IL-17 in the rheumatologist's line of sight.

ABSTRACT: Over the past decades, the identification of several new cytokines, including interleukin (IL)-17 and IL-23, and of new T helper cell subsets, including Th17 cells, has changed the vision of immunological processes. The IL-17/Th17 pathway plays a critical role during the development of inflammation and autoimmunity, and targeting this pathway has become an attractive strategy for a number of diseases. This review aims to describe the effects of IL-17 in the joint and its roles in the development of autoimmune and inflammatory arthritis. Furthermore, biotherapies targeting directly or indirectly IL-17 in inflammatory rheumatisms will be developed.

A dataset for Wi-Fi-based human activity recognition in line-of-sight and non-line-of-sight indoor environments.

Project description:The aim of this paper is to present a dataset for Wi-Fi-based human activity recognition. The dataset is comprised of five experiments performed by 30 different subjects in three different indoor environments. The experiments performed in the first two environments are of a line-of-sight (LOS) nature, while the experiments performed in the third environment are of a non-line-of-sight (NLOS) nature. Each subject performed 20 trials for each of the experiments which makes the overall number of recorded trials in the dataset equals to 3000 trials (30 subjects × 5 experiments × 20 trials). To record the data, we used the channel state information (CSI) tool [1] to capture the exchanged Wi-Fi packets between a Wi-Fi transmitter and receiver. The utilized transmitter and receiver are retrofitted with the Intel 5300 network interface card which enabled us to capture the CSI values that are contained in the recorded transmissions. Unlike other publicly available human activity datasets, this dataset provides researchers with the ability to test their developed methodologies on both LOS and NLOS environments, in addition to many different variations of human movements, such as walking, falling, turning, and pen pick up from the ground.

IL-17 and IL-17-producing cells in protection versus pathology.

Project description:IL-17 cytokine family members have diverse biological functions, promoting protective immunity against many pathogens but also driving inflammatory pathology during infection and autoimmunity. IL-17A and IL-17F are produced by CD4+ and CD8+ T cells, γδ T cells, and various innate immune cell populations in response to IL-1β and IL-23, and they mediate protective immunity against fungi and bacteria by promoting neutrophil recruitment, antimicrobial peptide production and enhanced barrier function. IL-17-driven inflammation is normally controlled by regulatory T cells and the anti-inflammatory cytokines IL-10, TGFβ and IL-35. However, if dysregulated, IL-17 responses can promote immunopathology in the context of infection or autoimmunity. Moreover, IL-17 has been implicated in the pathogenesis of many other disorders with an inflammatory basis, including cardiovascular and neurological diseases. Consequently, the IL-17 pathway is now a key drug target in many autoimmune and chronic inflammatory disorders; therapeutic monoclonal antibodies targeting IL-17A, both IL-17A and IL-17F, the IL-17 receptor, or IL-23 are highly effective in some of these diseases. However, new approaches are needed to specifically regulate IL-17-mediated immunopathology in chronic inflammation and autoimmunity without compromising protective immunity to infection.

Non-line-of-sight reconstruction with signal-object collaborative regularization.

Project description:Non-line-of-sight imaging aims at recovering obscured objects from multiple scattered lights. It has recently received widespread attention due to its potential applications, such as autonomous driving, rescue operations, and remote sensing. However, in cases with high measurement noise, obtaining high-quality reconstructions remains a challenging task. In this work, we establish a unified regularization framework, which can be tailored for different scenarios, including indoor and outdoor scenes with substantial background noise under both confocal and non-confocal settings. The proposed regularization framework incorporates sparseness and non-local self-similarity of the hidden objects as well as the smoothness of the signals. We show that the estimated signals, albedo, and surface normal of the hidden objects can be reconstructed robustly even with high measurement noise under the proposed framework. Reconstruction results on synthetic and experimental data show that our approach recovers the hidden objects faithfully and outperforms state-of-the-art reconstruction algorithms in terms of both quantitative criteria and visual quality.

Passive optical time-of-flight for non line-of-sight localization.

Project description:Optical imaging through diffusive, visually-opaque barriers and around corners is an important challenge in many fields, ranging from defense to medical applications. Recently, novel techniques that combine time-of-flight (TOF) measurements with computational reconstruction have allowed breakthrough imaging and tracking of objects hidden from view. These light detection and ranging (LiDAR)-based approaches require active short-pulsed illumination and ultrafast time-resolved detection. Here, bringing notions from passive radio detection and ranging (RADAR) and passive geophysical mapping approaches, we present an optical TOF technique that allows passive localization of light sources and reflective objects through diffusive barriers and around corners. Our approach retrieves TOF information from temporal cross-correlations of scattered light, via interferometry, providing temporal resolution that surpasses state-of-the-art ultrafast detectors by three orders of magnitude. While our passive approach is limited by signal-to-noise to relatively sparse scenes, we demonstrate passive localization of multiple white-light sources and reflective objects hidden from view using a simple setup.

Non-line-of-sight imaging with arbitrary illumination and detection pattern.

Project description:Non-line-of-sight (NLOS) imaging aims at reconstructing targets obscured from the direct line of sight. Existing NLOS imaging algorithms require dense measurements at regular grid points in a large area of the relay surface, which severely hinders their availability to variable relay scenarios in practical applications such as robotic vision, autonomous driving, rescue operations and remote sensing. In this work, we propose a Bayesian framework for NLOS imaging without specific requirements on the spatial pattern of illumination and detection points. By introducing virtual confocal signals, we design a confocal complemented signal-object collaborative regularization (CC-SOCR) algorithm for high-quality reconstructions. Our approach is capable of reconstructing both the albedo and surface normal of the hidden objects with fine details under general relay settings. Moreover, with a regular relay surface, coarse rather than dense measurements are enough for our approach such that the acquisition time can be reduced significantly. As demonstrated in multiple experiments, the proposed framework substantially extends the application range of NLOS imaging.

Tracking objects outside the line of sight using 2D intensity images.

Project description:The observation of objects located in inaccessible regions is a recurring challenge in a wide variety of important applications. Recent work has shown that using rare and expensive optical setups, indirect diffuse light reflections can be used to reconstruct objects and two-dimensional (2D) patterns around a corner. Here we show that occluded objects can be tracked in real time using much simpler means, namely a standard 2D camera and a laser pointer. Our method fundamentally differs from previous solutions by approaching the problem in an analysis-by-synthesis sense. By repeatedly simulating light transport through the scene, we determine the set of object parameters that most closely fits the measured intensity distribution. We experimentally demonstrate that this approach is capable of following the translation of unknown objects, and translation and orientation of a known object, in real time.

IL-17-receptor-associated adaptor Act1 directly stabilizes mRNAs to mediate IL-17 inflammatory signaling.

Project description:Mechanisms that degrade inflammatory mRNAs are well known; however, stabilizing mechanisms are poorly understood. Here, we show that Act1, an interleukin-17 (IL-17)-receptor-complex adaptor, binds and stabilizes mRNAs encoding key inflammatory proteins. The Act1 SEFIR domain binds a stem-loop structure, the SEFIR-binding element (SBE), in the 3' untranslated region (UTR) of Cxcl1 mRNA, encoding an inflammatory chemokine. mRNA-bound Act1 directs formation of three compartmentally distinct RNA-protein complexes (RNPs) that regulate three disparate events in inflammatory-mRNA metabolism: preventing mRNA decay in the nucleus, inhibiting mRNA decapping in P bodies and promoting translation. SBE RNA aptamers decreased IL-17-mediated mRNA stabilization in vitro, IL-17-induced skin inflammation and airway inflammation in a mouse asthma model, thus providing a therapeutic strategy for autoimmune diseases. These results reveal a network in which Act1 assembles RNPs on the 3' UTRs of select mRNAs and consequently controls receptor-mediated mRNA stabilization and translation during inflammation.

Reconfigurable beam system for non-line-of-sight free-space optical communication.

Project description:In this paper, we propose a reconfigurable beam-shaping system to permit energy-efficient non-line-of-sight (NLOS) free-space optical communication. Light is steered around obstacles blocking the direct communication pathway and reaches a receiver after reflecting off of a diffuse surface. A coherent array optical transmitter (CAO-Tx) is used to spatially shape the wavefront of the light incident on a diffuse surface. Wavefront shaping is used to enhance the amount of diffusely reflected light reaching the optical receiver. Synthetic NLOS experiments for a signal reflected over an angular range of 20° are presented. A record-breaking 30-Gbit/s orthogonal frequency-division multiplexing signal is transmitted over a diffused optical wireless link with a >17-dB gain.

IL-23/IL-17 Axis in Inflammatory Rheumatic Diseases.

Project description:In inflammatory rheumatic disorders, the immune system attacks and damages the connective tissues and invariably internal organs. During the past decade, remarkable advances having been made towards our understanding on the cellular and molecular mechanisms involved in rheumatic diseases. The discovery of IL-23/IL-17 axis and the delineation of its important role in the inflammation led to the introduction of many needed new therapeutic tools. We will present an overview of the rationale for targeting therapeutically the IL-23/IL-17 axis in rheumatic diseases and the clinical benefit which has been realized so far. Finally, we will discuss the complex interrelationship between IL-23 and IL-17 and the possible uncoupling in certain disease settings.

Differential requirement for P2X7R function in IL-17 dependent vs. IL-17 independent cellular immune responses.

Project description:IL17-dependent autoimmunity to collagen type V (Col V) has been associated with lung transplant obliterative bronchiolitis. Unlike the T helper 1 (Th1)-dependent immune responses to Tetanus Toxoid (TT), the Th17 response to Col V in lung transplant patients and its Th1/17 variant observed in coronary artery disease patients requires IL-1?, tumor necrosis factor ? and CD14(+) cells. Given the involvement of the P2X7 receptor (P2X7R) in monocyte IL-1? responses, we investigated its role in Th17-, Th1/17- and Th1-mediated proinflammatory responses. Transfer of antigen-pulsed peripheral blood mononucleated cells (PBMCs) from Col V-reactive patients into SCID mouse footpads along with P2X7R antagonists revealed a selective inhibition of Col V-, but not TT-specific swelling responses. P2X7R inhibitors blocked IL-1? induction from monocytes, including both Col V-?1 peptide-induced (T-dependent), as well as native Col V-induced (T-independent) responses. Significantly higher P2X7R expression was found on CXCR3(neg) CCR4(+)/6(+) CD4(+) [Th17] versus CXCR3(+)CCR4/6(neg) CD4(+) [Th1] subsets in PBMCs, suggesting that the paradigm of selective dependence on P2X7R might extend beyond Col V autoimmunity. Indeed, P2X7R inhibitors suppressed not only anti-Col V, but also Th1/17-mediated alloimmunity, in a heart transplant patient without affecting anti-viral Epstein-Barr virus responses. These results suggest that agents targeting the P2X7R might effectively treat Th17-related transplant pathologies, while maintaining Th1-immunity to infection.
