Project description:Freshwater availability is changing worldwide. Here we quantify 34 trends in terrestrial water storage observed by the Gravity Recovery and Climate Experiment (GRACE) satellites during 2002-2016 and categorize their drivers as natural interannual variability, unsustainable groundwater consumption, climate change or combinations thereof. Several of these trends had been lacking thorough investigation and attribution, including massive changes in northwestern China and the Okavango Delta. Others are consistent with climate model predictions. This observation-based assessment of how the world's water landscape is responding to human impacts and climate variations provides a blueprint for evaluating and predicting emerging threats to water and food security.

Project description:Emerging infectious diseases (EIDs) are a significant burden on global economies and public health. Their emergence is thought to be driven largely by socio-economic, environmental and ecological factors, but no comparative study has explicitly analysed these linkages to understand global temporal and spatial patterns of EIDs. Here we analyse a database of 335 EID 'events' (origins of EIDs) between 1940 and 2004, and demonstrate non-random global patterns. EID events have risen significantly over time after controlling for reporting bias, with their peak incidence (in the 1980s) concomitant with the HIV pandemic. EID events are dominated by zoonoses (60.3% of EIDs): the majority of these (71.8%) originate in wildlife (for example, severe acute respiratory virus, Ebola virus), and are increasing significantly over time. We find that 54.3% of EID events are caused by bacteria or rickettsia, reflecting a large number of drug-resistant microbes in our database. Our results confirm that EID origins are significantly correlated with socio-economic, environmental and ecological factors, and provide a basis for identifying regions where new EIDs are most likely to originate (emerging disease 'hotspots'). They also reveal a substantial risk of wildlife zoonotic and vector-borne EIDs originating at lower latitudes where reporting effort is low. We conclude that global resources to counter disease emergence are poorly allocated, with the majority of the scientific and surveillance effort focused on countries from where the next important EID is least likely to originate.

Project description:Recent satellite observations confirm that the Arctic is absorbing more solar radiation now than at the start of this century in response to declining Arctic sea ice and snow covers. Trends in the solar radiation input to Arctic ocean and land surfaces now each exceed interannual variability at the 95% confidence level, although all-sky trends have taken 20%-40% longer to emerge compared to clear-sky conditions. Clouds reduce mean solar absorption and secular trends over both land and ocean, but the effect of clouds on natural variability depends on the underlying surface. While clouds increase the time needed to unambiguously identify trends in nearly all Arctic regions, their masking effects are strongest over oceans. Clouds have extended the time to emergence of already observed clear-sky trends beyond the existing 21 years Clouds and Earth's Radiant Energy System record in half of eight Arctic seas, supporting the need for continued satellite-based radiative flux observations over the Arctic.

Project description:Although all nations in the America's face a common demographic reality of longevity, declining fertility rates and changes in family roles a growing body of research points to a dramatic demographic transformation in Mexico. Although Mexico's population is relatively young, with a median age of 27.9 in 2015, it will age rapidly in coming years, increasing to 42 years by 2050. The rapid median age in the nation also reflects the growing proportion of people 65 or older, and is expected to triple to 20.2% by 2050. This article examines how the age and gender structure of Mexico offers important insights about current and future political and social stability, as well as economic development. Mexico is the world's eleventh largest country in terms of population size and the "demographic dividend" of a large youthful population is giving way to a growing older population that will inevitably place demands on health care and social security. The shift in age structure will result in increased dependency of retirees on the working-age population in the next 20 years. Mexico does not provide universal coverage of social security benefits and less than half of the labor force is covered by any pension or retirement plan. As a result, elderly Mexicans often continue working into old age. The high total poverty rate in the country, especially among the older population magnifies the problem of the potential dependency burden. The article ends with a discussion of key public policy issues related to aging in Mexico.

Project description:Advances in omics and immunology over the past 20 years have revolutionized the approach to cancer prevention, with the goal now focused on identifying populations at higher risk for developing cancer in their lifetime as a result of either extensive exposure to environmental carcinogens or harboring precancer lesions or inherited genetic mutations that predispose them to specific types of cancer(s). Thus, the naïve idea that cancer could be "prevented" in the general population has evolved to a more practical approach based on the understanding that the target population for preventive agents will be individuals who already have alterations, in gene pathways, whether inherited or environmentally caused, and the goal will be to "intercept" these lesions at the earliest stages in the path from an initial genetic lesion to full-blown cancer. The Division of Cancer Prevention of the National Cancer Institute and the Office of Disease Prevention at the National Institutes of Health recently sponsored the second biennial "Translational Advances in Cancer Preventive Agent Development Meeting," held virtually from September 7-9th. In this Meeting Report, we highlight the scientific sessions of this meeting that covered the most recent advances in preventive agent development that also highlighted these rapidly emerging trends in this research area.

Project description:"Classical peer review" has been subject to intense criticism for slowing down the publication process, bias against specific categories of paper and author, unreliability, inability to detect errors and fraud, unethical practices, and the lack of recognition for unpaid reviewers. This paper surveys innovative forms of peer review that attempt to address these issues. Based on an initial literature review, we construct a sample of 82 channels of scientific communication covering all forms of review identified by the survey, and analyze the review mechanisms used by each channel. We identify two major trends: the rapidly expanding role of preprint servers (e.g., ArXiv) that dispense with traditional peer review altogether, and the growth of "non-selective review," focusing on papers' scientific quality rather than their perceived importance and novelty. Other potentially important developments include forms of "open review," which remove reviewer anonymity, and interactive review, as well as new mechanisms for post-publication review and out-of-channel reader commentary, especially critical commentary targeting high profile papers. One of the strongest findings of the survey is the persistence of major differences between the peer review processes used by different disciplines. None of these differences is likely to disappear in the foreseeable future. The most likely scenario for the coming years is thus continued diversification, in which different review mechanisms serve different author, reader, and publisher needs. Relatively little is known about the impact of these innovations on the problems they address. These are important questions for future quantitative research.

Project description:INTRODUCTION:Research on electronic cigarettes is an emerging field, with the number of articles in this field noted to have grown exponentially over recent years. We used a bibliometric analysis method (co-word analysis) to analyze the emerging trends and research hotspots in this field. METHODS:Publication data on electronic cigarettes from 2010 to 2018 were retrieved and downloaded from the PubMed database. Theme trends and knowledge structures were analyzed on the relevant research fields of electronic cigarettes by using a biclustering analysis, strategic diagram analysis, and social network analysis methods. Research hotspots were extracted and compared from three periods. RESULTS:Core topics that have continuously develop between the years 2010 and 2018 include: tobacco use cessation devices; tobacco products; tobacco use cessation devices/adverse effects; smoking prevention and adverse effects; electronic nicotine delivery systems/economics; and public health. Some currently undeveloped topics that could be considered as new future research directions include: tobacco use disorder/therapy; tobacco use disorder/epidemiology; students/psychology; students/statistics and numerical data; adolescent behavior/psychology; nicotine/toxicity; nicotinic agonists/administration and dosage; and electronic nicotine delivery systems/legislation and jurisprudence. CONCLUSIONS:Results suggest that some currently immature topics in strategic coordinates and emerging hotspots in social network graphs can be used as future research directions.

Project description:While promising, immunotherapy has yet to be fully unlocked for the preponderance of cancers where conventional chemoradiation reigns. This remains particularly evident in pediatric sarcomas where standard of care has not appreciably changed in decades. Importantly, pediatric bone sarcomas, like osteosarcoma and Ewing's sarcoma, possess unique tumor microenvironments driven by distinct molecular features, as do rhabdomyosarcomas and soft tissue sarcomas. A better understanding of each malignancy's biology, heterogeneity, and tumor microenvironment may lend new insights toward immunotherapeutic targets in novel platform technologies for cancer vaccines and adoptive cellular therapy. These advances may pave the way toward new treatments requisite for pediatric sarcomas and patients in need of new therapies.

Project description:Orodispersible films (ODFs) are thin, mechanically strong, and flexible polymeric films that are designed to dissolve or disintegrate rapidly in the oral cavity for local and/or systemic drug delivery. This review examines various aspects of ODFs and their potential as a drug delivery system. Recent advancements, including the detailed exploration of formulation components, such as polymers and plasticizers, are briefed. The review highlights the versatility of preparation methods, particularly the solvent-casting production process, and novel 3D printing techniques that bring inherent flexibility. Three-dimensional printing technology not only diversifies active compounds but also enables a multilayer approach, effectively segregating incompatible drugs. The integration of nanoparticles into ODF formulations marks a significant breakthrough, thus enhancing the efficiency of oral drug delivery and broadening the scope of the drugs amenable to this route. This review also sheds light on the diverse in vitro evaluation methods utilized to characterize ODFs, ongoing clinical trials, approved marketed products, and recent patents, providing a comprehensive outlook of the evolving landscape of orodispersible drug delivery. Current patient-centric approaches involve developing ODFs with patient-friendly attributes, such as improved taste masking, ease of administration, and enhanced patient compliance, along with the personalization of ODF formulations to meet individual patient needs. Investigating novel functional excipients with the potential to enhance the permeation of high-molecular-weight polar drugs, fragile proteins, and oligonucleotides is crucial for rapid progress in the advancing domain of orodispersible drug delivery.

Project description:Metalloproteins have long been recognized as key determinants of endogenous contrast in magnetic resonance imaging (MRI) of biological subjects. More recently, both natural and engineered metalloproteins have been harnessed as biotechnological tools to probe gene expression, enzyme activity, and analyte concentrations by MRI. Metalloprotein MRI probes are paramagnetic and function by analogous mechanisms to conventional gadolinium or iron oxide-based MRI contrast agents. Compared with synthetic agents, metalloproteins typically offer worse sensitivity, but the possibilities of using protein engineering and targeted gene expression approaches in conjunction with metalloprotein contrast agents are powerful and sometimes definitive strengths. This review summarizes theoretical and practical aspects of metalloprotein-based contrast agents, and discusses progress in the exploitation of these proteins for molecular imaging applications.
