Generation of diffraction-free optical beams using wrinkled membranes.

ABSTRACT: Wrinkling has become a well developed bottom-up technique to make artificial surface textures in about the last decade. However, application of the optical properties of long range ordered wrinkles has been limited to one dimensional gratings to date. We report the demonstration of macroscopic optical focusing using wrinkled membranes, in which concentric wrinkle rings on a gold-PDMS bilayer membrane convert collimated illuminations to diffraction-free focused beams. Beam diameters of 300-400 μm have been observed in the visible range, which are dominantly limited by the eccentricity of the current devices. Based upon agreement between theoretical and experimental results on eccentricity effects, we predict a decrease of the beam diameter to no more than around 50 μm, if eccentricity is eliminated.

Compensation-free high-dimensional free-space optical communication using turbulence-resilient vector beams.

Project description:Free-space optical communication is a promising means to establish versatile, secure and high-bandwidth communication between mobile nodes for many critical applications. While the spatial modes of light offer a degree of freedom to increase the information capacity of an optical link, atmospheric turbulence can introduce severe distortion to the spatial modes and lead to data degradation. Here, we demonstrate experimentally a vector-beam-based, turbulence-resilient communication protocol, namely spatial polarization differential phase shift keying (SPDPSK), that can reliably transmit high-dimensional information through a turbulent channel without the need of any adaptive optics for beam compensation. In a proof-of-principle experiment with a controllable turbulence cell, we measure a channel capacity of 4.84 bits per pulse using 34 vector modes through a turbulent channel with a scintillation index of 1.09, and 4.02 bits per pulse using 18 vector modes through even stronger turbulence corresponding to a scintillation index of 1.54.

Diffraction-free beams in fractional Schrodinger equation.

Project description:We investigate the propagation of one-dimensional and two-dimensional (1D, 2D) Gaussian beams in the fractional Schrödinger equation (FSE) without a potential, analytically and numerically. Without chirp, a 1D Gaussian beam splits into two nondiffracting Gaussian beams during propagation, while a 2D Gaussian beam undergoes conical diffraction. When a Gaussian beam carries linear chirp, the 1D beam deflects along the trajectories z = ±2(x - x0), which are independent of the chirp. In the case of 2D Gaussian beam, the propagation is also deflected, but the trajectories align along the diffraction cone z = 2√(x(2) + y(2)) and the direction is determined by the chirp. Both 1D and 2D Gaussian beams are diffractionless and display uniform propagation. The nondiffracting property discovered in this model applies to other beams as well. Based on the nondiffracting and splitting properties, we introduce the Talbot effect of diffractionless beams in FSE.

Characterizing Organelles in Live Stem Cells Using Label-Free Optical Diffraction Tomography.

Project description:Label-free optical diffraction tomography (ODT), an imaging technology that does not require fluorescent labeling or other pre-processing, can overcome the limitations of conventional cell imaging technologies, such as fluorescence and electron microscopy. In this study, we used ODT to characterize the cellular organelles of three different stem cells-namely, human liver derived stem cell, human umbilical cord matrix derived mesenchymal stem cell, and human induced pluripotent stem cell-based on their refractive index and volume of organelles. The physical property of each stem cell was compared with that of fibroblast. Based on our findings, the characteristic physical properties of specific stem cells can be quantitatively distinguished based on their refractive index and volume of cellular organelles. Altogether, the method employed herein could aid in the distinction of living stem cells from normal cells without the use of fluorescence or specific biomarkers.

Mitotic Chromosomes in Live Cells Characterized Using High-Speed and Label-Free Optical Diffraction Tomography.

Project description:The cell nucleus is a three-dimensional, dynamic organelle organized into subnuclear compartments such as chromatin and nucleoli. The structure and function of these compartments are maintained by diffusion and interactions between related factors as well as by dynamic and structural changes. Recent studies using fluorescent microscopic techniques suggest that protein factors can access and are freely mobile in heterochromatin and in mitotic chromosomes, despite their densely packed structure. However, the physicochemical properties of the chromosome during cell division are not fully understood. In the present study, characteristic properties such as the refractive index (RI), volume of the mitotic chromosomes, and diffusion coefficient (D) of fluorescent probes inside the chromosome were quantified using an approach combining label-free optical diffraction tomography with complementary confocal laser-scanning microscopy and fluorescence correlation spectroscopy. Variations in these parameters correlated with osmotic conditions, suggesting that changes in RI are consistent with those of the diffusion coefficient for mitotic chromosomes and cytosol. Serial RI tomography images of chromosomes in live cells during mitosis were compared with three-dimensional confocal micrographs to demonstrate that compaction and decompaction of chromosomes induced by osmotic change were characterized by linked changes in chromosome RI, volume, and the mobilities of fluorescent proteins.

Three-dimensional label-free imaging and analysis of Pinus pollen grains using optical diffraction tomography.

Project description:The structure of pollen grains is related to the reproductive function of the plants. Here, three-dimensional (3D) refractive index maps were obtained for individual conifer pollen grains using optical diffraction tomography (ODT). The 3D morphological features of pollen grains from pine trees were investigated using measured refractive index maps, in which distinct substructures were clearly distinguished and analyzed. Morphological and physiochemical parameters of the pollen grains were quantified from the obtained refractive index (RI) maps and used to quantitatively study the interspecific differences of pollen grains from different strains. Our results demonstrate that ODT can assess the structure of pollen grains. This label-free and rapid 3D imaging approach may provide a new platform for understanding the physiology of pollen grains.

Particle manipulation beyond the diffraction limit using structured super-oscillating light beams.

Project description:The diffraction-limited resolution of light focused by a lens was derived in 1873 by Ernst Abbe. Later in 1952, a method to reach sub-diffraction light spots was proposed by modulating the wavefront of the focused beam. In a related development, super-oscillating functions, that is, band-limited functions that locally oscillate faster than their highest Fourier component, were introduced and experimentally applied for super-resolution microscopy. Up till now, only simple Gaussian-like sub-diffraction spots were used. Here we show that the amplitude and phase profile of these sub-diffraction spots can be arbitrarily controlled. In particular, we utilize Hermite-Gauss, Laguerre-Gauss and Airy functions to structure super-oscillating beams with sub-diffraction lobes. These structured beams are then used for high-resolution trapping and manipulation of nanometer-sized particles. The trapping potential provides unprecedented localization accuracy and stiffness, significantly exceeding those provided by standard diffraction-limited beams.

Generation of wavelength-independent subwavelength Bessel beams using metasurfaces.

Project description:Bessel beams are of great interest due to their unique non-diffractive properties. Using a conical prism or an objective paired with an annular aperture are two typical approaches for generating zeroth-order Bessel beams. However, the former approach has a limited numerical aperture (NA), and the latter suffers from low efficiency, as most of the incident light is blocked by the aperture. Furthermore, an additional phase-modulating element is needed to generate higher-order Bessel beams, which in turn adds complexity and bulkiness to the system. We overcome these problems using dielectric metasurfaces to realize meta-axicons with additional functionalities not achievable with conventional means. We demonstrate meta-axicons with high NA up to 0.9 capable of generating Bessel beams with full width at half maximum about as small as ~?/3 (?=405?nm). Importantly, these Bessel beams have transverse intensity profiles independent of wavelength across the visible spectrum. These meta-axicons can enable advanced research and applications related to Bessel beams, such as laser fabrication, imaging and optical manipulation.

Generation of Ince-Gaussian Beams Using Azocarbazole Polymer CGH.

Project description:Ince-Gaussian beams, defined as a solution to a wave equation in elliptical coordinates, have shown great advantages in applications such as optical communication, optical trapping and optical computation. However, to ingress these applications, a compact and scalable method for generating these beams is required. Here, we present a simple method that satisfies the above requirement, and is capable of generating arbitrary Ince-Gaussian beams and their superposed states through a computer-generated hologram of size 1 mm2, fabricated on an azocarbazole polymer film. Other structural beams that can be derived from the Ince-Gaussian beam were also successfully generated by changing the elliptical parameters of the Ince-Gaussian beam. The orthogonality relations between different Ince-Gaussian modes were investigated in order to verify applicability in an optical communication regime. The complete python source code for computing the Ince-Gaussian beams and their holograms are also provided.

Real-Time Monitoring of Multitarget Antimicrobial Mechanisms of Peptoids Using Label-Free Imaging with Optical Diffraction Tomography.

Project description:Antimicrobial peptides (AMPs) are promising therapeutics in the fight against multidrug-resistant bacteria. As a mimic of AMPs, peptoids with N-substituted glycine backbone have been utilized for antimicrobials with resistance against proteolytic degradation. Antimicrobial peptoids are known to kill bacteria by membrane disruption; however, the nonspecific aggregation of intracellular contents is also suggested as an important bactericidal mechanism. Here,structure-activity relationship (SAR) of a library of indole side chain-containing peptoids resulting in peptoid 29 as a hit compound is investigated. Then, quantitative morphological analyses of live bacteria treated with AMPs and peptoid 29 in a label-free manner using optical diffraction tomography (ODT) are performed. It is unambiguously demonstrated that both membrane disruption and intracellular biomass flocculation are primary mechanisms of bacterial killing by monitoring real-time morphological changes of bacteria. These multitarget mechanisms and rapid action can be a merit for the discovery of a resistance-breaking novel antibiotic drug.

Elasto-plasticity in wrinkled polymerized lipid membranes.

Project description:Biomembranes shown to behave like elastic sheets, can also suffer plastic deformations. Neutron scattering experiments on partially polymerised wrinkled membranes revealed that when a critical degree of polymerisation is crossed, the wrinkled membranes do not resume their spherical shapes. Instead they remain wrinkled and rigid while their non-polymerised counterparts resume their spherical floppy shapes. The yield stress of these membranes, measured for the first time via the fractal dimension, is intimately related to the degree of polymerisation probably through a 2D disorder that quenches the lateral diffusion of the lipid molecules. This work might shed light on the physical reason behind the irreversible deformation of echinocytes, acanthocytes and malaria infected red blood cells.
