A new species of the genus Arhynchite (Annelida, Echiura) from sandy flats of Japan, previously referred to as Thalassema owstoni Ikeda, 1904.

ABSTRACT: A new echiuran, Arhynchite hayaoi sp. n., is described from newly collected specimens from sandy flats of the Seto Inland Sea, Japan, together with many museum specimens, including those once identified as Thalassema owstoni Ikeda, 1904 or Arhynchite arhynchite (Ikeda, 1924). The new species is clearly distinguishable from its congeners by the smooth margin of gonostomal lips and lack of rectal caecum. Brief references are also made to the morphological distinction between the new species and Thalassema owstoni, originally described from the deep bottom on the Japanese Pacific coast.

Muscular Development in Urechis unicinctus (Echiura, Annelida).

Project description:Echiura is one of the most intriguing major subgroups of phylum Annelida because, unlike most other annelids, echiuran adults lack metameric body segmentation. Urechis unicinctus lives in U-shape burrows of soft sediments. Little is known about the molecular mechanisms underlying the development of U. unicinctus. Herein, we overviewed the developmental process from zygote to juvenile U. unicinctus using immunohistochemistry and F-actin staining for the nervous and muscular systems, respectively. Through F-actin staining, we found that muscle fibers began to form in the trochophore phase and that muscles for feeding were produced first. Subsequently, in the segmentation larval stage, the transversal muscle was formed in the shape of a ring in an anterior-to-posterior direction with segment formation, as well as a ventromedian muscle for the formation of a ventral nerve cord. After that, many muscle fibers were produced along the entire body and formed the worm-shaped larva. Finally, we investigated the spatiotemporal expression of Uun_st-mhc, Uun_troponin I, Uun_calponin, and Uun_twist genes found in U. unicinctus. During embryonic development, the striated and smooth muscle genes were co-expressed in the same region. However, the adult body wall muscles showed differential gene expression of each muscle layer. The results of this study will provide the basis for the understanding of muscle differentiation in Echiura.

Homology and evolution of the chaetae in Echiura (Annelida).

Project description:Echiura is traditionally regarded as a small phylum of unsegmented spiralian worms. Molecular analyses, however, provide unquestionable evidence that Echiura are derived annelids that lost segmentation. Like annelids, echiurans possess chaetae, a single ventral pair in all species and one or two additional caudal hemi-circles of chaetae in two subgroups, but their evolutionary origin and affiliation to annelid chaetae are unresolved. Since annelids possess segmental pairs of dorsal (notopodial) and ventral (neuropodial) chaetae that are arranged in a row, the ventral chaetae in Echiura either represent a single or a paired neuropodial group of chaetae, while the caudal circle may represent fused rows of chaetae. In annelids, chaetogenesis is generally restricted to the ventral part of the notopodial chaetal sac and to the dorsal part of the neuropodial chaetal sac. We used the exact position of the chaetal formation site in the echiuran species, Thalassema thalassemum (Pallas, 1766) and Echiurus echiurus (Pallas, 1767), to test different hypotheses of the evolution of echiurid chaetae. As in annelids, a single chaetoblast is responsible for chaetogenesis in both species. Each chaeta of the ventral pair arises from its own chaetal sac and possesses a lateral formation site, evidencing that the pair of ventral chaetae in Echiura is homologous to a pair of neuropodia that fused on the ventral side, while the notopodia were reduced. Both caudal hemi-circles of chaetae in Echiurus echiurus are composed of several individual chaetal sacs, each with its own formative site. This finding argues against a homology of these hemi-circles of chaetae and annelids' rows of chaetae and leads to the hypothesis that the caudal chaetal rings evolved once within the Echiura by multiplication of ventral chaetae.

A new species of the rare, deep-sea polychaete genus Benthoscolex from the Sea of Kumano, Japan (Annelida, Amphinomidae).

Project description:A new species of amphinomid polychaete, Benthoscolex seisuiaesp. n., is described from the Sea of Kumano, Japan, from depths of 487-596 m. The species is distinguishable from its congeners by the following features: i) palps 1.8 times as long as lateral antennae; ii) branchiae do not reach to the tip of the notochaetae. This is the first record of Benthoscolex from Japan. A partial mitochondrial cytochrome c oxidase subunit I gene sequence from the holotype of B. seisuiaesp. n. is provided for reliable species identification in the future.

New species of Trophoniella from Shimoda, Japan (Annelida, Flabelligeridae).

Project description:Trophoniella hephaistos sp. n. was collected from a tank irrigated with seawater pumped directly from Nabeta Bay, Japan. This species is discriminated from other Trophoniella by having dorsal tubercles, a tongue-shaped branchial plate, a tunic covered with large sediment grains dorsally and ventrally, having eyes, and anchylosed neurohooks starting from chaetigers 17-20. This is the first record of Trophoniella from Japanese waters. Identification keys to species of Trophoniella and four gene sequences (COI, 16S, 18S, 28S) of this species are provided. Phylogenetic analysis was conducted to clarify phylogenetic position of Trophoniella in Flabelligeridae using four genes.

Somatic Cough Syndrome (Previously Referred to as Psychogenic Cough) and Tic Cough (Previously Referred to as Habit Cough) in Adults and Children: CHEST Guideline and Expert Panel Report.

Project description:We conducted a systematic review on the management of psychogenic cough, habit cough, and tic cough to update the recommendations and suggestions of the 2006 guideline on this topic.We followed the American College of Chest Physicians (CHEST) methodologic guidelines and the Grading of Recommendations, Assessment, Development, and Evaluation framework. The Expert Cough Panel based their recommendations on data from the systematic review, patients' values and preferences, and the clinical context. Final grading was reached by consensus according to Delphi methodology.The results of the systematic review revealed only low-quality evidence to support how to define or diagnose psychogenic or habit cough with no validated diagnostic criteria. With respect to treatment, low-quality evidence allowed the committee to only suggest therapy for children believed to have psychogenic cough. Such therapy might consist of nonpharmacologic trials of hypnosis or suggestion therapy, or combinations of reassurance, counseling, and referral to a psychologist, psychotherapy, and appropriate psychotropic medications. Based on multiple resources and contemporary psychologic, psychiatric, and neurologic criteria (Diagnostic and Statistical Manual of Mental Disorders, 5th edition and tic disorder guidelines), the committee suggests that the terms psychogenic and habit cough are out of date and inaccurate.Compared with the 2006 CHEST Cough Guidelines, the major change in suggestions is that the terms psychogenic and habit cough be abandoned in favor of somatic cough syndrome and tic cough, respectively, even though the evidence to do so at this time is of low quality.

Two new species of Mediomastus (Annelida, Capitellidae) from Tokyo Bay, Japan.

Project description:Two undescribed species of polychaetes in Mediomastus (Annelida: Capitellidae) were collected from intertidal to shallow habitats in Tokyo Bay, Japan. These are M. duobalteus sp. n. and M. hanedaensis sp. n. Mediomastus duobalteus sp. n. is distinguishable from all congeners by the following characters: 1) segments 3, 4, 8-11 stainable with methyl green, 2) thoracic capillary chaetae unilimbate, 3) abdominal capillary chaetae absent, 4) paddle-like chaetae in the thorax absent, and 5) abdominal hooded hooks not flared. Mediomastus hanedaensis sp. n. is similar to M. warrenae Green, 2002, but differs from the latter in the shapes of the thoracic capillary chaetae and the abdominal hooded hooks, and the staining pattern with methyl green. In addition, a key to all Mediomastus species is provided.

Molecular identification and larval morphology of spionid polychaetes (Annelida, Spionidae) from northeastern Japan.

Project description:Planktonic larvae of spionid polychaetes are among the most common and abundant group in coastal meroplankton worldwide. The present study reports the morphology of spionid larvae collected mainly from coastal waters of northeastern Japan that were identified by the comparison of adult and larval 18S and 16S rRNA gene sequences. The molecular analysis effectively discriminated the species. Adult sequences of 48 species from 14 genera (Aonides Claparède, 1864; Boccardia Carazzi, 1893; Boccardiella Blake & Kudenov, 1978; Dipolydora Verrill, 1881; Laonice Malmgren, 1867; Malacoceros Quatrefages, 1843; Paraprionospio Caullery, 1914; Polydora Bosc, 1802; Prionospio Malmgren, 1867; Pseudopolydora Czerniavsky, 1881; Rhynchospio Hartman, 1936; Scolelepis Blainville, 1828; Spio Fabricius, 1785; Spiophanes Grube, 1860) and larval sequences of 41 species from 14 genera (Aonides; Boccardia; Boccardiella; Dipolydora; Laonice; Paraprionospio; Poecilochaetus Claparède in Ehlers, 1875; Polydora; Prionospio; Pseudopolydora; Rhynchospio; Scolelepis; Spio; Spiophanes) of spionid polychaetes were obtained; sequences of 27 of these species matched between adults and larvae. Morphology of the larvae was generally species-specific, and larvae from the same genus mostly shared morphological features, with some exceptions. Color and number of eyes, overall body shape, and type and arrangement of pigmentation are the most obvious differences between genera or species. The morphological information on spionid larvae provided in this study contributes to species or genus level larval identification of this taxon in the studied area. Identification keys to genera and species of planktonic spionid larvae in northeastern Japan are provided. The preliminary results of the molecular phylogeny of the family Spionidae using 18S and 16S rRNA gene regions are also provided.

The diagnosis of inborn errors of metabolism in previously undiagnosed adults referred for medical genetics evaluation.

Project description:Traditionally thought of as a pediatric diagnostic and therapeutic dilemma, the diagnostic rate and spectrum of inborn errors of metabolism (IEM) in the adult population is largely unknown. A retrospective chart review of patients seen by the Michigan Medicine Adult Medical Genetics Clinic for clinical evaluation from 2014 to 2018 was conducted. Patients referred for a primary indication possibly consistent with an IEM were considered. Variables included age at genetic evaluation, symptom onset age, sex, clinical course, organ systems involved, developmental history, family history and prior genetic testing. Of patients evaluated during the study period, 112 were referred for an indication possibly consistent with an IEM and underwent a complete biochemical workup with an IEM diagnostic rate of 9.8% achieved. An additional 9.8% were diagnosed with a non-IEM genetic diagnosis. Management changes were implemented in all IEM diagnoses. Metabolic disorders in the adult population are under-recognized and under-diagnosed. This report demonstrates the need for clinicians to consider these diagnoses in adults and either refer to a genetics clinic or initiate a biochemical workup. As advances in diagnosis, treatment, and life expectancy of patients with IEMs increases, recognizing and diagnosing these conditions can significantly impact care.

Thauera butanivorans sp. nov., a C2-C9 alkane-oxidizing bacterium previously referred to as 'Pseudomonas butanovora'.

Project description:The placement of 'Pseudomonas butanovora' in the genus Thauera was proposed previously, based on 16S rRNA gene sequence analysis, upon further studies of taxonomical characteristics. In this study, physiological characteristics and DNA-DNA reassociation data are presented and the transfer of 'P. butanovora' to the genus Thauera is proposed. The original description of the strain (strain Bu-B1211) indicated that it was capable of denitrification but not anaerobic growth. 'P. butanovora' is capable of anaerobic respiration and growth, utilizing nitrate as a terminal electron acceptor during the oxidation of organic acids and alcohols, but not aromatic hydrocarbons or open-chain terpenoids. The total fatty acid composition supported the assignment of strain Bu-B1211 to the Betaproteobacteria and resembled that of members of the genus Thauera. The combination of 16S rRNA gene phylogenetic evidence, physiological and taxonomical characteristics and DNA-DNA reassociation data supported the placement of 'Pseudomonas butanovora' Bu-B1211 in the genus Thauera as representing a novel species, for which the name Thauera butanivorans sp. nov. is proposed. The type strain is Bu-B1211(T) (=IAM 12574(T)=ATCC 43655(T)=DSM 2080(T)).

A new genus and species of Serpulidae (Annelida, Polychaeta, Sabellida) from the Caribbean Sea.

Project description:A new genus and species of Serpulidae (Annelida, Polychaeta) from the Caribbean Sea. Turbocavus secretus (gen. nov. and sp. nov.) is described from shallow hard substrates (0.5-3 m) in wind-sheltered bays of St. John, U.S. Virgin Islands and Curaçao, as well as from diving depths (46-49 m) around Bonaire (Leeward Antilles), Caribbean Sea. The new taxon, which has from 7 to 19 thoracic chaetigers and up to 335 abdominal chaetigers, bears a unique type of thoracic chaeta which is multifolded at the base and continues with a groove tapering to the capillary tip. The new serpulid has unique 18S rRNA sequences and genetic analysis of the 18S rRNA gene situates the new genus at the basis of the serpulid cladogram, well separated from other genera, and close to Filograna/Salmacina and Protula.
