Preliminary molecular identification of a predatory bug, Orius albidipennis, collected from ornamental plants.

ABSTRACT: Orius albidipennis (Reuter) (Hemiptera: Anthocoridae) is a generalist predator used for biological control of insects attacking ornamental plants. Molecular identification of this species using internal transcribed spacer 1 (ITS1) of ribosomal DNA was conducted for the first time. The complete sequence of ITS1 and fragments of its flanking 18S and 5.8S rDNA genes are reported herein. The estimated length of ITS1 of O. albidipennis was 305 bp. This spacer was nearly identical to its counterpart of Orius sp-Taif strain in spite of the difference in their length. The phylogentic relationships were determined using the maximum-likelihood method supported with strong bootstrap probabilities clustering of both taxa together. Further molecular markers could be useful to identify the Taif strain and support its sister relationship to the Egyptian O. albidipennis.

Project description:BackgroundOviposition site selection is an important factor in determining the success of insect populations. Orius spp. are widely used in the biological control of a wide range of soft-bodied insect pests such as thrips, aphids, and mites. Orius strigicollis (Heteroptera: Anthocoridae) is the dominant Orius species in southern China; however, what factor drives its selection of an oviposition site after mating currently remains unknown.MethodsHere, kidney bean pods (KBPs) were chosen as the oviposition substrate, and choice and nonchoice experiments were conducted to determine the preferences concerning oviposition sites on the KBPs of O. strigicollis. The mechanism of oviposition behavior was revealed through observation and measurement of oviposition action, the egg hatching rate, and the oviposition time.ResultsWe found that O. strigicollis preferred the seams of the pods for oviposition, especially the seams at the tips of the KBPs. Choice and nonchoice experiments showed that females did not lay eggs when the KBP tail parts were unavailable. The rates of egg hatching on different KBP parts were not significantly different, but the time required for females to lay eggs on the tip seam was significantly lower. Decreased oviposition time is achieved on the tip seam because the insect can exploit support points found there and gain leverage for insertion of the ovipositor.DiscussionThe preferences for oviposition sites of O. strigicollis are significantly influenced by the topography of the KBP surface. Revealing such behavior and mechanisms will provide an important scientific basis for the mass rearing of predatory bugs.

Stable Isotope Enrichment (?15N) in the Predatory Flower Bug (Orius majusculus) Predicts Fitness-Related Differences between Diets.

Project description:Mass rearing of insects, used both as biological control agents and for food and feed, is receiving increasing attention. Efforts are being made to improve diets that are currently in use, and to identify alternative diets, as is the case with the predatory flower bug (Orius majusculus) and other heteropteran predators, due to the high costs of their current diet, the eggs of the Mediterranean flour moth (E. kuehniella). The assessment of alternative diets may include measurements of the predator's fitness-related traits (development time, weight, etc.), and biochemical analyses such as lipid and protein content in the diet and the insects. However, assessing diet quality via the predator's fitness-related traits is laborious, and biochemical composition is often difficult to relate to the measured traits. Isotope analysis, previously used for diet reconstruction studies, can also serve as a tool for the assessment of diet quality. Here, the variation in discrimination factors or isotope enrichment (?15N and ?13C) indicates the difference in isotopic ratio between the insect and its diet. In this study, we investigated the link between ?15N and diet quality in the predatory bug Orius majusculus. Three groups of bugs were fed different diets: Ephestia kuehniella eggs, protein-rich Drosophila melanogaster and lipid-rich D. melanogaster. The isotopic enrichment and fitness-related measurements were assessed for each group. Results show a relation between ?15N and fitness-related measurements, which conform to the idea that lower ?15N indicates a higher diet quality.

Mechanisms of Salt Tolerance and Molecular Breeding of Salt-Tolerant Ornamental Plants.

Project description:As the area of salinized soils increases, and freshwater becomes more scarcer worldwide, an urgent measure for agricultural production is to use salinized land and conserve freshwater resources. Ornamental flowering plants, such as carnations, roses, chrysanthemums, and gerberas, are found around the world and have high economic, ornamental, ecological, and edible value. It is therefore prudent to improve the salt tolerance of these important horticultural crops. Here, we summarize the salt-adaptive mechanisms, genes, and molecular breeding of ornamental flowering crops. We also review the genome editing technologies that provide us with the means to obtain novel varieties with high salinity tolerance and improved utility value, and discuss future directions of research into ornamental plants like salt exclusion mechanism. We considered that the salt exclusion mechanism in ornamental flowering plants, the acquisition of flowers with high quality and novel color under salinity condition through gene editing techniques should be focused on for the future research.

Ethylene resistance in flowering ornamental plants - improvements and future perspectives.

Project description:Various strategies of plant breeding have been attempted in order to improve the ethylene resistance of flowering ornamental plants. These approaches span from conventional techniques such as simple cross-pollination to new breeding techniques which modify the plants genetically such as precise genome-editing. The main strategies target the ethylene pathway directly; others focus on changing the ethylene pathway indirectly via pathways that are known to be antagonistic to the ethylene pathway, e.g. increasing cytokinin levels. Many of the known elements of the ethylene pathway have been addressed experimentally with the aim of modulating the overall response of the plant to ethylene. Elements of the ethylene pathway that appear particularly promising in this respect include ethylene receptors as ETR1, and transcription factors such as EIN3. Both direct and indirect approaches seem to be successful, nevertheless, although genetic transformation using recombinant DNA has the ability to save much time in the breeding process, they are not readily used by breeders yet. This is primarily due to legislative issues, economic issues, difficulties of implementing this technology in some ornamental plants, as well as how these techniques are publically perceived, particularly in Europe. Recently, newer and more precise genome-editing techniques have become available and they are already being implemented in some crops. New breeding techniques may help change the current situation and pave the way toward a legal and public acceptance if products of these technologies are indistinguishable from plants obtained by conventional techniques.

Molecular and Morphological Assessment of Septoria Species Associated with Ornamental Plants in Yunnan Province, China.

Project description:The Karst landform is the main geographic characteristic in South China. Such areas are rich in vegetation and especially suitable for growth of shrubs and herbaceous plants. In this study, 11 Septoria strains were obtained from different plants' leaves collected in the Kunming Botanical Garden, Yunnan Province, China. Based on single-gene and multi-gene analyses of five gene loci (tef1, rpb2, tub2, ITS, and LSU) and four gene regions (without LSU), these strains were found to belong to three independent phylogenetic lineages representing five species, including four novel taxa, and one new record for China. Five single gene trees were also provided to evaluate the effectiveness of each gene for discriminating the species, as a result of which tub2 was found to have the most suitable DNA barcode for rapid identification. Morphological descriptions, illustrations, and comparisons are provided for a more comprehensive assessment. Genealogical Concordance Phylogenetic Species Recognition (GCPSR) with a pairwise homoplasy index (PHI) test was used to evaluate the conclusions of the phylogenetic analyses.

Herbaceous perennial ornamental plants can support complex pollinator communities.

Project description:Human-designed landscapes can host diverse pollinator communities, and the availability of floral resources is central to supporting insect biodiversity in highly modified environments. However, some urban landscapes have relatively few pollinator-attractive plant species and management in urban environments rarely considers the function of these plants in generating and supporting a stable ecological community. Evaluations of 25 cultivars within five commercially popular herbaceous perennial ornamental plant genera (Agastache, Echinacea, Nepeta, Rudbeckia, and Salvia) revealed variation in the total and proportional abundance of visitors attracted. These varieties supported multiple pollinator functional groups, however bees were the primary visitors to in this system. Cultivars were assessed according to their function within a plant-pollinator network. Comparisons of artificial networks created with the six most attractive and six least attractive cultivars demonstrated that a planting scheme using the most attractive cultivars would attract nearly four times as many bee species, including several specialists and rare species. Plant diversity in the landscape was correlated with abundance and diversity of pollinator visitors, demonstrating that community context shapes a plant's relative attractiveness to pollinators. We conclude that herbaceous perennial cultivars can support an abundance and diversity of pollinator visitors, however, planting schemes should take into consideration the effects of cultivar, landscape plant diversity, floral phenology, floral area, and contribution to a stable ecological community.

Complex floral traits shape pollinator attraction to ornamental plants.

Project description:Background and aimsOrnamental flowering plant species are often used in managed greenspaces to attract and support pollinator populations. In natural systems, selection by pollinators is hypothesized to result in convergent multimodal floral phenotypes that are more attractive to specific pollinator taxa. In contrast, ornamental cultivars are bred via artificial selection by humans, and exhibit diverse and distinct phenotypes. Despite their prevalence in managed habitats, the influence of cultivar phenotypic variation on plant attractiveness to pollinator taxa is not well resolved.MethodsWe used a combination of field and behavioural assays to evaluate how variation in floral visual, chemical and nutritional traits impacted overall attractiveness and visitation by pollinator taxonomic groups and bee species to 25 cultivars of five herbaceous perennial ornamental plant genera.Key resultsDespite significant phenotypic variation, cultivars tended to attract a broad range of pollinator species. Nonetheless, at the level of insect order (bee, fly, butterfly, beetle), attraction was generally modulated by traits consistent with the pollination syndrome hypothesis. At the level of bee species, the relative influence of traits on visitation varied across plant genera, with some floral phenotypes leading to a broadening of the visitor community, and others leading to exclusion of visitation by certain bee species.ConclusionsOur results demonstrate how pollinator choice is mediated by complex multimodal floral signals. Importantly, the traits that had the greatest and most consistent effect on regulating pollinator attraction were those that are commonly selected for in cultivar development. Though variation among cultivars in floral traits may limit the pollinator community by excluding certain species, it may also encourage interactions with generalist taxa to support pollinator diversity in managed landscapes.

Identification of metabolizing enzyme genes associated with xenobiotics and odorants in the predatory stink bug Arma custos based on transcriptome analysis.

Project description:The predatory stink bug, Arma custos, is a highly effective beneficial predator of crop pests. The lack of gene information related to xenobiotic detoxification and odorant degrading enzymes in the predator stink bugs to date has limited our ability for more in-depth studies of biological control. Hence, we conducted de novo assembly of the A. custos transcriptome from guts, antennae, and other tiussue samples of 5th instar larvae using Illumina sequencing technology. A total of 91, 50 and 23 genes of cytochrome P450 monooxygenases (CYPs), carboxyl/choline esterases (CCEs) and glutathione S-transferases (GSTs) genes were identified, respectively. Gene expansions of CYP3 and CYP4 clans and the hormone and pheromone processing CCE class were found in A. custos. Analysis of tissue-specific expression patterns showed that 37 CYPs, 14 CCEs and 8 GSTs were enriched in guts, and 6 CYPs, 5 CCEs and 2 GSTs were up-regulated in antennae, suggesting their potential roles on xenobiotics detoxification and ordorant degradation. Gene information data presented here could be useful for a deeper understanding of the ecology, physiology and behavior of this beneficial species and could be helpful to improve their bio-control efficiency.
