.).

Project description:Insect antennae are important mechanosensory and chemosensory organs. Insect appendages, such as antennae, are encased in a cuticular exoskeleton and are thought to bend only between segments or subsegments where the cuticle is thinner, more flexible, or bent into a fold. There is a growing appreciation of the dominating influence of folds in the mechanical behavior of a structure, and the bending of cricket antennae was considered in this context. Antennae will bend or deflect in response to forces, and the resulting bending behavior will affect the sensory input of the antennae. In some cricket antennae, such as in those of Acheta domesticus, there are a large number (>100) of subsegments (flagellomeres) that vary in their length. We evaluated whether these antennae bend only at the joints between flagellomeres, which has always been assumed but not tested. In addition we questioned whether an antenna undergoes a length change as it bends, which would result from some patterns of joint deformation. Measurements using light microscopy and SEM were conducted on both male and female adult crickets (Acheta domesticus) with bending in four different directions: dorsal, ventral, medial, and lateral. Bending occurred only at the joints between flagellomeres, and antennae shortened a comparable amount during bending, regardless of sex or bending direction. The cuticular folds separating antennal flagellomeres are not very deep, and therefore as an antenna bends, the convex side (in tension) does not have a lot of slack cuticle to "unfold" and does not lengthen during bending. Simultaneously on the other side of the antenna, on the concave side in compression, there is an increasing overlap in the folded cuticle of the joints during bending. Antennal shortening during bending would prevent stretching of antennal nerves and may promote hemolymph exchange between the antenna and head.

Project description:The genome structure of Acheta domesticus mini ambidensovirus, isolated from crickets, resembled that of ambisense densoviruses from Lepidoptera but was 20% smaller. It had the highest (<25%) protein sequence identity with the nonstructural protein 1 (NS1) of Iteravirus and VP of Densovirus members (both with 25% coverage) and smaller (0.2- versus 0.55-kb) Y-shaped inverted terminal repeats.
