Seladonia (Pachyceble) henanensis sp. n. (Hymenoptera, Halictidae) from China.

ABSTRACT: Seladonia (Pachyceble) henanensis sp. n., is described from Henan Province, the eastern-central part of China. This species is separated from its allied species by a combination of the following morphological characters: head broad in female, inner hind tibial spur of female with 7-8 slender teeth, T1 basolaterally with appressed hair tuft in both sexes, and genitalia with long and large lower gonostylus in male. Important taxonomic characters are illustrated with photographs, scanning electron micrographs, and line drawings.

Project description:The available information about the cleptoparastic bees of the genus Sphecodes in the Palaearctic part of China is summarized. Twenty-four species are currently known from this area including 16 newly recorded. Based on type specimens, new synonymies have been proposed for Sphecodescristatus Hagens, 1882 = S.alfkeni Meyer, 1922, syn. n.; S.longulus Hagens, 1882 = S.subfasciatus Blüthgen, 1934, syn. n.; S.nippon Meyer, 1922 = S.kansuensis Blüthgen, 1934, syn. n.; Sphecodespieli Cockerell, 1931 = S.orientalis Astafurova & Proshchalykin, 2014, syn. n. Lectotypes are designated for Sphecodesalfkeni Meyer, 1922 and S.pellucidusniveipennis Meyer, 1925. Illustrated keys to males and females of all species known from Palaearctic China and an updated checklist of the 33 Chinese species of Sphecodes are provided.

The number of Chinese species of the subfamily Nomiinae and genus Lipotriches are updated to 47 and 15, respectively.

The complete mitogenome of Lasioglossum affine (Hymenoptera: Halictidae) and phylogenetic analysis.

Project description:The complete mitogenome of Lasioglossum affine (Hymenoptera: Halictidae) was sequenced and analyzed. The whole mitogenome is 17,352 bp (AT%=84.1%) and encodes 37 typical eukaryotic mitochondrial genes, including 13 protein-coding genes (PCGs), 22 tRNAs, two rRNAs, and an AT-rich region. Further analysis found three gene rearrangements, where trn I-Q-M → trn M-I-Q, trn W-C-Y → trn C-W-Y, and trn K-D → trn D-K were shuffled. The phylogenetic relationships of 19 species of Hymenoptera were established using maximum-likelihood method based on 13 concatenated PCGs. The result showed that Lasioglossum affine is a sister of Lasioglossum sp. SJW-2017.

The complete mitogenome of Nomia chalybeata (Hymenoptera: Halictidae) and phylogenetic analysis.

Project description:The complete mitogenome of Nomia chalybeata was sequenced. The mitochondrial length of N. chalybeata was 16,692 bp (AT content 85.4%), with 37 classic invertebrate mitochondrial genes (including 13 protein-coding genes, 22 transporter RNAs, and two ribosomal RNAs) and AT-rich region (AT content 91.7%). The maximum-likelihood phylogenetic relationship was constructed using 11 species from Hymenoptera. Through the phylogenetic relationship, our research team successfully used the molecular data of the mitochondrial genome to verify that N. chalybeata belongs to the family Halictidae, and also provides molecular data for the database of the family Halictidae.

Computational genome-wide survey of odorant receptors from two solitary bees Dufourea novaeangliae (Hymenoptera: Halictidae) and Habropoda laboriosa (Hymenoptera: Apidae).

Project description:Olfactory/odorant receptors (ORs) probably govern eusocial behaviour in honey bees through detection of cuticular hydrocarbons (CHCs) and queen mandibular gland pheromones (QMP). CHCs are involved in nest-mate recognition whereas QMP acts as sex pheromone for drones and as retinue pheromone for female workers. Further studies on the effect of eusociality on the evolution of ORs are hindered by the non-availability of comprehensive OR sets of solitary species. We report complete OR repertoires from two solitary bees Dufourea novaeangliae (112 ORs) and Habropoda laboriosa (151 ORs). We classify these ORs into 34 phylogenetic clades/subfamilies. Differences in the OR sets of solitary and eusocial bees are observed in individual subfamilies like subfamily 9-exon (putative CHC receptors) and L (contains putative QMP receptor group). A subfamily (H) including putative floral scent receptors is expanded in the generalist honey bees only, but not in the specialists. On the contrary, subfamily J is expanded in all bees irrespective of their degree of social complexity or food preferences. Finally, we show species-lineage specific and OR-subfamily specific differences in the putative cis-regulatory DNA motifs of the ORs from six hymenopteran species. Out of these, [A/G]CGCAAGCG[C/T] is a candidate master transcription factor binding site for multiple olfactory genes.

Identification of 24 new microsatellite loci in the sweat bee Lasioglossum malachurum (Hymenoptera: Halictidae).

Project description:ObjectiveThe objective here is to identify highly polymorphic microsatellite loci for the Palaearctic sweat bee Lasioglossum malachurum. Sweat bees (Hymenoptera: Halictidae) are widespread pollinators that exhibit an unusually large range of social behaviours from non-social, where each female nests alone, to eusocial, where a single queen reproduces while the other members of the colony help to rear her offspring. They thus represent excellent models for understanding social evolution.Results24 new microsatellite loci were successfully optimized. When amplified across 23-40 unrelated females, the number of alleles per locus ranged from 3 to 17 and the observed heterozygosities 0.45 to 0.95. Only one locus showed evidence of significant deviation from Hardy-Weinberg equilibrium. No evidence of linkage disequilibrium was found. These 24 loci will enable researchers to gain greater understanding of colony relationships within this species, an important model for the study of eusociality. Furthermore, 22 of the same loci were also successfully amplified in L. calceatum, suggesting that these loci may be useful for investigating the ecology and evolution of sweat bees in general.
