Fabrication of core-shell nanoparticles via controlled aggregation of semi-flexible conjugated polymer and hyaluronic acid.

ABSTRACT: Core-shell conjugated polymer nanoparticles (CPNs) were fabricated by complexing a semi-flexible, primary amine-containing conjugated polymer (CP) with hyaluronic acid (HA). Flexibility introduced in the rigid rod conjugated backbone allows backbone reorganization to increase ?-? interaction under ionic complexation, resulting in core-shell nanoparticles with a hydrophobic CP core wrapped with a HA shell. The core-shell nanoparticles exhibited no cellular toxicity and high cancer cell specificity with minimal binding to normal cells.

Fabrication of Anisotropic Cu Ferrite-Polymer Core-Shell Nanoparticles for Photodynamic Ablation of Cervical Cancer Cells.

Project description:In this work we developed methylene blue-immobilized copper-iron nanoparticles (MB-CuFe NPs) through a facile one-step hydrothermal reaction to achieve a better phototherapeutic effect. The Fe/Cu ratio of the CuFe NPs was controllable by merely changing the loading amount of iron precursor concentration. The CuFe NPs could serve as a Fenton catalyst to convert hydrogen peroxide (H2O2) into reactive oxygen species (ROS), while the superparamagnetic properties also suggest magnetic resonance imaging (MRI) potential. Furthermore, the Food and Drug Administration (FDA)-approved MB photosensitizer could strongly adsorb onto the surface of CuFe NPs to facilitate the drug delivery into cells and improve the photodynamic therapy at 660 nm via significant generation of singlet oxygen species, leading to enhanced cancer cell-damaging efficacy. An MTT (thiazolyl blue tetrazolium bromide) assay proved the low cytotoxicity of the CuFe NPs to cervical cancer cells (HeLa cells), namely above 80% at 25 ppm of the sample dose. A slight dissolution of Cu and Fe ions from the CuFe NPs in an acidic environment was obtained, providing direct evidence for CuFe NPs being degradable without the risk of long-term retention in the body. Moreover, the tremendous photo-to-thermal conversion of CuFe NPs was examined, which might be combined with photodynamic therapy (PDT) for promising development in the depletion of cancer cells after a single pulse of deep-red light irradiation at high laser power.

Controlled aggregation in conjugated polymer nanoparticles via organic acid treatments.

Project description:Understanding and controlling aggregation structures of conjugated polymers (CPs) in aqueous solutions is critical to improving the physical and photophysical properties of CPs for biological applications. Here, we present spectroscopic evidence, including nuclear magnetic resonance (NMR) spectroscopic results, that different organic acid treatment induces different aggregation structures and photophysical properties of CPs in water. Conjugated polymer nanoparticles (CPNs) were fabricated by treating a non-aqueous soluble, primary amine-containing poly(phenylene ethynylene) (PPE-NH(2)) with organic acids followed by dialysis. CPNs formed by acetic acid (AA) treatment (CPN-AAs) exhibit characteristics of loose aggregation with minimal π-π stacking, while CPNs formed by tartaric acid (TA) treatment (CPN-TAs) exhibit a high degree of π-π stacking among PPE-NH(2) chains. The controlled aggregation for a specific application was demonstrated by comparing the fluorescence quenching abilities of the CPN-AAs and the CPN-TAs. A doubled Stern-Volmer constant was obtained from the densely packed CPN-TAs compared to that of the loosely aggregated CPN-AAs.

Inhibition of protein aggregation by zwitterionic polymer-based core-shell nanogels.

Project description:Protein aggregation is a process by which misfolded proteins polymerizes into aggregates and forms fibrous structures with a ?-sheet conformation, known as amyloids. It is an undesired outcome, as it not only causes numerous neurodegenerative diseases, but is also a major deterrent in the development of protein biopharmaceuticals. Here, we report a rational design for the synthesis of novel zwitterionic polymer-based core-shell nanogels via controlled radical polymerization. Nanogels with different sizes and functionalities in the core and shell were prepared. The nanogels exhibit remarkable efficiency in the protection of lysozyme against aggregation. Addition of nanogels suppresses the formation of toxic fibrils and also enables lysozyme to retain its enzymatic activity. Increasing the molecular weight and degree of hydrophobicity markedly increases its overall efficiency. Investigation of higher order structures revealed that lysozyme when heated without any additive loses its secondary structure and transforms into a random coil conformation. In contrast, presence of nanogels facilitates the retention of higher order structures by acting as molecular chaperones, thereby reducing molecular collisions. The present study is the first to show that it is possible to design zwitterionic nanogels using appropriate polymerization techniques that will protect proteins under conditions of extreme stress and inhibit aggregation.

Catechol Redox Induced Formation of Metal Core-Polymer Shell Nanoparticles.

Project description:A novel strategy was developed to synthesize polymer-coated metal nanoparticles (NPs) through reduction of metal cations with 3,4-dihydroxyphenylalanine (DOPA)-containing polyethylene glycol (PEG) polymers. Catechol redox chemistry was used to both synthesize metal NPs and simultaneously form a cross-linked shell of PEG polymers on their surfaces. DOPA reduced gold and silver cations into neutral metal atoms, producing reactive quinones that covalently cross-linked the PEG molecules around the surface of the NP. Importantly, these PEG-functionalized metal NPs were stable in physiological ionic strengths and under centrifugation, and hold broad appeal since they absorb and scatter light in aqueous solutions.

Flexible transparent displays based on core/shell upconversion nanophosphor-incorporated polymer waveguides.

Project description:Core/shell (C/S)-structured upconversion nanophosphor (UCNP)-incorporated polymer waveguide-based flexible transparent displays are demonstrated. Bright green- and blue-emitting Li(Gd,Y)F4:Yb,Er and Li(Gd,Y)F4:Yb,Tm UCNPs are synthesized via solution chemical route. Their upconversion luminescence (UCL) intensities are enhanced by the formation of C/S structure with LiYF4 shell. The Li(Gd,Y)F4:Yb,Er/LiYF4 and Li(Gd,Y)F4:Yb,Tm/LiYF4 C/S UCNPs exhibit 3.3 and 2.0 times higher UCL intensities than core counterparts, respectively. In addition, NaGdF4:Yb,Tm/NaGdF4:Eu C/S UCNPs are synthesized and they show red emission via energy transfer and migration of Yb3+ → Tm3+ → Gd3+ → Eu3+. The C/S UCNPs are incorporated into bisphenol A ethoxylate diacrylate which is used as a core material of polymer waveguides. The fabricated stripe-type polymer waveguides are highly flexible and transparent (transmittance > 90% in spectral range of 443-900 nm). The polymer waveguides exhibit bright blue, green, and red luminescence, depending on the incorporated UCNPs into the polymer core, under coupling with a near infrared (NIR) laser. Moreover, patterned polymer waveguide-based display devices are fabricated by reactive ion etching process and they realize bright blue-, green-, and red-colored characters under coupling with an NIR laser.

Strong Plasmon-Exciton Coupling in Ag Nanoparticle-Conjugated Polymer Core-Shell Hybrid Nanostructures.

Project description:Strong plasmon-exciton coupling between tightly-bound excitons in organic molecular semiconductors and surface plasmons in metal nanostructures has been studied extensively for a number of technical applications, including low-threshold lasing and room-temperature Bose-Einstein condensates. Typically, excitons with narrow resonances, such as J-aggregates, are employed to achieve strong plasmon-exciton coupling. However, J-aggregates have limited applications for optoelectronic devices compared with organic conjugated polymers. Here, using numerical and analytical calculations, we demonstrate that strong plasmon-exciton coupling can be achieved for Ag-conjugated polymer core-shell nanostructures, despite the broad spectral linewidth of conjugated polymers. We show that strong plasmon-exciton coupling can be achieved through the use of thick shells, large oscillator strengths, and multiple vibronic resonances characteristic of typical conjugated polymers, and that Rabi splitting energies of over 1000 meV can be obtained using realistic material dispersive relative permittivity parameters. The results presented herein give insight into the mechanisms of plasmon-exciton coupling when broadband excitonic materials featuring strong vibrational-electronic coupling are employed and are relevant to organic optoelectronic devices and hybrid metal-organic photonic nanostructures.

Core/Shell Conjugated Polymer/Quantum Dot Composite Nanofibers through Orthogonal Non-Covalent Interactions.

Project description:Nanostructuring organic polymers and organic/inorganic hybrid materials and controlling blend morphologies at the molecular level are the prerequisites for modern electronic devices including biological sensors, light emitting diodes, memory devices and solar cells. To achieve all-around high performance, multiple organic and inorganic entities, each designed for specific functions, are commonly incorporated into a single device. Accurate arrangement of these components is a crucial goal in order to achieve the overall synergistic effects. We describe here a facile methodology of nanostructuring conjugated polymers and inorganic quantum dots into well-ordered core/shell composite nanofibers through cooperation of several orthogonal non-covalent interactions including conjugated polymer crystallization, block copolymer self-assembly and coordination interactions. Our methods provide precise control on the spatial arrangements among the various building blocks that are otherwise incompatible with one another, and should find applications in modern organic electronic devices such as solar cells.

Polymer Topology Determines the Formation of Protein Corona on Core-Shell Nanoparticles.

Project description:Linear and cyclic poly(2-ethyl-2-oxazoline) (PEOXA) adsorbates provide excellent colloidal stability to superparamagnetic iron oxide nanoparticles (FexOy NPs) within protein-rich media. However, dense shells of linear PEOXA brushes cannot prevent weak but significant attractive interactions with human serum albumin. In contrast, their cyclic PEOXA counterparts quantitatively hinder protein adsorption, as demonstrated by a combination of dynamic light scattering and isothermal titration calorimetry. The cyclic PEOXA brushes generate NP shells that are denser and more compact than their linear counterparts, entirely preventing the formation of a protein corona as well as aggregation, even when the lower critical solution temperature of PEOXA in a physiological buffer is reached.

Core-Shell Polymer-Based Nanoparticles Deliver miR-155-5p to Endothelial Cells.

Project description:Heart failure occurs in over 30% of the worldwide population and most commonly originates from cardiovascular diseases such as myocardial infarction. microRNAs (miRNAs) target and silence specific mRNAs, thereby regulating gene expression. Because the endogenous miR-155-5p has been ascribed to vasculoprotection, loading it onto positively charged, core-shell poly(isobutylcyanoacrylate) (PIBCA)-polysaccharide nanoparticles (NPs) was attempted. NPs showed a decrease (p < 0.0001) in surface electrical charge (ζ potential), with negligible changes in size or shape when loaded with the anionic miR-155-5p. Presence of miR-155-5p in loaded NPs was further quantified. Cytocompatibility up to 100 μg/mL of NPs for 2 days with human coronary artery endothelial cells (hCAECs) was documented. NPs were able to enter hCAECs and were localized in the endoplasmic reticulum (ER). Expression of miR-155-5p was increased within the cells by 75-fold after 4 hours of incubation (p < 0.05) and was still noticeable at day 2. Differences between loaded NP-cultured cells and free miRNA, at days 1 (p < 0.05) and 2 (p < 0.001) suggest the ability of prolonged load release in physiological conditions. Expression of miR-155-5p downstream target BACH1 was decreased in the cells by 4-fold after 1 day of incubation (p < 0.05). This study is a first proof of concept that miR-155-5p can be loaded onto NPs and remain intact and biologically active in endothelial cells (ECs). These nanosystems could potentially increase an endogenous cytoprotective response and decrease damage within infarcted hearts.

Shell Architecture Strongly Influences the Glass Transition, Surface Mobility, and Elasticity of Polymer Core-Shell Nanoparticles.

Project description:Despite the growing application of nanostructured polymeric materials, there still remains a large gap in our understanding of polymer mechanics and thermal stability under confinement and near polymer-polymer interfaces. In particular, the knowledge of polymer nanoparticle thermal stability and mechanics is of great importance for their application in drug delivery, phononics, and photonics. Here, we quantified the effects of a polymer shell layer on the modulus and glass-transition temperature (T g) of polymer core-shell nanoparticles via Brillouin light spectroscopy and modulated differential scanning calorimetry, respectively. Nanoparticles consisting of a polystyrene (PS) core and shell layers of poly(n-butyl methacrylate) (PBMA) were characterized as model systems. We found that the high T g of the PS core was largely unaffected by the presence of an outer polymer shell, whereas the lower T g of the PBMA shell layer decreased with increasing PBMA thickness. The surface mobility was revealed at a temperature about 15 K lower than the T g of the PBMA shell layer. Overall, the modulus of the core-shell nanoparticles decreased with increasing PBMA shell layer thickness. These results suggest that the nanoparticle modulus and T g can be tuned independently through the control of nanoparticle composition and architecture.
