Device-tissue interactions: a collaborative communications system.

ABSTRACT: Medical devices, including surgical staplers, energy-based devices, and access enabling devices, are used routinely today in the majority of surgical procedures. Although these technically advanced devices have proved to be of immense benefit to both surgeons and patients, their rapid development and continuous improvement have had the unintended consequence of creating a knowledge gap for surgeons due to a lack of adequate training and educational programs. Thus, there is an unmet need in the surgical community to collect existing data on device-tissue interactions and subsequently develop research and educational programs to fill this gap in surgical training. Gathering data and developing these new programs will require collaboration between doctors, engineers, and scientists, from both clinical practice and industry. This paper presents a communications system to enable this unique collaboration that can potentially result in significantly improved patient care.

Delay minimization based uplink resource allocation for device-to-device communications considering mmWave propagation.

Project description:This paper addresses the resource allocation problem in multi-sharing uplink for device-to-device (D2D) communication, one aspect of 5G communication networks. The main advantage and motivation in relation to the use of D2D communication is the significant improvement in the spectral efficiency of the system when exploiting the proximity of communication pairs and reusing idle resources of the network, mainly in the uplink mode, where there are more idle available resources. An approach is proposed for allocating resources to D2D and cellular user equipments (CUE) users in the uplink of a 5G based network which considers the estimation of delay bound value. The proposed algorithm considers minimization of total delay for users in the uplink and solves the problem by forming conflict graph and by finding the maximal weight independent set. For the user delay estimation, an approach is proposed that considers the multifractal traffic envelope process and service curve for the uplink. The performance of the algorithm is evaluated through computer simulations in comparison with those of other algorithms in the literature in terms of throughput, delay, fairness and computational complexity in a scenario with channel modeling that describes the propagation of millimeter waves at frequencies above 6 GHz. Simulation results show that the proposed allocation algorithm outperforms other algorithms in the literature, being highly efficient to 5G systems.

Digital micromirror device projection printing system for meniscus tissue engineering.

Project description:Meniscus degeneration due to age or injury can lead to osteoarthritis. Although promising, current cell-based approaches show limited success. Here we present three-dimensional methacrylated gelatin (GelMA) scaffolds patterned via projection stereolithography to emulate the circumferential alignment of cells in native meniscus tissue. Cultured human avascular zone meniscus cells from normal meniscus were seeded on the scaffolds. Cell viability was monitored, and new tissue formation was assessed by gene expression analysis and histology after 2weeks in serum-free culture with transforming growth factor β1 (10ngml(-1)). Light, confocal and scanning electron microscopy were used to observe cell-GelMA interactions. Tensile mechanical testing was performed on unseeded, fresh scaffolds and 2-week-old cell-seeded and unseeded scaffolds. 2-week-old cell-GelMA constructs were implanted into surgically created meniscus defects in an explant organ culture model. No cytotoxic effects were observed 3weeks after implantation, and cells grew and aligned to the patterned GelMA strands. Gene expression profiles and histology indicated promotion of a fibrocartilage-like meniscus phenotype, and scaffold integration with repair tissue was observed in the explant model. We show that micropatterned GelMA scaffolds are non-toxic, produce organized cellular alignment, and promote meniscus-like tissue formation. Prefabrication of GelMA scaffolds with architectures mimicking the meniscus collagen bundle organization shows promise for meniscal repair. Furthermore, the technique presented may be scaled up to repair larger defects.

DAS writeback: a collaborative annotation system.

Project description:BACKGROUND: Centralised resources such as GenBank and UniProt are perfect examples of the major international efforts that have been made to integrate and share biological information. However, additional data that adds value to these resources needs a simple and rapid route to public access. The Distributed Annotation System (DAS) provides an adequate environment to integrate genomic and proteomic information from multiple sources, making this information accessible to the community. DAS offers a way to distribute and access information but it does not provide domain experts with the mechanisms to participate in the curation process of the available biological entities and their annotations. RESULTS: We designed and developed a Collaborative Annotation System for proteins called DAS Writeback. DAS writeback is a protocol extension of DAS to provide the functionalities of adding, editing and deleting annotations. We implemented this new specification as extensions of both a DAS server and a DAS client. The architecture was designed with the involvement of the DAS community and it was improved after performing usability experiments emulating a real annotation task. CONCLUSIONS: We demonstrate that DAS Writeback is effective, usable and will provide the appropriate environment for the creation and evolution of community protein annotation.

Laser communications system with drones as relay medium for healthcare applications.

Project description:This article introduces a prototype laser communication system integrated with uncrewed aerial vehicles (UAVs), aimed at enhancing data connectivity in remote healthcare applications. Traditional radio frequency systems are limited by their range and reliability, particularly in challenging environments. By leveraging UAVs as relay points, the proposed system seeks to address these limitations, offering a novel solution for real-time, high-speed data transmission. The system has been empirically tested, showcasing its ability to maintain data transmission integrity under various conditions. Results indicate a substantial improvement in connectivity, with high data transmission success rate (DTSR) scores, even amidst environmental disturbances. This study underscores the system's potential for critical applications such as emergency response, public health monitoring, and extending services to remote or underserved areas.

Creating Interactions between Tissue-Engineered Skeletal Muscle and the Peripheral Nervous System.

Project description:Effective models of mammalian tissues must allow and encourage physiologically (mimetic) correct interactions between co-cultured cell types in order to produce culture microenvironments as similar as possible to those that would normally occur in vivo. In the case of skeletal muscle, the development of such a culture model, integrating multiple relevant cell types within a biomimetic scaffold, would be of significant benefit for investigations into the development, functional performance, and pathophysiology of skeletal muscle tissue. Although some work has been published regarding the behaviour of in vitro muscle models co-cultured with organotypic slices of CNS tissue or with stem cell-derived neurospheres, little investigation has so far been made regarding the potential to maintain isolated motor neurons within a 3D biomimetic skeletal muscle culture platform. Here, we review the current state of the art for engineering neuromuscular contacts in vitro and provide original data detailing the development of a 3D collagen-based model for the co-culture of primary muscle cells and motor neurons. The devised culture system promotes increased myoblast differentiation, forming arrays of parallel, aligned myotubes on which areas of nerve-muscle contact can be detected by immunostaining for pre- and post-synaptic proteins. Quantitative RT-PCR results indicate that motor neuron presence has a positive effect on myotube maturation, suggesting neural incorporation influences muscle development and maturation in vitro. The importance of this work is discussed in relation to other published neuromuscular co-culture platforms along with possible future directions for the field.

Redressing the interactions between stem cells and immune system in tissue regeneration.

Project description:Skeletal muscle has an extraordinary regenerative capacity reflecting the rapid activation and effective differentiation of muscle stem cells (MuSCs). In the course of muscle regeneration, MuSCs are reprogrammed by immune cells. In turn, MuSCs confer immune cells anti-inflammatory properties to resolve inflammation and facilitate tissue repair. Indeed, MuSCs can exert therapeutic effects on various degenerative and inflammatory disorders based on their immunoregulatory ability, including effects primed by interferon-γ (IFN-γ) and tumor necrosis factor-α (TNF-α). At the molecular level, the tryptophan metabolites, kynurenine or kynurenic acid, produced by indoleamine 2,3-dioxygenase (IDO), augment the expression of TNF-stimulated gene 6 (TSG6) through the activation of the aryl hydrocarbon receptor (AHR). In addition, insulin growth factor 2 (IGF2) produced by MuSCs can endow maturing macrophages oxidative phosphorylation (OXPHOS)-dependent anti-inflammatory functions. Herein, we summarize the current understanding of the immunomodulatory characteristics of MuSCs and the issues related to their potential applications in pathological conditions, including COVID-19.

Dynamic Appliances Scheduling in Collaborative MicroGrids System.

Project description:In this paper a new approach which is based on a collaborative system of MicroGrids (MG's), is proposed to enable household appliance scheduling. To achieve this, appliances are categorized into flexible and non-flexible Deferrable Loads (DL's), according to their electrical components. We propose a dynamic scheduling algorithm where users can systematically manage the operation of their electric appliances. The main challenge is to develop a flattening function calculus (reshaping) for both flexible and non-flexible DL's. In addition, implementation of the proposed algorithm would require dynamically analyzing two successive multi-objective optimization (MOO) problems. The first targets the activation schedule of non-flexible DL's and the second deals with the power profiles of flexible DL's. The MOO problems are resolved by using a fast and elitist multi-objective genetic algorithm (NSGA-II). Finally, in order to show the efficiency of the proposed approach, a case study of a collaborative system that consists of 40 MG's registered in the load curve for the flattening program has been developed. The results verify that the load curve can indeed become very flat by applying the proposed scheduling approach.

Project description:Male germ cell development depends on multiple biological events that combine epigenetic reprogramming, cell cycle regulation, and cell migration in a spatio-temporal manner. Sertoli cells are a crucial component of the spermatogonial stem cell niche and provide essential growth factors and chemokines to developing germ cells. This review focuses mainly on the activation of master regulators of the niche in Sertoli cells and their targets, as well as on novel molecular mechanisms underlying the regulation of growth and differentiation factors such as GDNF and retinoic acid by NOTCH signaling and other pathways.

A semi-centralized blockchain system with multi-chain for auditing communications of Wide Area Protection System.

Project description:Wide Area Protection System (WAPS) undertakes the important task of maintaining system reliability and stability when the power system is subject to abnormal or predetermined unstable conditions. The existing WAPS adopts a centralized mechanism to record and audit communication messages, which faces the risk of excessive authority and tampering with communication records and audit logs, thus making it impossible to achieve true transparency and fairness. Due to the involvement of multiple parties and equipment maintained by different manufacturers in the communication of WAPS, there are difficulties in tracing the cause of the accident and determining the at-fault party following misoperations and miss trips. To address this issue, we propose a semi-centralized blockchain system with multi-chain for auditing communications of WAPS. We first propose a semi-centralized system architecture according to the system architecture and management requirements of WAPS. Then, we utilize the blockchain network as a self-recording channel to achieve tamper-proof and non-repudiation verification interaction. We also design a multi-chain structure and classification node mechanism to meet the communication auditing requirements of multiple WAPS. We have designed a new block structure that conforms to the communication protocol of WAPS. To reduce the storage burden caused by the ever-expanding blockchain ledger, we propose a deletable blockchain scheme while maintaining the integrity and security of blockchain. Analysis and experiments show that the proposed blockchain system can support the secure, transparent, tamper-proof and traceable communication recording and auditing of WAPS along with high performance.
